Abstract

The present invention relates to N-heterocyclic sulphonamide compounds, in particular pyrazole sulphonamide compounds, and their use as N-myristoyl transferase inhibitors.

Claims (36)

1. A compound having the structure:

wherein

n is 0, 1, 2, 3, 4, 5 or 6;

E is independently selected from C and N;

W is selected from a hydrocarbyl optionally substituted with R 11 , an optionally substituted aryl or heteroaryl group and a carbocyclyl optionally substituted with one or more of halogen, cyano, amino, hydroxy, C 1-6 alkyl and C 1-6 alkoxy groups;

where M is selected from C and N;

R 3 , R 4 and R 5 are independently selected from hydrogen, R 12 , hydrocarbyl optionally substituted with R 12 , and —(CH 2 ) l -heterocyclyl optionally substituted with R 12 ; wherein R 1 and R 2 taken together with the atoms to which they are attached may form a heterocycle, optionally substituted with one or more R 12 ;

l is 0, 1, 2, 3, 4, 5 or 6;

wherein each R 11 and R 12 is independently selected from halogen, trifluoromethyl, cyano, thio, nitro, oxo, ═NR 13 , —OR 13 , —SR 13 , —C(O)R 13 , —C(O)OR 13 , —OC(O)R 13 , —NR 13 COR 14 , —NR 13 CONR 13 2 , —NR 13 COR 14 , —NR 13 CO 2 R 14 , —S(O)R 13 , —S(O) 2 R 13 , —SONR 13 2 , —NR 13 S(O) 2 R 14 ; —CSR 13 , —N(R 13 )R 14 , —C(O)N(R 13 )R 14 , —SO 2 N(R 13 )R 14 and R 15 ;

wherein R 13 and R 14 are each independently selected from hydrogen or R 15 ;

wherein R 15 is selected from hydrocarbyl, carbocyclyl and —(CH 2 ) m -heterocyclyl, and each R 15 is optionally and independently substituted with one or more of halogen, cyano, amino, hydroxy, C 1-6 alkyl and C 1-6 alkoxy;

ring D is an optionally substituted nitrogen containing 6 or 7 membered heterocycle, wherein each substitutable carbon or nitrogen in Ring D is optionally and independently substituted by one or more R 7 ;

R 7 is independently selected from hydrogen, R 20 , hydrocarbyl optionally substituted with R 20 , and —(CH 2 ) v -heterocyclyl optionally substituted with R 20 ;

v is 0, 1, 2, 3, 4, 5 or 6;

wherein each R 20 is independently selected from halogen, trifluoromethyl, cyano, thio, nitro, oxo, ═NR 21 , —OR 21 , —SR 21 , —C(O)R 21 , —C(O)OR 21 , —OC(O)R 21 , —NR 21 COR 22 , —NR 21 CONR 22 2 , —NR 21 COR 22 , —NR 21 CO 2 R 22 , —S(O)R 21 , —S(O) 2 R 21 , —SONR 21 2 , —NR 21 S(O) 2 R 22 ; —CSR 21 , N(R 21 )R 22 , —C(O)N(R 21 )R 22 , —SO 2 N(R 21 )R 22 and R 23 ;

wherein R 21 and R 22 are each independently selected from hydrogen or R 23 ; wherein R 23 is selected from hydrocarbyl, carbocyclyl and —(CH 2 ) w -heterocyclyl, and each R 23 is optionally and independently substituted with one or more of halogen, cyano, amino, hydroxy, C 1-6 alkyl and C 1-6 alkoxy;

w is 0, 1, 2, 3, 4, 5 or 6;

R 8 is selected from the list of optional substituents represented by the group R 4 ;

m is 0, 1, 2, 3, 4, 5 or 6;

p is 0, 1, 2, 3 or 4, wherein the values of R 4 may be the same or different;

q is 0, 1, 2, 3 or 4, wherein the values of R 5 may be the same or different; and

t is 0, 1, 2, 3, 4, 5 or 6, wherein the values of R 7 may be the same or different;

or a pharmaceutically acceptable salt thereof.

2. A compound as claimed in claim 1 wherein E is C.

3. A compound as claimed in claim 1 having the structure:

wherein R 5a , R 5b and R 5c are independently selected from hydrogen, R 12 , hydrocarbyl optionally substituted with R 12 , and —(CH 2 ) l -heterocyclyl optionally substituted with R 12 ;

wherein R 3a and R 3b are independently selected from hydrogen, R 12 , hydrocarbyl optionally substituted with R 12 , and —(CH 2 ) l -heterocyclyl optionally substituted with R 12 .

4. A compound as claimed in claim 3 wherein R 5a , R 5b and R 5c independently represent C 1-6 alkyl.

5. A compound as claimed in claim 1 having the structure:

6. A compound as claimed in claim 1 wherein W is C 1-6 alkyl or cycloalkyl.

7. A compound as claimed in claim 1 wherein R 4 is selected from hydrogen or C 1-6 alkyl optionally substituted with halogen.

8. A compound as claimed in claim 1 selected from the group consisting of:

9. A compound as claimed in claim 1 wherein one or more of R 3 and R 5 taken together with the atoms to which they are attached form a carbocycle, optionally substituted with R 12 .

10. A compound as claimed in claim 1 wherein R 4 is selected from hydrogen and C 1-6 alkyl optionally substituted with fluoro.

11. A compound as claimed in claim 3 wherein R 5a , R 5b and R 5c are independently selected from hydrogen, R 12 , C 1-6 alkyl, alkenyl, alkynyl and haloalkyl, optionally substituted with R 12 .

12. A compound as claimed in claim 3 wherein R 3a and R 3b are independently selected from hydrogen, R 12 , C 1-6 alkyl, alkenyl, alkynyl and haloalkyl, optionally substituted with R 12 .
