Method of selectively inhibiting Ras-mediated tumor growth in humans
Disclosed are compounds, for example, a compound of formula I, wherein R, R 0 , R 1 -R 8 , n, X, Y, Y′, and E are as described herein, pharmaceutical compositions containing such compounds, and methods of treating or preventing a disease or condition for example, cancer, mediated by the ras gene.
1. A method of inhibiting growth of a tumor in a patient, the method comprising assaying the tumor to determine if the tumor contains a mutant ras gene encoding for expression of a hyperactive Ras; and
if the tumor is found to contain a mutant ras gene encoding for expression of a hyperactive Ras, administering to said patient an effective amount of a compound selected from the group consisting of:
(Z)-2-(1-(4-hydroxy-3,5-dimethoxybenzylidene)-5-methoxy-2-methyl-1H-inden-3-yl)-N-(pyridin-3-yl)acetamide (004),
(Z)-2-(5-fluoro-1-(4-hydroxy-3,5-dimethoxybenzylidene)-2-methyl-1H-inden-3-yl)-N-(pyridin-2-ylmethyl)acetamide (010),
(Z)-2-(5-fluoro-1-(4-hydroxy-3,5-dimethoxybenzylidene)-2-methyl-1H-inden-3-yl)-N-(pyridin-3-ylmethyl)acetamide (011),
(Z)—N-benzyl-2-(5-fluoro-1-(4-hydroxy-3,5-dimethoxybenzylidene)-2-methyl-1H-inden-3-yl)acetamide (015), and
(Z)-2-(5-fluoro-1-(4-hydroxy-3,5-dimethoxybenzylidene)-2-methyl-1H-inden-3-yl)-N-phenylacetamide (035), wherein the tumor expresses a hyperactive Ras.
2. The method of claim 1 , wherein the tumor is selected from the group consisting of colorectal tumor, lung tumor, pancreatic tumor, melanoma, ovarian tumor, renal tumor, prostate tumor, head and neck tumor, endocrine tumor, uterine tumor, breast tumor, sarcoma, gastric tumor, hepatic tumor, esophageal tumor, central nervous system tumor, peripheral nervous system tumor, germline tumor, lymphoma, and leukemia.
3. The method of claim 2 , wherein the tumor is selected from colorectal tumor, lung tumor, and pancreatic tumor.
