Abstract

Methods of suppressing the activation of microglial cells in the Central Nervous System (CNS), methods of ameliorating or treating the neurological effects of cerebral ischemia or cerebral inflammation, and methods of combating specific diseases that affect the CNS by administering a compound that binds to microglial receptors and prevents or reduces microglial activation are described. Also described are methods of screening compounds for the ability to suppress or reduce microglial activation.

Claims (12)

1. A therapeutic composition comprising an ApoE peptide of about 18 to about 50 amino acids containing the sequence of SEQ ID No.: 10, wherein said ApoE peptide is conjugated to a carrier molecule capable of penetrating the blood brain barrier.

2. The therapeutic composition of claim 1 , wherein the presence of the carrier molecule increases transport of the ApoE peptide across the blood brain barrier compared to that which would occur in the absence of said carrier molecule.

The therapeutic composition of claim 1 , wherein said ApoE peptide contains SEQ ID No. 10 linked to one to five additional amino acids or amino acid analogues at the N-terminus or C-terminus or both the N- and C-terminus, wherein such additional amino acids do not adversely affect the therapeutic function of the ApoE peptide.

4. The therapeutic composition of claim 1 , wherein said ApoE peptide contains about 18 amino acids.

5. The therapeutic composition of claim 1 , wherein said ApoE peptide contains about 20 amino acids.

6. The therapeutic composition of claim 1 , wherein said ApoE peptide contains about 22 amino acids.

7. The therapeutic composition of claim 1 , wherein said ApoE peptide contains about 24 amino acids.

8. The therapeutic composition of claim 1 , wherein said ApoE peptide contains about 26 amino acids.

9. The therapeutic composition of claim 1 , wherein said ApoE peptide contains about 28 amino acids.

10. The therapeutic composition of claim 1 , wherein said ApoE peptide contains about 30 amino acids.

11. The therapeutic composition of claim 1 , wherein said ApoE peptide contains about 40 amino acids.

12. The therapeutic composition of claim 1 , wherein said carrier molecule is selected from the group consisting of pyridinium, fatty acids, inositol, cholesterol, glucose derivatives, hemoglobin, lysozyme, cytochrome c, ceruloplasmin, calmodulin, ubiquitin, substance P, histone, insulin and transferrin and peptides thereof, lipid vesicles and liposomes.
