Abstract

Compounds and compositions for eliciting or enhancing immune reactivity to HER-2/neu protein are disclosed. The compounds include polypeptides and nucleic acid molecules encoding such peptides. The compounds may be used for the prevention or treatment of malignancies in which the HER-2/neu oncogene is associated.

Claims (10)

1. An isolated nucleic acid molecule directing the expression of a polypeptide encoded by a DNA sequence selected from: (a) nucleotides 2026 through 3765 of SEQ ID NO: 1; and (b) DNA sequences that hybridize to a nucleotide sequence complementary to nucleotides 2026 through 3765 of SEQ ID NO: 1 under moderately stringent conditions, wherein the DNA sequence encodes a polypeptide that produces an immune response to HER-2/neu protein, and wherein the hybridization conditions comprise prewashing in a solution of 5×SSC, 0.5% SDS, 1.0 mM EDTA (pH8.0); hybridization at 50° to 65° C. in 5×SSC overnight; followed by washing twice at 65° for 20 minutes with each of 2×, 0.5× and 0.2×SSC containing 0.1% SDS; with the provisos that the polypeptide of (b) is not intact HER-2/neu protein and does not include the extracellular domain of HER-2/neu protein.

2. An isolated nucleic acid molecule according to claim 1 wherein the nucleic acid molecule directs the expression of a polypeptide encoded by nucleotides 2026 through 3765 of SEQ ID NO: 1.

3. An isolated nucleic acid molecule according to claim 1 wherein the nucleic acid molecule directs the expression of a polypeptide consisting of the amino acid sequence of SEQ ID NO:2 from amino acid 676 through amino acid 1255.

4. An isolated nucleic acid molecule according to claim 1 wherein the nucleic acid molecule directs the expression of a polypeptide consisting of a portion of the amino acid sequence of SEQ ID NO:2 from amino acid 676 through amino acid 1255.

5. An isolated nucleic acid molecule according to any one of claims 1 - 4 in combination with a pharmaceutically acceptable carrier or diluent.

An isolated viral vector directing the expression of a polypeptide encoded by a DNA sequence selected from: (a) nucleotides 2026 through 3765 of SEQ ID NO: 1; and (b) DNA sequences that hybridize to a nucleotide sequence complementary to nucleotides 2026 through 3765 of SEQ ID NO: 1 under moderately stringent conditions, wherein the DNA sequence encodes a polypeptide that produces an immune response to HER-2/neu protein, and wherein the hybridization conditions comprise prewashing in a solution of 5×SSC, 0.5% SDS, 1.0 mM EDTA (pH8.0); hybridization at 50° to 65° C. in 5×SSC overnight; followed by washing twice at 65° for 20 minutes with each of 2×, 0.5× and 0.2×SSC containing 0.1% SDS; with the provisos that the polypeptide of (b) is not intact HER-2/neu protein and does not include is not the extracellular domain of HER-2/neu protein.

7. An isolated viral vector according to claim 6 wherein the viral vector directs the expression of a polypeptide encoded by nucleotides 2026 through 3765 of SEQ ID NO: 1.

8. An isolated viral vector according to claim 6 wherein the viral vector directs the expression of a polypeptide consisting of the amino acid sequence of SEQ ID NO:2 from amino acid 676 through amino acid 1255.

9. An isolated viral vector according to claim 6 wherein the viral vector directs the expression of a polypeptide consisting of a portion of the amino acid sequence of SEQ ID NO:2 from amino acid 676 through amino acid 1255.

10. An isolated viral vector according to any one of claims 6 - 9 in combination with a pharmaceutically acceptable carrier or diluent.
