Abstract

The invention provides composition and methods for repairing a ruptured anterior cruciate ligament.

Claims (20)

1. A scaffold for repair of a tissue defect, comprising a three-dimensional matrix, adapted to facilitate cell in-growth, wherein said matrix is shaped for implantation into a patient at the site of the tissue defect, wherein said matrix comprises a natural matrix material wherein the natural matrix material is a collagen glycosaminoglycan copolymer comprised of collagen type I and wherein said scaffold further comprises plasma.

2. The scaffold of claim 1 , wherein the matrix is in the shape of a solid cylindrical member, a cylindrical member having hollow cavities, a tube, or a flat sheet rolled into a tube.

3. The scaffold of claim 1 , wherein the matrix further comprises one or more of growth factors, antibiotics, glycosaminoglycans, extracellular matrix protein, platelets, cytokines, neutralizing agents, crosslinking agents, stem cells, or cell media supplements.

4. The scaffold of claim 3 , wherein the one or more growth factors are selected from the group consisting of epidermal growth factor (EGF); fibroblast growth factor (FGF); transforming growth factor (TGF); and platelet derived growth factor (PDGF).

5. The scaffold of claim 3 , wherein the one or more extracellular matrix proteins are selected from the group consisting of elastin, laminin, fibronectin and entectin.

6. The scaffold of claim 1 wherein the scaffold is resistant to degradation by synovial fluid.

7. The scaffold of claim 1 , wherein the scaffold is freeze dried.

8. The scaffold of claim 1 , further comprising a mechanical device that is a suture or a clip for attachment of the scaffold to the site of the tissue defect to aid in the repair.

9. A method of treating a tissue defect in a subject, the method comprising placing the scaffold of claim 1 at the site of the tissue defect in the subject.

10. A scaffold for repair of a tissue defect, comprising a three-dimensional collagen matrix, wherein said collagen matrix comprises a collagen polymer comprised of type I collagen and wherein said scaffold further comprises plasma.

11. The scaffold of claim 10 , further comprising a mechanical device that is a suture or a clip for attachment of the scaffold to the site of the tissue defect to aid in the repair.

12. The scaffold of claim 10 , wherein the collagen matrix is in the form of or forming a tube or cylinder.

13. The scaffold of claim 10 , wherein the collagen matrix further comprises collagen or collagen glycosaminoglycan copolymer.

14. The scaffold of claim 10 , wherein the collagen matrix comprises one or more growth factors selected from the group consisting of epidermal growth factor (EGF); fibroblast growth factor (FGF); transforming growth factor (TGF); and platelet derived growth factor (PDGF).

15. The scaffold of claim 10 , wherein the collagen matrix comprises one or more extracellular matrix proteins selected from the group consisting of elastin, laminin, fibronectin and entectin.

16. The scaffold of claim 10 , wherein the tissue defect is an intra-articular tissue defect.

17. The scaffold of claim 16 , wherein the intra-articular tissue defect is a partial or complete tear of a ligament exposed to synovial fluid.

18. The scaffold of claim 17 , wherein the ligament is an anterior cruciate ligament (ACL).

19. The scaffold of claim 16 , wherein the intra-articular tissue defect is of meniscus, cartilage, labrum, tendon, muscle, or bone.

20. A method of treating a tissue defect in a subject, the method comprising placing the scaffold of claim 10 at the site of the tissue defect in the subject.
