Abstract

A microarray-based sample analysis (MBSA) system includes a cartridge holder adapted to receive a replaceable cartridge that is configured to receive a detachable, replaceable sample analysis unit containing one or more reaction chambers for sample analysis; a fluid control subsystem that controls fluid flow; and an optical subsystem configured to capture an image of the microarray.

Claims (22)

1. A microarray-based sample analysis (MBSA) system, comprising:

a cartridge holder having attached thereon an integrated cartridge, wherein the integrated cartridge comprises:

a cartridge body;

a fluidic interface that allows the integrated cartridge to be connected to the cartridge holder; and

a replaceable sample analysis unit comprising one or more reaction chambers for sample analysis,

wherein the one or more reaction chambers comprise one or more microarrays, wherein the replaceable sample analysis unit is detachable from the integrated cartridge and wherein the integrated cartridge is detachable from the cartridge holder;

a fluid control subsystem that controls fluid flow; and

an optical subsystem configured to capture an image of the one or more microarrays.

2. The MBSA system of claim 1 , further comprising:

a thermocycling system that is configured to control temperature in the reaction chamber during a reaction process.

3. The MBSA system of claim 2 , wherein the thermocycling system comprises a bladder thermocycler comprising two flexible bladders facing each other.

4. The MBSA system of claim 1 , wherein the optical subsystem comprises a laser light source.

5. The MBSA system of claim 1 , wherein the optical subsystem comprises a high-brightness LED light source.

6. The MBSA system of claim 1 , wherein the optical subsystem is capable of fluorescence imaging and fluorescence-independent imaging.

7. The MBSA system of claim 6 , wherein the optical subsystem comprises a first light source for fluorescence imaging and a second light source for fluorescence-independent imaging.

8. The MBSA system of claim 7 , wherein the second light source is a light-emitting diode (LED).

9. The MBSA system of claim 8 , wherein the LED is configured for darkfield illumination in reflected light.

10. The MBSA system of claim 8 , wherein the LED is configured for oblique illumination in transmitted light.

11. The MBSA system of claim 1 , wherein the one or more microarrays comprise a DNA microarray.

12. The MBSA system of claim 1 , wherein the one or more microarrays comprise a protein microarray.

13. The MBSA system of claim 1 , wherein the one or more reaction chambers comprise hydrophilic interior surfaces.

14. The MBSA system of claim 1 , wherein the detachable, replaceable sample analysis unit is attached to one side of the integrated cartridge so that the reaction chamber is extended from the side of the integrated cartridge.
