Abstract

This document relates to methods and materials for detecting mutations that can be linked to dementia. For example, methods and materials for detecting one or more mutations within PGRN nucleic acid are provided. This document also provides methods and materials for detecting the level of progranulin expression. In addition, this document relates to methods and materials for treating mammals having a neurodegenerative disorder (e.g., dementia). For example, methods and materials for increasing PGRN polypeptide levels in mammals are provided, as are methods and materials for identifying agents that can be used to increase PGRN polypeptide levels in mammals.

Claims (6)

1. A method for increasing the level of a PGRN polypeptide within a mammal having frontotemporal dementia, wherein the genome of said mammal comprises a null mutation in a PGRN nucleic acid, wherein said method comprises administering, to said mammal, a nucleic acid comprising a nucleic acid sequence that encodes said PGRN polypeptide.

2. The method of claim 1 , wherein said mammal is a human.

3. The method of claim 1 , wherein said nucleic acid comprises the nucleotide sequence set forth in SEQ ID NO:2.

4. The method of claim 1 , wherein said nucleic acid encodes a polypeptide comprising the amino acid sequence set forth in SEQ ID NO:1.

5. The method of claim 1 , wherein said nucleic acid is administered using a viral vector.

6. The method of claim 5 , wherein said viral vector is selected from the group consisting of an adeno-associated virus vector, a lentivirus vector, and an adenovirus vector.
