Abstract

The present invention concerns a monoclonal antibody and corresponding hybridoma cells and antigens suitable for isolating fetal cells from maternal blood. The inventive monoclonal antibody reacts with a surface antigen present on fetal red blood cells including their nucleated precursor cells, but not with surface antigens on adult erythroid cell.

Claims (10)

1. A method for detecting whether fetal red blood cells or their nucleated precursor cells are present in a sample comprising:

(a) contacting a sample with a monoclonal antibody which is (i) produced by the hybridoma 4B9 deposited with the Deutsche Sammlung von Mikroorganismen and Zellkulturen GmbH under accession number DSM ACC 2666 and (ii) fluorescently labeled, under conditions in which the antibody binds to fetal red blood cells and their nucleated precursor cells if present in the sample; and

(b) detecting cells bound by said fluorescently labeled antibody if present in the sample, wherein binding of the antibody indicates that the cell is a fetal red blood cell or a nucleated precursor cell thereof.

2. The method according to claim 1 , wherein fetal red blood cells or their nucleated precursor cells are present in the sample and wherein step (b) comprises detecting cells bound by said fluorescently labeled antibody.

3. The method according to claim 2 , further comprising analyzing cells bound by said fluorescently labeled antibody for a chromosomal or genetic defect.

4. The method according to claim 2 , wherein the sample is maternal blood.

5. The method according to claim 3 , wherein the sample is maternal blood.

6. The method according to claim 1 , which comprises separating cells bound by said fluorescently labeled antibody, wherein the separating is performed using flow cytometry, solid phase separation, immunomagnetic bead separation, or panning on plastic surfaces.

7. The method according to claim 6 , wherein the sample is maternal blood.

8. The method according to claim 1 , wherein the sample is maternal blood.
