Abstract

The present invention relates to isolated polypeptides having cellobiohydrolase activity and isolated polynucleotides encoding the polypeptides. The invention also relates to nucleic acid constructs, vectors, and host cells comprising the polynucleotides as well as methods of producing and using the polypeptides.

Claims (49)

1. A nucleic acid construct or expression vector comprising a polynucleotide encoding a polypeptide having cellobiohydrolase activity and one or more heterologous control sequences that direct the production of the polypeptide in an expression host, wherein the polynucleotide is operably linked to the one or more heterologous control sequences that direct the production of the polypeptide in a host cell, and wherein the polypeptide having cellobiohydrolase activity is selected from the group consisting of:

(a) a polypeptide comprising an amino acid sequence having at least 90% sequence identity to the amino acid sequence of amino acids 26 to 537 of SEQ ID NO: 4;

(b) a polypeptide encoded by a polynucleotide comprising a nucleotide sequence having at least 90% sequence identity to the nucleotide sequence of nucleotides 76 to 1614 of SEQ ID NO: 3;

(c) a fragment of the polypeptide of (a) or (b), wherein the fragment has cellobiohydrolase activity;

(d) a polypeptide comprising an amino acid sequence having at least 90% sequence identity to the amino acid sequence of amino acids 26 to 465 of SEQ ID NO: 4;

(e) a polypeptide encoded by a polynucleotide comprising a nucleotide sequence having at least 90% sequence identity to the nucleotide sequence of nucleotides 76 to 1395 of SEQ ID NO: 3; and

(f) a fragment of the polypeptide of (d) or (e), wherein the fragment has cellobiohydrolase activity.

2. An isolated recombinant host cell comprising the nucleic acid construct or expression vector of claim 1 .

3. A method of producing a polypeptide having cellobiohydrolase activity, comprising:

(a) cultivating the recombinant host cell of claim 2 under conditions conducive for production of the polypeptide; and

(b) recovering the polypeptide.

4. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide comprises an amino acid sequence having at least 95% sequence identity to the amino acid sequence of amino acids 26 to 537 of SEQ ID NO: 4.

5. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide comprises an amino acid sequence having at least 96% sequence identity to the amino acid sequence of amino acids 26 to 537 of SEQ ID NO: 4.

6. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide comprises an amino acid sequence having at least 97% sequence identity to the amino acid sequence of amino acids 26 to 537 of SEQ ID NO: 4.

7. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide comprises an amino acid sequence having at least 98% sequence identity to the amino acid sequence of amino acids 26 to 537 of SEQ ID NO: 4.

8. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide comprises an amino acid sequence having at least 99% sequence identity to the amino acid sequence of amino acids 26 to 537 of SEQ ID NO: 4.

9. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide comprises the amino acid sequence of SEQ ID NO: 4.

10. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide comprises the amino acid sequence of amino acids 26 to 537 of SEQ ID NO: 4.

11. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide comprises an amino acid sequence having at least 95% sequence identity to the amino acid sequence of amino acids 26 to 465 of SEQ ID NO: 4.

12. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide comprises an amino acid sequence having at least 96% sequence identity to the amino acid sequence of amino acids 26 to 465 of SEQ ID NO: 4.

13. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide comprises an amino acid sequence having at least 97% sequence identity to the amino acid sequence of amino acids 26 to 465 of SEQ ID NO: 4.

14. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide comprises an amino acid sequence having at least 98% sequence identity to the amino acid sequence of amino acids 26 to 465 of SEQ ID NO: 4.

15. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide comprises an amino acid sequence having at least 99% sequence identity to the amino acid sequence of amino acids 26 to 465 of SEQ ID NO: 4.

16. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide comprises the amino acid sequence of amino acids 26 to 465 of SEQ ID NO: 4.

17. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide is the polypeptide of (d), (e), or (f) and the polypeptide further comprises a cellulose binding domain.

18. An isolated recombinant host cell transformed with a polynucleotide encoding a polypeptide having cellobiohydrolase activity, wherein the polynucleotide is heterologous to the recombinant host cell, and wherein the polypeptide having cellobiohydrolase activity is selected from the group consisting of:

(a) a polypeptide comprising an amino acid sequence having at least 90% sequence identity to the amino acid sequence of amino acids 26 to 537 of SEQ ID NO: 4;

(b) a polypeptide encoded by a polynucleotide comprising a nucleotide sequence having at least 90% sequence identity to the nucleotide sequence of nucleotides 76 to 1614 of SEQ ID NO: 3;

(c) a fragment of the polypeptide of (a) or (b), wherein the fragment has cellobiohydrolase activity;

(d) a polypeptide comprising an amino acid sequence having at least 90% sequence identity to the amino acid sequence of amino acids 26 to 465 of SEQ ID NO: 4;

(e) a polypeptide encoded by a polynucleotide comprising a nucleotide sequence having at least 90% sequence identity to the nucleotide sequence of nucleotides 76 to 1395 of SEQ ID NO: 3; and

(f) a fragment of the polypeptide of (d) or (e), wherein the fragment has cellobiohydrolase activity.

19. The recombinant host cell of claim 18 , wherein the polypeptide comprises an amino acid sequence having at least 95% sequence identity to the amino acid sequence of amino acids 26 to 537 of SEQ ID NO: 4.

20. The recombinant host cell of claim 18 , wherein the polypeptide comprises an amino acid sequence having at least 96% sequence identity to the amino acid sequence of amino acids 26 to 537 of SEQ ID NO: 4.

21. The recombinant host cell of claim 18 , wherein the polypeptide comprises an amino acid sequence having at least 97% sequence identity to the amino acid sequence of amino acids 26 to 537 of SEQ ID NO: 4.

22. The recombinant host cell of claim 18 , wherein the polypeptide comprises an amino acid sequence having at least 98% sequence identity to the amino acid sequence of amino acids 26 to 537 of SEQ ID NO: 4.

23. The recombinant host cell of claim 18 , wherein the polypeptide comprises an amino acid sequence having at least 99% sequence identity to the amino acid sequence of amino acids 26 to 537 of SEQ ID NO: 4.

24. The recombinant host cell of claim 18 , wherein the polypeptide comprises the amino acid sequence of SEQ ID NO: 4.

25. The recombinant host cell of claim 18 , wherein the polypeptide comprises the amino acid sequence of amino acids 26 to 537 of SEQ ID NO: 4.

26. The recombinant host cell of claim 18 , wherein the polypeptide comprises an amino acid sequence having at least 95% sequence identity to the amino acid sequence of amino acids 26 to 465 of SEQ ID NO: 4.

27. The recombinant host cell of claim 18 , wherein the polypeptide comprises an amino acid sequence having at least 96% sequence identity to the amino acid sequence of amino acids 26 to 465 of SEQ ID NO: 4.

28. The recombinant host cell of claim 18 , wherein the polypeptide comprises an amino acid sequence having at least 97% sequence identity to the amino acid sequence of amino acids 26 to 465 of SEQ ID NO: 4.

29. The recombinant host cell of claim 18 , wherein the polypeptide comprises an amino acid sequence having at least 98% sequence identity to the amino acid sequence of amino acids 26 to 465 of SEQ ID NO: 4.

30. The recombinant host cell of claim 18 , wherein the polypeptide comprises an amino acid sequence having at least 99% sequence identity to the amino acid sequence of amino acids 26 to 465 of SEQ ID NO: 4.

31. The recombinant host cell of claim 18 , wherein the polypeptide comprises the amino acid sequence of amino acids 26 to 465 of SEQ ID NO: 4.

32. The recombinant host cell of claim 18 , wherein the polypeptide is the polypeptide of (d), (e), or (f) and the polypeptide further comprises a cellulose binding domain.

33. A method of producing a polypeptide having cellobiohydrolase activity, comprising:

(a) cultivating the recombinant host cell of claim 18 under conditions conducive for production of the polypeptide; and

(b) recovering the polypeptide.
