Abstract

The present invention relates to isolated polypeptides having xylanase activity and polynucleotides encoding the polypeptides. The invention also relates to nucleic acid constructs, vectors, and host cells comprising the polynucleotides as well as methods of producing and using the polypeptides.

Claims (45)

1. A nucleic acid construct comprising a polynucleotide encoding a polypeptide having xylanase activity operably linked to one or more heterologous control sequences that direct the production of the polypeptide in an expression host and wherein the polypeptide having xylanase activity is selected from the group consisting of:

(a) a polypeptide comprising an amino acid sequence having at least 85% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4; and

(b) a fragment of the sequence of amino acids 17 to 389 of SEQ ID NO: 4, wherein the fragment has xylanase activity.

2. The nucleic acid construct of claim 1 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 85% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

3. The nucleic acid construct of claim 1 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 90% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

4. The nucleic acid construct of claim 1 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 95% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

5. The nucleic acid construct of claim 1 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 97% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

6. The nucleic acid construct of claim 1 , wherein the polypeptide having xylanase activity comprises the sequence of SEQ ID NO: 4.

7. The nucleic acid construct of claim 1 , wherein the polypeptide having xylanase activity comprises the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

8. The nucleic acid construct of claim 1 , wherein the polypeptide having xylanase activity is a fragment of the sequence of SEQ ID NO: 4, wherein the fragment has xylanase activity.

The nucleic acid construct of claim 1 , wherein the polypeptide having xylanase activity is encoded by a polynucleotide that hybridizes under very high stringency conditions with the full-length complement of the sequence of nucleotides 49 to 1705 of SEQ ID NO: 3, wherein the very high stringency conditions are defined as prehybridization and hybridization at 42° C. in 5×SSPE, 0.3% SDS, 200 micrograms/ml sheared and denatured salmon sperm DNA, and 50% formamide, and washing three times each for 15 minutes using 2×SSC, 0.2% SDS at 70° C.

The nucleic acid construct of claim 1 , wherein the polypeptide having xylanase activity is a variant of the sequence of amino acids 17 to 389 of SEQ ID NO: 4 comprising a substitution, deletion and/or insertion at one or more positions and wherein the variant comprises an amino acid sequence that has at least 95% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

11. An isolated recombinant host cell transformed with the nucleic acid construct of claim 1 .

12. A method of producing a polypeptide having xylanase activity, comprising:

(a) cultivating the recombinant host cell of claim 11 under conditions conducive for production of the polypeptide; and

(b) recovering the polypeptide.

An isolated recombinant host cell transformed with a heterologous nucleic acid construct comprising a polynucleotide operably linked to one or more control sequences that direct the production of a polypeptide in the recombinant host cell, wherein the polypeptide has xylanase activity and is selected from the group consisting of:

(a) a polypeptide comprising an amino acid sequence having at least 85% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4; and

(b) a fragment of the sequence of amino acids 17 to 389 of SEQ ID NO: 4, wherein the fragment has xylanase activity.

14. The recombinant host cell of claim 13 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 85% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

15. The recombinant host cell of claim 13 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 90% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

16. The recombinant host cell of claim 13 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 95% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

17. The recombinant host cell of claim 13 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 97% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

18. The recombinant host cell of claim 13 , wherein the polypeptide having xylanase activity comprises the sequence of SEQ ID NO: 4.

19. The recombinant host cell of claim 13 , wherein the polypeptide having xylanase activity comprises the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

20. The recombinant host cell of claim 13 , wherein the polypeptide having xylanase activity is a fragment of the sequence of SEQ ID NO: 4, wherein the fragment has xylanase activity.

The recombinant host cell of claim 13 , wherein the polypeptide having xylanase activity is encoded by a polynucleotide that hybridizes under very high stringency conditions with the full-length complement of the sequence of nucleotides 49 to 1705 of SEQ ID NO: 3, wherein the very high stringency conditions are defined as prehybridization and hybridization at 42° C. in 5×SSPE, 0.3% SDS, 200 micrograms/ml sheared and denatured salmon sperm DNA, and 50% formamide, and washing three times each for 15 minutes using 2×SSC, 0.2% SDS at 70° C.

22. The recombinant host cell of claim 13 , wherein the polypeptide having xylanase activity is a variant of the sequence of amino acids 17 to 389 of SEQ ID NO: 4 comprising a substitution, deletion and/or insertion at one or more positions and wherein the variant comprises an amino acid sequence that has at least 95% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

23. A method of producing a polypeptide having xylanase activity, comprising:

(a) cultivating the recombinant host cell of claim 13 under conditions conducive for production of the polypeptide; and

(b) recovering the polypeptide.

24. The nucleic acid construct of claim 1 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 91% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

25. The nucleic acid construct of claim 1 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 92% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

26. The nucleic acid construct of claim 1 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 93% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

27. The nucleic acid construct of claim 1 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 94% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

28. The nucleic acid construct of claim 1 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 96% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

29. The nucleic acid construct of claim 1 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 98% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

30. The nucleic acid construct of claim 1 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 99% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

31. The recombinant host cell of claim 13 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 91% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

32. The recombinant host cell of claim 13 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 92% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

33. The recombinant host cell of claim 13 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 93% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

34. The recombinant host cell of claim 13 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 94% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

35. The recombinant host cell of claim 3 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 96% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

36. The recombinant host cell of claim 13 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 98% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.

37. The recombinant host cell of claim 13 , wherein the polypeptide having xylanase activity comprises an amino acid sequence that has at least 99% sequence identity to the sequence of amino acids 17 to 389 of SEQ ID NO: 4.
