Abstract

A method and apparatus for separating cell suspension into centrate and concentrate includes a solid wall rotatable centrifuge bowl ( 82 ). A single use rotatable core ( 88 ) is positioned in the bowl and bounds a cavity ( 90 ) which serves as a separation chamber. The bowl rotates about an axis ( 84 ). An inlet tube ( 92 ) and an outlet tube ( 102 ) extend along the axis in the bowl cavity. The centripetal pump ( 98 ) in the cavity is in connection with the outlet tube. The centrate discharge pump ( 120 ) is in connection with the outlet tube. A pressure damping reservoir ( 122 ) is fluidly connected between the outlet tube and the discharge pump. At least one control circuit ( 142 ) is operative to control the apparatus to maintain a positive pressure above atmospheric pressure within the cavity, so as to maintain the cell suspension away from at least one seal ( 106 ) that bounds the interior of the cavity.

Claims (260)

1. Apparatus comprising:

a single use structure configured for use in a centrifuge system including a multi-use rotatable centrifuge bowl, wherein the structure is configured to be positioned in the rotatable centrifuge bowl and to separate within an interior area of the single use structure, cells in a cell culture batch material into cell concentrate and cell centrate,

the single use structure in an operative position, including

an upper portion,

a lower portion,

a hollow cylindrical core intermediate of the upper portion and the lower portion,

a separation chamber in radially surrounding relation of the hollow cylindrical core,

an outer wall, wherein the outer wall

extends in fluid tight relation with the upper and lower portions and bounds the separation chamber,

extends in surrounding relation of the hollow cylindrical core, and

has an internal truncated cone shape with a smaller inside radius adjacent the lower portion,

a vertically extending feed tube,

a vertically extending centrate discharge tube,

a vertically extending concentrate discharge tube,

wherein the upper and lower portions and the outer wall are rotatable within the rotatable centrifuge bowl about a vertical axis,

wherein during rotation of the rotatable centrifuge bowl, the upper and lower portions and the outer wall rotate relative to each of the feed tube, the centrate discharge tube, and the concentrate discharge tube,

a centrate centripetal pump, wherein the centrate centripetal pump is axially aligned with the hollow cylindrical core, disposed coaxially about the feed tube, and in fluid communication with the centrate discharge tube,

wherein the centrate centripetal pump is positioned in the interior area in a centrate centripetal pump chamber, wherein the centrate centripetal pump chamber is in fluid communication with the separation chamber,

wherein during rotation of the rotatable centrifuge bowl, the upper and lower portions and the outer wall rotate relative to the centrate centripetal pump,

a concentrate centripetal pump, wherein the concentrate centripetal pump is axially aligned with the hollow cylindrical core, disposed coaxially about the feed tube, positioned vertically above the centrate centripetal pump, and in fluid communication with the concentrate discharge tube,

wherein the concentrate centripetal pump is positioned in the interior area in a concentrate centripetal pump chamber,

wherein the concentrate centripetal pump chamber is in fluid communication with the separation chamber, and wherein during rotation of the rotatable centrifuge bowl the upper and lower portions and the outer wall rotate relative to the concentrate centripetal pump.

2. Apparatus comprising:

a single use structure configured for use in a centrifuge system including a multi-use rotatable centrifuge bowl, wherein the single use structure is configured to be releasably positioned in the rotatable centrifuge bowl and to separate within an interior area of the single use structure, cells in a cell culture batch material into cell concentrate and cell centrate,

the single use structure including:

a first disc shape portion at a first axial end,

a second disc shape portion at a second axial end axially disposed below the first axial end,

a hollow cylindrical core coaxially arranged with the first and second disc shape portions and axially intermediate of the first disc shape portion and the second disc shape portion,

an outer wall, wherein the outer wall

is in fluid tight relation with the first and second disc shape portions,

extends in surrounding relation of at least a portion of the hollow cylindrical core,

bounds a separation chamber that extends radially between the hollow cylindrical core and the outer wall,

is tapered such that the outer wall has a radius that is larger axially adjacent to the first disc shape portion than axially adjacent to the second disc shape portion,

an axially extending feed tube, wherein the feed tube includes an inlet opening within the interior area,

an axially extending centrate discharge tube coaxial with and radially outward of the feed tube,

an axially extending concentrate discharge tube coaxial with and radially outward of the centrate discharge tube,

a centrate centripetal pump, wherein the centrate centripetal pump is disposed coaxially about the feed tube and in fluid communication with the centrate discharge tube,

wherein the centrate centripetal pump is positioned in the interior area in a centrate centripetal pump chamber, wherein the centrate centripetal pump chamber is in fluid communication with the separation chamber,

wherein during rotation of the single use structure with the rotatable centrifuge bowl, the first and second disc shape portions and the outer wall rotate relative to the centrate centripetal pump,

a concentrate centripetal pump, wherein the concentrate centripetal pump is disposed coaxially about the feed tube, is in fluid communication with the concentrate discharge tube, and is positioned in axially intermediate relation of the centrate centripetal pump and the first disc shape portion,

wherein the concentrate centripetal pump is positioned in the interior area in a concentrate centripetal pump chamber,

wherein the concentrate centripetal pump chamber is in fluid communication with the separation chamber, and wherein during rotation of the single use structure with the rotatable centrifuge bowl, the first and second disc shape portions and the outer wall rotate relative to the concentrate centripetal pump.

3. Apparatus comprising:

a single use structure configured for use in a centrifuge system including a multi-use rotatable centrifuge bowl, wherein the single use structure is configured to be releasably positioned in the rotatable centrifuge bowl and to separate within an interior area of the single use structure, cells in a cell culture batch material into cell concentrate and cell centrate,

the single use structure including:

a first disc shape portion adjacent a first axial end,

a second axial end axially disposed below the first axial end in an operative position of the single use structure,

a hollow cylindrical core coaxially arranged and in operative connection with the first disc shape portion and axially intermediate of the first disc shape portion and the second axial end,

a flexible outer wall, wherein the flexible outer wall

is more flexible than the first disc shape portion and in the operative position conforms to an internal structure of the rotatable centrifuge bowl in which the single use structure is positioned,

is in fluid tight relation with the first disc shape portion,

extends in surrounding relation of at least a portion of the hollow cylindrical core,

bounds a separation chamber that extends radially between the hollow cylindrical core and the flexible outer wall,

is tapered such that the flexible outer wall has a radius that is larger axially adjacent to the first disc shape portion than axially adjacent to the second axial end,

an axially extending feed tube, wherein the feed tube includes an inlet opening within the interior area,

an axially extending centrate discharge tube coaxial with and radially outward of the feed tube,

an axially extending concentrate discharge tube coaxial with and radially outward of the centrate discharge tube,

a centrate centripetal pump, wherein the centrate centripetal pump is disposed coaxially about the feed tube and in fluid communication with the centrate discharge tube,

wherein the centrate centripetal pump is positioned in the interior area in a centrate centripetal pump chamber, wherein the centrate centripetal pump chamber is in fluid communication with the separation chamber,

wherein during rotation of the single use structure with the rotatable centrifuge bowl, the first disc shape portion and the outer wall rotate relative to the centrate centripetal pump,

a concentrate centripetal pump, wherein the concentrate centripetal pump is disposed coaxially about the feed tube, is in fluid communication with the concentrate discharge tube, and is positioned in axially intermediate relation of the centrate centripetal pump and the first axial end,

wherein the concentrate centripetal pump is positioned in the interior area in a concentrate centripetal pump chamber,

wherein the concentrate centripetal pump chamber is in fluid communication with the separation chamber, and wherein during rotation of the single use structure with the multi-use rotatable centrifuge bowl, the first disc shape portion and the outer wall rotate relative to the concentrate centripetal pump.

4. The apparatus according to claim 3

a plurality of radially extending accelerator vanes, wherein the accelerator vanes are disposed axially intermediate of the hollow cylindrical core and the second axial end.

5. The apparatus according to claim 3

wherein the single use structure includes a second disc shape portion adjacent the second axial end, wherein the hollow cylindrical core is in operative connection with the second disc shape portion,

wherein the flexible outer wall is in engagement with the second disc shape portion.

6. Apparatus comprising:

a single use structure configured for use in a centrifuge system including a multi-use rotatable centrifuge bowl, wherein the single use structure is configured to be positioned in the rotatable centrifuge bowl and to separate within an interior area of the single use structure, cells in a cell culture batch material into cell concentrate and cell centrate,

the single use structure in an operative position including

an upper portion,

a lower portion,

a hollow cylindrical core intermediate of the upper portion and the lower portion,

a separation chamber in radially surrounding relation of the core,

an outer wall, wherein the outer wall

extends in fluid tight relation with the upper and lower portions and bounds the separation chamber,

extends in surrounding relation of the hollow cylindrical core, and

has an internal truncated cone shape with a smaller inside radius adjacent to the lower portion,

a vertically extending feed tube,

a vertically extending centrate discharge tube,

a vertically extending concentrate discharge tube,

wherein the upper and lower portions and the outer wall are rotatable within the rotatable centrifuge bowl about a vertical axis,

wherein during rotation of the rotatable centrifuge bowl, the upper and lower portions and the outer wall rotate relative to each of the feed tube, the centrate discharge tube, and the concentrate discharge tube,

a centrate centripetal pump, wherein the centrate centripetal pump is

axially aligned with the hollow cylindrical core,

disposed coaxially about the feed tube,

in fluid communication with the centrate discharge tube, and

positioned in the interior area in a centrate centripetal pump chamber,

wherein the centrate centripetal pump chamber is in fluid communication with the separation chamber,

wherein during rotation of the rotatable centrifuge bowl, the upper and lower portions and the outer wall rotate relative to the centrate centripetal pump,

a concentrate centripetal pump, wherein the concentrate centripetal pump is

axially aligned with the hollow cylindrical core,

disposed coaxially about the feed tube,

positioned vertically above the centrate centripetal pump,

in fluid communication with the concentrate discharge tube, and

positioned in the interior area in a concentrate centripetal pump chamber,

wherein the concentrate centripetal pump chamber is in fluid communication with the separation chamber,

wherein during rotation of the rotatable centrifuge bowl, the upper and lower portions and the outer wall rotate relative to the concentrate centripetal pump,

an external concentrate pump,

wherein the external concentrate pump is outside of the single use structure and is in fluid connection with the concentrate discharge tube,

an optical sensor, wherein the optical sensor is in operative connection with an interior of a concentrate discharge line,

wherein the concentrate discharge line is in operative connection with the concentrate discharge tube and the external concentrate pump,

a controller, wherein the controller is in operative connection with the external concentrate pump and the optical sensor,

wherein the controller is operative to control the external concentrate pump responsive at least in part to the optical sensor.

7. The apparatus according to claim 6

wherein the concentrate centripetal pump has a larger peripheral diameter than the centrate centripetal pump.

8. The apparatus according to claim 6

wherein the upper portion includes a rotatable upper flange,

wherein the lower portion includes a rotatable lower flange,

and wherein the outer wall includes a flexible liner that extends between the upper flange and the lower flange, wherein the flexible liner is more flexible than the upper and lower flanges and in the operative position conforms to an internal structure of the rotatable centrifuge bowl in which the single use structure is positioned.

9. The apparatus according to claim 6

wherein the upper portion includes an upper flange, wherein the upper flange

is configured to releasably operatively engage the rotatable centrifuge bowl.

10. The apparatus according to claim 6

wherein the lower portion includes a plurality of radially extending accelerator vanes.

11. The apparatus according to claim 6 and further including

a vertically extending diluent feed tube, wherein the diluent feed tube extends coaxially with the feed tube, centrate discharge tube and concentrate discharge tube,

wherein the diluent feed tube is configured to enable diluent to be added to the interior area.

12. The apparatus according to claim 6

wherein the single use structure enables the centrifuge system to operate with at least one of continuous or semi continuous discharge of cell concentrate.

13. The apparatus according to claim 6

wherein the centrate centripetal pump comprises a pair of discs that are disposed from each other an axial distance, wherein the axial distance is selectively variable.

14. The apparatus according to claim 6

wherein the concentrate centripetal pump comprises a pair of discs that are disposed from each other an axial distance, wherein the axial distance is selectively variable.

15. The apparatus according to claim 6

wherein the centrate centripetal pump and the concentrate centripetal pump are coaxially aligned.

16. The apparatus according to claim 6

wherein the outer wall has a greater inside radius at a vertical level corresponding to a top portion of the hollow cylindrical core than at a vertical level corresponding to a bottom portion of the hollow cylindrical core.

17. The apparatus according to claim 6 and further comprising

a plurality of radially extending accelerator vanes, wherein the plurality of accelerator vanes extend in the centrate centripetal pump chamber vertically above the centripetal centrate pump.

18. The apparatus according to claim 6

wherein the upper portion includes an upper flange, and the bottom portion includes a lower flange,

and wherein the hollow cylindrical core extends between and in operative engagement with each of the upper flange and the lower flange.

19. The apparatus according to claim 6 and further comprising

at least one annular air tight seal,

wherein the at least one annular air tight seal extends in sealing relation between the upper portion and an outer wall of one of the feed tube, the centrate discharge tube and the concentrate discharge tube,

wherein the at least one annular air tight seal is operative to maintain air within the interior area, wherein the air isolates the at least one annular air tight seal from the cell culture batch material being processed, while maintaining the cell culture batch material in fluid connection with each of the centrate centripetal pump and the concentrate centripetal pump.

20. Apparatus comprising:

a single use structure configured for use in a centrifuge system including a multi-use rotatable centrifuge bowl, wherein the single use structure is configured to be releasably positioned in the rotatable centrifuge bowl and to separate within an interior area of the single use structure, cells in a cell culture batch material into cell concentrate and cell centrate,

the single use structure in an operative position including:

a first disc shape portion at a first axial end,

a second disc shape portion at a second axial end axially disposed below the first axial end,

a hollow cylindrical core coaxially arranged with the first and second disc shape portions, wherein the hollow cylindrical core extends axially intermediate of the first disc shape portion and the second disc shape portion,

an outer wall, wherein the outer wall

is in fluid tight relation with the first and second disc shape portions,

extends in surrounding relation of at least a portion of the hollow cylindrical core,

bounds a separation chamber that extends radially between the hollow cylindrical core and the outer wall,

is tapered such that the outer wall has a radius that is larger axially adjacent to the first disc shape portion than axially adjacent to the second disc shape portion,

an axially extending feed tube,

wherein the feed tube includes an inlet opening within the interior area,

an axially extending centrate discharge tube coaxial with and radially outward of the feed tube,

an axially extending concentrate discharge tube coaxial with and radially outward of the centrate discharge tube,

a centrate centripetal pump, wherein the centrate centripetal pump is

disposed coaxially about the feed tube,

in fluid communication with the centrate discharge tube, and

positioned in the interior area in a centrate centripetal pump chamber,

wherein the centrate centripetal pump chamber is in fluid communication with the separation chamber,

wherein during the rotation of the single use structure with the rotatable centrifuge bowl, the first and second disc shape portions and the outer wall rotate relative to the centrate centripetal pump,

a concentrate centripetal pump, wherein the concentrate centripetal pump is

disposed coaxially about the feed tube,

in fluid communication with the concentrate discharge tube,

positioned in axially intermediate relation of the centrate centripetal pump and the first disc shape portion,

positioned in the interior area in a concentrate centripetal pump chamber,

wherein the concentrate centripetal pump chamber is in fluid communication with the separation chamber,

wherein during rotation of the single use structure with the rotatable centrifuge bowl, the first and second disc shape portions and the outer wall rotate relative to the concentrate centripetal pump,

an external concentrate pump, wherein the external concentrate pump is

outside of the single use structure, and

in fluid communication with the concentrate discharge tube,

an optical sensor, wherein the optical sensor is in operative connection with an interior of a concentrate discharge line,

wherein the concentrate discharge line is in operative connection with the concentrate discharge tube and the external concentrate pump,

a controller, wherein the controller is

in operative connection with the external concentrate pump and the optical sensor, and

is operative to control the external concentrate pump responsive at least in part to the optical sensor.

21. The apparatus according to claim 20

wherein the concentrate centripetal pump has a larger peripheral diameter than the centrate centripetal pump.

22. The apparatus according to claim 20

wherein the outer wall comprises a flexible liner,

wherein the flexible liner is

in fluid tight connection with each of the first and second disc shape portions, and

more flexible than the first and second disc shape portions and in an operative position conforms to an internal structure of the rotatable centrifuge bowl in which the single use structure is positioned.

23. The apparatus according to claim 20 and further comprising:

at least one annular air tight seal,

wherein the at least one annular air tight seal extends in operative sealing relation between the first disc shape portion and an outer wall of the concentrate discharge tube,

wherein the at least one annular air tight seal is operative to maintain air within the interior area during cell culture batch processing, wherein the air isolates the at least one annular air tight seal from the cell culture batch, while maintaining the cell culture batch in fluid connection with each of the centrate centripetal pump and the concentrate centripetal pump.

24. The apparatus according to claim 20

wherein the second disc shape portion includes a plurality of radially extending accelerator vanes.

25. Apparatus comprising:

a structure configured for use in a centrifuge system including a rotatable centrifuge bowl, wherein the structure is configured to be releasably positioned in the rotatable centrifuge bowl and to separate within an interior area of the structure, cells in a cell culture batch material into cell concentrate and cell centrate,

the structure in an operative position including

an upper portion,

a lower portion,

a cylindrical core, wherein the cylindrical core extends axially intermediate of the upper portion and the lower portion,

a separation chamber in radially surrounding relation of the core,

an outer wall, wherein the outer wall

extends in fluid tight relation with the upper and lower portions and bounds the separation chamber,

extends in surrounding relation of the core, and

has an internal truncated cone shape with a smaller inside radius adjacent to the lower portion,

a vertically extending feed tube,

a vertically extending centrate discharge tube,

a vertically extending concentrate discharge tube,

wherein the upper and lower portions and the outer wall are rotatable in the rotatable centrifuge bowl about a vertical axis,

wherein during rotation of the rotatable centrifuge bowl, the upper and lower portions and the outer wall rotate relative to each of the feed tube, the centrate discharge tube, and the concentrate discharge tube,

a centrate centripetal pump, wherein the centrate centripetal pump is

axially aligned with the core,

disposed coaxially about the feed tube,

in fluid communication with the centrate discharge tube, and

positioned in the interior area in a centrate centripetal pump chamber,

wherein the centrate centripetal pump chamber is in fluid communication with the separation chamber,

wherein during rotation of the rotatable centrifuge bowl, the upper and lower portions and the outer wall rotate relative to the centrate centripetal pump,

a concentrate centripetal pump, wherein the concentrate centripetal pump is

axially aligned with the core,

disposed coaxially about the feed tube,

positioned vertically above the centrate centripetal pump,

in fluid communication with the concentrate discharge tube, and

positioned in the interior area in a concentrate centripetal pump chamber,

wherein the concentrate centripetal pump chamber is in fluid communication with the separation chamber,

wherein during rotation of the rotatable centrifuge bowl, the upper and lower portions and the outer wall rotate relative to the concentrate centripetal pump,

an external concentrate pump,

wherein the external concentrate pump is outside of the single use structure and is in fluid connection with the concentrate discharge tube,

an optical sensor, wherein the optical sensor is in operative connection with an interior of a concentrate discharge line,

wherein the concentrate discharge line is in fluid connection with the concentrate discharge tube and the external concentrate pump,

a controller, wherein the controller is in operative connection with the external concentrate pump and the optical sensor, and

wherein the controller is operative to control the external concentrate pump responsive at least in part to the optical sensor.

26. Apparatus comprising:

a structure configured for use in a centrifuge system including a rotatable centrifuge bowl, wherein the structure is configured to be releasably positioned in the rotatable centrifuge bowl and to separate within an interior area of the structure, cells in a cell culture batch material into cell concentrate and cell centrate,

the structure in an operative position including:

a first disc shape portion at a first axial end,

a second disc shape portion at a second axial end axially disposed below the first axial end,

a cylindrical core coaxially arranged with the first and second disc shape portions, wherein the core extends axially intermediate of the first disc shape portion and the second disc shape portion,

an outer wall, wherein the outer wall

is in fluid tight relation with the first and second disc shape portions,

extends in surrounding relation of at least a portion of the core,

bounds a separation chamber that extends radially between the core and the outer wall,

is tapered such that the outer wall has a radius that is larger axially adjacent to the first disc shape portion than axially adjacent to the second disc shape portion,

an axially extending feed tube,

wherein the feed tube includes an inlet opening within the interior area,

an axially extending centrate discharge tube coaxial with and radially outward of the feed tube,

an axially extending concentrate discharge tube coaxial with and radially outward of the centrate discharge tube,

a centrate centripetal pump, wherein the centrate centripetal pump is

disposed coaxially about the feed tube,

in fluid communication with the centrate discharge tube, and

positioned in the interior area in a centrate centripetal pump chamber,

wherein the centrate centripetal pump chamber is in fluid communication with the separation chamber,

wherein during the rotation of the structure with the rotatable centrifuge bowl, the first and second disc shape portions and the outer wall rotate relative to the centrate centripetal pump,

a concentrate centripetal pump, wherein the concentrate centripetal pump is

disposed coaxially about the feed tube,

in fluid communication with the concentrate discharge tube,

positioned in axially intermediate relation of the centrate centripetal pump and the first disc shape portion,

positioned in the interior area in a concentrate centripetal pump chamber,

wherein the concentrate centripetal pump chamber is in fluid communication with the separation chamber,

wherein during rotation of the structure with the rotatable centrifuge bowl, the first and second disc shape portions and the outer wall rotate relative to the concentrate centripetal pump,

an external concentrate pump, wherein the external concentrate pump is

outside of the structure, and

in fluid communication with the concentrate discharge tube,

an optical sensor, wherein the optical sensor is in operative connection with an interior of a concentrate discharge line,

wherein the concentrate discharge line is in operative connection with the concentrate discharge tube and the external concentrate pump,

a controller, wherein the controller is

in operative connection with the external concentrate pump and the optical sensor, and

operative to control the external concentrate pump responsive at least in part to the optical sensor.
