Abstract

The present invention is directed to a method of producing compositions including embryonic proteins. The method includes culturing cells under hypoxic conditions on a biocompatible surface in vitro. The culturing method produces both soluble and non-soluble fractions, which may be used separately or in combination to obtain physiologically acceptable compositions useful in a variety of medical and therapeutic applications.

Claims (10)

1. A surface coating for a medical device comprising a composition made by the method comprising:

culturing fibroblast cells under hypoxic conditions of 1-5% oxygen on microcarrier beads or a three dimensional surface in a suitable growth medium, for at least 2 weeks, thereby producing multipotent stem cells, wherein the multipotent stem cells produce and secrete into the growth medium the composition; and

collecting the composition after said 2 weeks.

2. The surface coating of claim 1 , wherein the medical device is selected from the group consisting of a pacemaker, a stent, a stent graft, a vascular prosthesis, a heart valve, a shunt, a drug delivery port and a catheter.

3. The surface coating of claim 1 , wherein the composition further comprises a growth factor or a biological agent.

The surface coating of claim 1 , wherein the composition is applied to the medical device, and wherein applying the composition to the medical device comprises using a photoactive crosslinker to generate permanent covalent bonding of the composition to the device surface upon activation of the crosslinker.

5. The surface coating of claim 4 , wherein activation of the crosslinker comprises applying ultraviolet radiation.

6. The surface coating of claim 1 , wherein the composition promotes wound healing, modifies inflammation, modifies a fibrous capsule formation, modifies tissue ingrowth, or modifies cell ingrowth.

7. The surface coating of claim 1 , wherein the composition treats damaged tissue.

8. The surface coating of claim 7 , wherein the damaged tissue is heart tissue.
