Abstract

The present disclosure provides small molecule drugs and pharmaceutical compositions for the treatment and prevention of diseases related to the formation of Aβ42 oligomers in a subject. It further provides a method of reducing formation of or disrupting Aβ42 oligomers in a subject, the method comprising the step of administering to the subject in need thereof a therapeutically effective amount of a pharmaceutical composition. One such compound is a compound of formula 120:

Claims (28)

1. A compound of Formula 120:

or a pharmaceutically acceptable salt, stereoisomer, or tautomer thereof, wherein:

R 1 is H;

R 3 is H;

R 4 is H;

R 5 is H;

R 6 is alkynyl;

R 7 is H;

R 8 is H;

R 20 is H, alkyl, alkyl(Oaryl), alkyl(aryl), alkyl(heterocyclyl), alkyl(heteroaryl), alkenyl, or alkynyl;

R 21 is H, alkyl, alkyl(Oaryl), alkyl(heterocyclyl), alkyl(heteroaryl), alkenyl, or alkynyl;

R 22 is H, NR′R″, or NR′C(O)R;

R 23 is H, alkyl, alkyl(Oaryl), alkyl(heterocyclyl), alkyl(heteroaryl), alkenyl, or alkynyl;

R 24 is H, alkyl, alkyl(Oaryl), alkyl(aryl), alkyl(heterocyclyl), alkyl(heteroaryl), alkenyl, or alkynyl;

R 25 is H, alkyl, alkyl(Oaryl), alkyl(aryl), alkyl(heterocyclyl), alkyl(heteroaryl), alkenyl, or alkynyl;

R 26 is halogen;

R 27 is H, alkyl, alkyl(Oaryl), alkyl(aryl), alkyl(heterocyclyl), alkyl(heteroaryl), alkenyl, or alkynyl;

R 28 is H, alkyl, alkyl(Oaryl), alkyl(aryl), alkyl(heterocyclyl), alkyl(heteroaryl), alkenyl, or alkynyl;

R 29 is H, alkyl, alkyl(Oaryl), alkyl(aryl), alkyl(heterocyclyl), alkyl(heteroaryl), alkenyl, or alkynyl;

R is alkyl or aryl;

R′ is H, alkyl, or aryl; and

R″ is H, alkyl, or aryl.

2. The compound of claim 1 , or a pharmaceutically acceptable salt, stereoisomer, or tautomer thereof, wherein R 6 is C≡CH.

3. The compound of claim 1 , or a pharmaceutically acceptable salt, stereoisomer, or tautomer thereof, wherein R 22 is H.

4. The compound of claim 1 , or a pharmaceutically acceptable salt, stereoisomer, or tautomer thereof, wherein R 26 is F.

5. The compound of claim 1 , or a pharmaceutically acceptable salt, stereoisomer, or tautomer thereof, wherein R 26 is Cl.

6. The compound of claim 1 , wherein the compound is:

or a pharmaceutically acceptable salt or stereoisomer thereof.
