CD200 inhibitors and methods of use thereof
The present invention provides in certain embodiments compositions comprising at least one CD200 inhibitor, and methods of reversing or modulating immune suppression in a patient having a disease or disorder arising from abnormal cell growth, function or behavior, which method comprises administering to a patient in need thereof a CD200 inhibitor composition.
1. A method of reversing or modulating immune suppression in a patient having a cancer associated with overexpression of CD200, which method comprises administering to the patient in need thereof a composition comprising a CD200 inhibitor, wherein the CD200 inhibitor is a mutant of naturally-occurring CD200 peptide, wherein the CD200 inhibitor is 13 to 20 amino acids in length, and wherein the peptide comprises the amino acid sequence VTWQKKAAVSPEN (SEQ ID NO: 21).
2. The method of reversing or modulating immune suppression of claim 1 , further comprising administering
(i) a cancer vaccine;
(ii) a cancer vaccine that is a tumor lysate; or
(iii) a tumor lysate that is substantially devoid of CD200.
3. The method of reversing or modulating immune suppression of claim 1 , wherein the composition further comprises a pharmaceutically acceptable carrier and/or diluent and/or an adjuvant.
4. The method of reversing or modulating immune suppression of claim 3 , wherein the adjuvant is administered before, concurrently or after administration of composition.
5. The method of reversing or modulating immune suppression of claim 1 , wherein the composition further comprises a cancer therapy.

The method of reversing or modulating immune suppression of claim 1 , wherein the composition is administered at least twice, or wherein the administration comprises administering at least three doses of the composition, or the administration comprises administering at least five doses of the composition, or the administration comprises administering at least ten doses of the composition.
7. The method of reversing or modulating immune suppression of claim 6 , wherein at least two consecutive dosages of the administration are separated by an interval of about one week, or wherein at least two consecutive dosages of the administration are separated by an interval of about one month.

A method of reversing or modulating immune suppression of claim 1 , wherein the cancer is selected from glioblastoma, melanoma, prostate, endometrial, ovarian, breast, lung, head and neck, hepatocellular, thyroid, breast, ovary, cervix, prostate, testis, genitourinary tract, esophagus, larynx, glioblastoma, neuroblastoma, stomach, skin, keratoacanthoma, lung, epidermoid carcinoma, large cell carcinoma, non-small cell lung carcinoma (NSCLC), small cell carcinoma, lung adenocarcinoma, bone, colon, adenoma, pancreas, adenocarcinoma, thyroid, follicular carcinoma, undifferentiated carcinoma, papillary carcinoma, seminoma, melanoma, sarcoma, bladder carcinoma, liver carcinoma and biliary passages, kidney carcinoma, myeloid disorders, lymphoid disorders, hairy cells, buccal cavity and pharynx (oral), lip, tongue, mouth, pharynx, small intestine, colon-rectum, large intestine, rectum, brain and central nervous system, Hodgkin's lymphoma and leukemia cancers.
9. The method of reversing or modulating immune suppression of claim 1 , wherein the patient is a human.
10. The method of reversing or modulating immune suppression of claim 1 , further comprising administering a chemotherapy agent.
11. The method of reversing or modulating immune suppression of claim 10 , wherein the chemotherapy agent is sunititib, ontak, cyclophosphamide, gemcitabine, and/or retionoic acid.
12. A method of reversing or modulating immune suppression in a tumor microenvironment or sentinel lymph nodes in a patient having a cancer associated with overexpression of CD200, which method comprises administering to the patient in need thereof a composition of comprising a CD200 inhibitor, wherein the CD200 inhibitor is a mutant of naturally-occurring CD200 peptide, wherein the CD200 inhibitor is 13 to 20 amino acids in length, and wherein the peptide comprises the amino acid sequence VTWQKKAAVSPEN (SEQ ID NO: 21).
13. The method of reversing or modulating immune suppression in a tumor microenvironment or sentinel lymph nodes of claim 12 , further comprising administering
(i) a cancer vaccine;
(ii) a cancer vaccine that is a tumor lysate; or
(iii) a tumor lysate that is substantially devoid of CD200.
14. The method of reversing or modulating immune suppression in a tumor microenvironment or sentinel lymph nodes of claim 12 , wherein the composition further comprises a pharmaceutically acceptable carrier and/or diluent and/or an adjuvant.
15. The method of reversing or modulating immune suppression in a tumor microenvironment or sentinel lymph nodes of claim 12 , wherein the adjuvant is administered before, concurrently or after administration of composition.
16. The method of reversing or modulating immune suppression in a tumor microenvironment or sentinel lymph nodes of claim 12 , wherein the composition further comprises a cancer therapy.
17. The method of reversing or modulating immune suppression in a tumor microenvironment or sentinel lymph nodes of claim 12 , wherein the composition is administered at least twice, or wherein the administration comprises administering at least three doses of the composition, or the administration comprises administering at least five doses of the composition, or the administration comprises administering at least ten doses of the composition.

The method of reversing or modulating immune suppression in a tumor microenvironment or sentinel lymph nodes of claim 17 , wherein at least two consecutive dosages of the administration are separated by an interval of about one week, or wherein at least two consecutive dosages of the administration are separated by an interval of about one month.

A method of reversing or modulating immune suppression in a tumor microenvironment or sentinel lymph nodes of claim 12 , wherein the cancer is selected from glioblastoma, melanoma, prostate, endometrial, ovarian, breast, lung, head and neck, hepatocellular, thyroid, breast, ovary, cervix, prostate, testis, genitourinary tract, esophagus, larynx, glioblastoma, neuroblastoma, stomach, skin, keratoacanthoma, lung, epidermoid carcinoma, large cell carcinoma, non-small cell lung carcinoma (NSCLC), small cell carcinoma, lung adenocarcinoma, bone, colon, adenoma, pancreas, adenocarcinoma, thyroid, follicular carcinoma, undifferentiated carcinoma, papillary carcinoma, seminoma, melanoma, sarcoma, bladder carcinoma, liver carcinoma and biliary passages, kidney carcinoma, myeloid disorders, lymphoid disorders, hairy cells, buccal cavity and pharynx (oral), lip, tongue, mouth, pharynx, small intestine, colon-rectum, large intestine, rectum, brain and central nervous system, Hodgkin's lymphoma and leukemia cancers.
20. The method of reversing or modulating immune suppression in a tumor microenvironment or sentinel lymph nodes of claim 12 , wherein the patient is a human.
21. The method of reversing or modulating immune suppression in a tumor microenvironment or sentinel lymph nodes of claim 12 , further comprising administering a chemotherapy agent.
22. The method of reversing or modulating immune suppression in a tumor microenvironment or sentinel lymph nodes of claim 12 , wherein the chemotherapy agent is sunititib, ontak, cyclophosphamide, gemcitabine, and/or retionoic acid.
