Abstract

The technology described herein generally relates to systems for extracting polynucleotides from multiple samples, particularly from biological samples, and additionally to systems that subsequently amplify and detect the extracted polynucleotides. The technology more particularly relates to microfluidic systems that carry out PCR on multiple samples of nucleotides of interest within microfluidic channels, and detect those nucleotides.

Claims (27)

1. A diagnostic system comprising:

a liquid dispenser configured to aspirate or dispense fluid, the liquid dispenser comprising:

a plurality of dispense heads, wherein each dispense head is configured to accept a pipette tip from a holder;

one or more valves;

wherein by controlling flow of gas through the one or more valves, pressure above the plurality of dispense heads is configured to be varied so that fluid is drawn up into or expelled from a pipette tip attached to a respective dispense head;

a sample tube holder comprising a first rack portion configured to be disposed around an upper portion of a sample tube containing a nucleic-acid containing sample, the sample tube holder further comprising a second rack portion configured to be disposed around a lower portion of the sample tube, wherein a middle portion the sample tube between the upper portion of the sample tube and the lower portion of the sample tube is visible when the sample tube is received in the sample tube holder;

a mirror positioned relative to the sample tube holder;

a barcode scanner configured to read a barcode of the sample tube containing the nucleic-acid containing sample when the sample tube is received in the sample tube holder, wherein the barcode is disposed on the middle portion of the sample tube, wherein the barcode scanner has a non-linear field of view from the barcode scanner to the mirror to the middle portion of the sample tube, enabling the barcode scanner to detect light reflected by the mirror from the sample tube; and

a stripper plate comprising a defined hole structure, wherein the stripper plate is configured to move relative to the pipette tip to detach the pipette tip from the respective dispense head.

2. The diagnostic system of claim 1 , wherein the liquid dispenser is configured to carry out fluid transfer operations simultaneously.

3. The diagnostic system of claim 1 , further comprising a horizontal rail along which the liquid dispenser moves in an x-direction.

4. The diagnostic system of claim 1 , further comprising one or more encoded stepper motors.

5. The diagnostic system of claim 1 , further comprising a variable force sensor configured to control an extent of vertical motion of the liquid dispenser during pipette tip pick-up and fluid dispensing operations.

6. The diagnostic system of claim 1 , wherein the plurality of dispense heads are arranged parallel to one another.

7. The diagnostic system of claim 1 , wherein the liquid dispenser is configured to accept or dispense, in a single operation, an amount of 1.0 ml of fluid or less.

8. The diagnostic system of claim 1 , wherein the barcode scanner moves with the liquid dispenser.

9. The diagnostic system of claim 1 , wherein the barcode scanner is configured to read a mirror image barcode.

10. The diagnostic system of claim 1 , wherein the barcode scanner is configured to communicate sample identifying information to a processor, thereby permitting sample identifying information to be associated with diagnostic results.

11. The diagnostic system of claim 1 , further comprising a sensor configured to detect a presence of the pipette tip.

12. The diagnostic system of claim 1 , wherein the stripper plate is configured to align pipette tips during dispensing of fluid.

13. The diagnostic system of claim 1 , wherein pipette tips extend perpendicular to the stripper plate.

14. The diagnostic system of claim 1 , wherein the stripper plate is configured to align pipette tips at a location closer to a bottom of the pipette tips.

15. The diagnostic system of claim 1 , wherein the stripper plate is configured to align pipette tips for dispensing fluid.

16. The diagnostic system of claim 1 , wherein the stripper plate is configured to be actuated by a stepper motor.

17. The diagnostic system of claim 1 , wherein the stripper plate is configured to move in a plane perpendicular to pipette tips.

18. The diagnostic system of claim 1 , wherein the barcode scanner is configured to read a barcode located on a top of a microfluidic cartridge.

19. The diagnostic system of claim 1 , wherein the barcode scanner is downward facing.
