Abstract

Claims (26)

1. A composition comprising: a delipidated, decellularized adipose tissue matrix derived from a first sample of adipose tissue which has been delipidated prior to decellularizing, wherein the delipidated, decellularized adipose tissue matrix contains a first proportion of lipids which is less than a second proportion of lipids contained in a processed adipose tissue derived from a second sample of adipose tissue which has been decellularized prior to delipidating; and a population of isolated viable adipose-derived stem cells (ASCs) seeded on the delipidated, decellularized adipose tissue matrix.

2. The composition of claim 1 , wherein the isolated viable ASCs have been cultured prior to seeding on the delipidated, decellularized adipose tissue matrix.

3. The composition of claim 1 , wherein the composition comprises a particulate form, an injectable form, or a combination thereof.

4. The composition of claim 1 , wherein the composition is dehydrated, partially dehydrated, or not dehydrated.

5. The composition of claim 1 , wherein the composition further comprises a carrier.

6. The composition of claim 1 , wherein the composition is a slurry, paste, or gel.

7. The composition of claim 1 , wherein the isolated viable ASCs comprise autologous cells, allogeneic cells, or a mixture thereof.

8. The composition of claim 7 , wherein at least a portion of the isolated viable ASCs comprise allogeneic cells derived from the first sample of adipose tissue.

9. The composition of claim 7 , wherein the composition is intended for implanting in or on a recipient patient and at least a portion of the isolated viable ASCs comprise autologous cells derived from the recipient patient.

10. A method of preparing a composition comprising a delipidated, decellularized adipose tissue matrix and a population of isolated viable adipose-derived stem cells (ASCs), comprising the steps of:

(a) obtaining a first adipose tissue;

(b) mechanically reducing the size of the first adipose tissue;

(c) delipidating the adipose tissue by contacting the first adipose tissue with a polar organic solvent such that lipids are transferred from the first adipose tissue to the polar organic solvent, thereby producing a delipidated adipose tissue from which substantially all of the lipids native to the adipose tissue have been removed;

(d) decellularizing the delipidated adipose tissue after step (c), thereby producing the delipidated, decellularized adipose tissue matrix which is essentially free of native cells and cellular components previously resident in the first adipose tissue;

(e) obtaining isolated viable adipose-derived stem cells (ASCs); and

(f) seeding the ASCs on the delipidated, decellularized adipose tissue matrix.

The method of claim 10 , wherein the step of (e) obtaining isolated viable ASCs comprises isolating viable ASCs from at least one sample of adipose tissue recovered from at least one living or deceased human donor to provide allogeneic isolated viable ASCs at least a portion of which are used in the step of (f) seeding.

12. The method of claim 11 , wherein the at least one sample of adipose tissue includes the first adipose tissue.

The method of claim 10 , wherein the composition is intended for implanting in or on a recipient patient and the step of (e) obtaining isolated viable ASCs comprises recovering autologous isolated viable ASCs from the recipient patient, at least a portion of which are used in the step of (f) seeding.

14. The method of claim 10 , wherein the step of (e) obtaining isolated viable ASCs comprises culturing the isolated viable ASCs.

The method of claim 10 , wherein the step of (f) seeding comprises the steps of: suspending the isolated viable ASCs in a medium to produce a suspended population of ASCs, and then adding the at least a portion of the suspended population of ASCs to a quantity of the delipidated, decellularized adipose tissue matrix to produce the composition.

16. The method of claim 15 , wherein the medium comprises basal medium, nutrient enriched medium, or a combination thereof.

17. The method of claim 15 , where in the step of (f) seeding further comprises: the steps of: incubating the composition at a temperature of greater than about 30° C., or rinsing the composition at least once with a buffered saline to wash away unwanted non-adherent ASCs, or both.

18. A method for treatment of a condition of a body feature, the method comprising implanting the composition of claim 1 in, on, proximate to, or a combination thereof, the body feature.

19. A method for treatment of a condition of a body feature, the method comprising implanting the composition of claim 3 in, on, proximate to, or a combination thereof, the body feature.

20. A method for treatment of a condition of a body feature, the method comprising implanting the composition of claim 5 in, on, proximate to, or a combination thereof, the body feature.
