Abstract

The present disclosure provides compositions, enema products, and enema delivery systems used for gastric, gastrointestinal and/or colonic treatments; and methods for making and using them. Methods of the invention are used for the stabilization, amelioration, treatment and/or prevention of constipation, for the treatment of abdominal pain, particularly non-specific abdominal pain, and diarrhea, including diarrhea caused by a drug side effect, a psychological condition, a disease or a condition such as Crohn's Disease, a poison, a toxin or an infection, e.g., a toxin-mediated traveler's diarrhea, or C. difficile or the pseudo-membranous colitis associated with this infection.

Claims (20)

1. An enema product configured for transporting to a remote facility, the enema product comprising a container and a pharmaceutical composition within the container, the pharmaceutical composition formulated for enema delivery directly from the container, wherein the container comprises an oxygen-resistant material, and wherein the pharmaceutical composition comprises saline, a cryoprotectant, and a microbiota of a stool sample comprising anaerobic bacteria, wherein particular bacterial species have not been pre-segregated from the microbiota, wherein the pharmaceutical composition comprises no more than 1% rough particulate matter of the stool sample, wherein the microbiota is in an amount effective for treating a C. difficile infection, and wherein the container is configured to provide a reduced oxygen environment to preserve the anaerobic bacteria.

2. The enema product of claim 1 , wherein the pharmaceutical composition comprises no more than 0.8% rough particulate matter of the stool sample.

3. The enema product of claim 1 , wherein the pharmaceutical composition comprises no more than 0.5% rough particulate matter of the stool sample.

4. The enema product of claim 1 , wherein the pharmaceutical composition comprises no more than 0.3% rough particulate matter of the stool sample.

5. The enema product of claim 1 , wherein the cryoprotectant comprises polyethylene glycol.

6. The enema product of claim 1 , wherein the enema product comprises flexible tubing for delivery of the pharmaceutical composition to a subject.

7. The enema product of claim 1 , wherein the stool sample is derived from a healthy human donor.

8. The enema product of claim 1 , wherein the stool sample has a weight of 50 grams.

9. The enema product of claim 1 , wherein the pharmaceutical composition is frozen.

10. An enema delivery system configured for transporting to a remote facility, the enema delivery system comprising a container, flexible tubing, and a pharmaceutical composition within the container, wherein the pharmaceutical composition is formulated for enema delivery from the container via the flexible tubing, wherein the pharmaceutical composition comprises saline, a cryoprotectant and a preparation of viable uncultured non-pathogenic fecal bacteria comprising anaerobic bacteria, wherein the fecal bacteria are from a stool of a human donor, wherein the container is sealed, wherein the pharmaceutical composition comprises no more than 1% rough particulate matter, wherein the pharmaceutical composition is in an amount effective for treating recurrence of C. difficile infection, and wherein the container is configured to provide a reduced oxygen environment to preserve the anaerobic bacteria.

11. The enema delivery system of claim 10 wherein the cryoprotectant comprises polyethylene glycol.

12. The enema delivery system of claim 10 , wherein the pharmaceutical composition comprises no more than 0.8% rough particulate matter of the stool sample.

13. The enema delivery system of claim 10 , wherein the pharmaceutical composition comprises no more than 0.5% rough particulate matter of the stool sample.

14. The enema delivery system of claim 10 wherein the pharmaceutical composition comprises no more than 0.3% rough particulate matter of the stool sample.

15. The enema delivery system of claim 10 , wherein the container comprises ethylene.

16. The enema delivery system of claim 10 , wherein the container comprises an oxygen-resistant material.

17. The enema delivery system of claim 10 , wherein the stool is derived from a healthy human donor.

18. The enema delivery system of claim 10 , wherein the fecal bacteria are separated from fiber of the stool.

19. The enema delivery system of claim 10 , wherein the pharmaceutical composition is frozen.

20. The enema delivery system of claim 10 , wherein the pharmaceutical composition further comprises an antioxidant.
