Human glycosylation enzymes
View Patent ↗The present invention relates to novel human glycosylation enzyme polypeptides and isolated nucleic acids containing the coding regions of the genes encoding such polypeptides. Also provided are vectors, host cells, antibodies, and recombinant methods for producing human glycosylation enzyme polypeptides. The invention further relates to diagnostic and therapeutic methods useful for diagnosing and treating disorders related to these novel human glycosylation enzyme polypeptides.
1. An isolated protein comprising an amino acid sequence selected from the group consisting of:
(a) amino acid residues 1 to 359 of SEQ ID NO:4; and
(b) amino acid residues 2 to 359 of SEQ ID NO:4.
2. The protein of claim 1 , wherein the amino acid sequence is (a).
3. The protein of claim 1 , wherein the amino acid sequence is (b).
4. The protein of claim 1 wherein the amino acid sequence further comprises a heterologous polypeptide.
5. The protein of claim 1 wherein said protein is glycosylated.
6. The protein of claim 1 wherein said protein is fused to polyethylene glycol.
7. An isolated protein produced by a method comprising:
(a) expressing the protein of claim 1 in a recombinant host cell comprising a nucleic acid molecule encoding said protein; and
(b) recovering the protein from the cell culture.
8. A composition comprising the protein of claim 1 and a carrier.
9. An isolated protein comprising an amino acid sequence selected from the group consisting of:
(a) the amino acid sequence of the full-length Sialic Acid Synthetase polypeptide encoded by the HA5AA37 cDNA clone contained in ATCC Deposit No. PTA-1410; and
(b) the amino acid sequence of the full-length Sialic Acid Synthetase polypeptide, excluding the N-terminal methionine residue, encoded by the HA5AA37 cDNA clone contained in ATCC Deposit No. PTA-1410.
10. The protein of claim 9 , wherein the amino acid sequence is (a).
11. The protein of claim 9 , wherein the amino acid sequence is (b).
12. The protein of claim 9 wherein the amino acid sequence further comprises a heterologous polypeptide.
13. The protein of claim 9 wherein said protein is glycosylated.
14. The protein of claim 9 wherein said protein is fused to polyethylene glycol.
15. An isolated protein produced by a method comprising:
(a) expressing the protein of claim 9 in a recombinant host cell comprising a nucleic acid molecule encoding said protein; and
(b) recovering the protein from the cell culture.
16. A composition comprising the protein of claim 9 and a carrier.
17. An isolated polypeptide consisting of a fragment of SEQ ID NO:4, wherein said fragment is at least 30 contiguous amino acid residues in length.
18. The polypeptide of claim 17 wherein the fragment is at least 50 contiguous amino acid residues in length.
19. The polypeptide of claim 17 fused to a heterologous polypeptide.
20. The polypeptide of claim 17 wherein said protein is glycosylated.
21. The polypeptide of claim 17 wherein said protein is fused to polyethylene glycol.
22. An isolated polypeptide produced by a method comprising:
(a) expressing the polypeptide of claim 17 in a recombinant host cell comprising a nucleic acid molecule encoding said polypeptide; and
(b) recovering the polypeptide from the cell culture.
23. A composition comprising the polypeptide of claim 17 and a carrier.
24. An isolated polypeptide consisting of a fragment of the full-length Sialic Acid Synthetase polypeptide encoded by the HA5AA37 cDNA clone contained in ATCC Deposit No. PTA-1410, wherein said fragment is at least 30 contiguous amino acid residues in length.
25. The polypeptide of claim 24 wherein the fragment is at least 50 contiguous amino acid residues in length.
26. The polypeptide of claim 24 fused to a heterologous polypeptide.
27. The polypeptide of claim 24 wherein said polypeptide is glycosylated.
28. The polypeptide of claim 24 wherein said polypeptide is fused to polyethylene glycol.
29. An isolated polypeptide produced by a method comprising:
(a) expressing the polypeptide of claim 24 in a recombinant host cell comprising a nucleic acid molecule encoding said polypeptide; and
(b) recovering the polypeptide from the cell culture.
30. A composition comprising the polypeptide of claim 24 and a carrier.
31. An isolated polypeptide consisting of a fragment of the polypeptide of SEQ ID NQ:4 with sialic acid synthetase activity.
32. The polypeptide of claim 31 wherein the polypeptide is fused to a heterologous polypeptide.
33. The polypeptide of claim 31 wherein said protein is glycosylated.
34. The polypeptide of claim 31 wherein said polypeptide is fused to polyethylene glycol.
35. An isolated polypeptide produced by a method comprising:
(a) expressing the polypeptide of claim 31 in a recombinant host cell comprising a nucleic acid molecule encoding said polypeptide; and
(b) recovering the polypeptide from the cell culture.
36. A composition comprising the polypeptide of claim 31 and a carrier.
