Abstract

The present invention relates to phosphonate compounds, compositions containing them, processes for obtaining them, and their use for treating a variety of medical disorders, e.g., osteoporosis and other disorders of bone metabolism, cancer, viral infections, and the like.

Claims (24)

1. An antiviral nucleoside compound, wherein the 5′-hydroxyl group has been substituted for a phosphonate or methyl phosphonate that is covalently linked to an alkylethanediol.

2. The compound of claim 1 , wherein the compound is selected from the group consisting of 2-chloro-deoxyadenosine, 1-β-D-arabinofuranosyl-cytidine (ara-C), fluorouridine, fluorodeoxyuridine (floxuridine), gemcitabine, cladribine, fludarabine, pentostatin (2′-deoxycoformycin), 1-β-D-arabinofuranosyl-guanine (ara-G), 1-β-D-arabinofuranosyl-adenosine (ara-A), and 1-β-D-arabinofuranosyl-uridine (ara-U).

A method for the treatment of a viral disease selected from human immunodeficiency virus, influenza, herpes simplex virus, human herpes virus, cytomegalovirus, hepatitis B and C virus, Epstein-Barr virus, varicella zoster virus, orthopox virus, ebola virus and papilloma virus comprising administering an effective amount of a compound of claim 1 optionally in a pharmaceutically acceptable carrier.

A method for the treatment of a viral disease selected from human immunodeficiency virus, influenza, herpes simplex virus, human herpes virus, cytomegalovirus, hepatitis B and C virus, Epstein-Barr virus, varicella zoster virus, orthopox virus, ebola virus and papilloma virus comprising administering an effective amount of a compound of claim 2 optionally in a pharmaceutically acceptable carrier.

5. The method of claim 3 , wherein orthopox is selected from variola major and minor, vaccinia, smallpox, cowpox, camelpox and monkeypox.

6. The method of claim 3 , wherein the virus is HIV.

7. The methods of claim 3 , wherein the virus is influenza.

8. The method of claim 3 , wherein the virus is herpes.

9. The method of claim 3 , wherein the virus is cytomegalovirus.

10. The method of claim 3 , wherein the virus is hepatitis B.

11. The method of claim 3 , wherein the virus is hepatitis C.

12. The method of claim 3 , wherein the virus is Epstein-Barr virus.

13. The method of claim 3 , wherein the virus is varicella zoster virus.

14. The method of claim 3 , wherein the virus is papilloma.

15. The method of claim 4 , wherein orthopox is selected from variola major and minor, vaccinia, smallpox, cowpox, camelpox and monkeypox.

16. The method of claim 4 , wherein the virus is HIV.

17. The method of claim 4 , wherein the virus is influenza.

18. The method of claim 4 , wherein the virus is herpes.

19. The method of claim 4 , wherein the virus is cytomegalovirus.

20. The method of claim 4 , wherein the virus is hepatitis B.

21. The method of claim 4 , wherein the virus is hepatitis C.

22. The method of claim 4 , wherein the virus is Epstein-Barr virus.

23. The method of claim 4 , wherein the virus is varicella zoster virus.

24. The method of claim 4 , wherein the virus is papilloma.
