TSG101-GAG interaction and use thereof
Isolated protein complexes are provided comprising Tsg101 and HIV GAG or GAGp6. The protein complexes are useful in screening assays for selecting compounds effective in modulating the Tsg101-HIV GAG or GAGp6 interaction within the protein complexes.
1. A method of inhibiting the binding of the human tumor susceptibility gene 101 protein (Tsg101) to the human immunodeficiency virus (HIV) GAG polyprotein (HIV GAG) comprising contacting Tsg101 with an antibody immunoreactive with the ubiquitin E2 variant (UEV) domain of Tsg101, in the presence of said HIV GAG, wherein said antibody inhibits said binding.
2. The method of claim 1 , wherein said binding of Tsg101 to HIV GAG occurs between the UEV domain of Tsg101 and the late assembly domain of HIV GAG p6.
3. The method of claim 1 , wherein said antibody is selectively immunoreactive with the UEV domain of Tsg101.
4. The method of claim 1 , wherein said antibody is a polyclonal antibody.
5. The method of claim 1 , wherein said antibody is a monoclonal antibody.
6. The method of claim 1 , wherein said antibody is an antibody fragment.
7. The method of claim 1 , wherein said antibody is a single chain antibody.
8. The method of claim 1 , wherein said antibody is a humanized antibody.
