Abstract

A rotatable rotor for separating components contained in a fluid, comprising an annular base having a first annular channel and an annular cover having a second annular channel for holding a flexible doughnut-shaped centrifuge bag therebetween. The cover has one or more radially spaced apart concentric indicator lines for monitoring the separation of the components. When the base and cover are superimposed, the first and second channels define an annular interior chamber having an off-centered figure eight configuration. The base and cover each comprise a grooved column axially centered and extending from the base top surface and the cover bottom surface, respectively. The flexible bag comprises inner and outer tubes extending from the core of the bag and which are seated in the column grooves to ensure that the bag is fixably rotatable with the rotor.

Claims (39)

1. A centrifuge apparatus for separating a predetermined component from a fluid, comprising:

a) a flexible centrifuge bag comprising:

upper and lower flexible sheets, the sheets having doughnut shaped configurations, inner perimeters defining a central core of the centrifuge bag, and outer perimeters, wherein the upper and lower sheets are superimposed and completely sealed together at their outer and inner perimeters;

a first compartment for receiving the fluid and separating the fluid into at least a first fraction and a second fraction;

a second compartment for separating the predetermined component from the first fraction;

a channel fluidly communicating the first compartment with the second compartment;

an inlet tube for introducing the fluid into the first compartment; and

an outlet tube for removing the predetermined component from the second compartment; and

b) a rotatable rotor comprising an annular base having a first annular channel and an annular cover having a second channel for holding the flexible centrifuge bag therebetween, whereby when the base and the cover are superimposed, the first and second annular channels define an annular interior chamber having an off-centered figure eight configuration, the cover further comprising one or more radially spaced apart concentric indicator lines for monitoring the separation of the predetermined component.

2. The apparatus of claim 1 , wherein the inlet tube extends radially from the central core and is sealed between the upper and lower sheets at the inner perimeter.

3. The apparatus of claim 1 , wherein the second compartment comprises diametrically opposed first and second wells for collecting and holding the predetermined component.

The apparatus of claim 3 , wherein the outlet tube extends across the diameter of the core and is sealed between the upper and lower sheets at the inner perimeter of the bag, wherein one end of the outlet tube is proximal to the first well and the other end of the outlet tube is proximal to the second well.

5. The apparatus of claim 1 , wherein the inlet and outlet tubes are fluidly connected to a multiple lumen tubing comprising an inlet lumen and an outlet lumen.

6. The apparatus of claim 5 , wherein the inlet tube is fluidly connected at its proximal end to the inlet lumen.

7. The apparatus of claim 5 , wherein the outlet tube is fluidly connected at its axis to the outlet lumen.

8. The apparatus of claim 5 , wherein the tubing is prevented from completely rotating about its axis during rotation of the centrifuge rotor.

The apparatus of claim 1 , wherein the base further comprises a first outer rim extending from the base top surface and a first grooved column axially centered and extending from the base top surface, wherein the height of the first rim is equal to the height of the first column, the first rim and the first column defining the first annular channel, and wherein the cover further comprises a second outer rim extending from the cover bottom surface and a second grooved column axially centered and extending from the cover bottom surface, wherein the height of the second rim is equal to the height of the second column, the second rim and the second column defining the second annular channel.

10. The apparatus of claim 9 , wherein when the centrifuge bag is positioned between the cover and base and wherein the tubes are seated in the grooves.

11. The apparatus of claim 1 , wherein the distal end of the outlet tube is positioned relative to a predetermined indicator line.

12. A method for removing a predetermined component from a fluid, comprising

a) providing a flexible centrifuge bag comprising:

upper and lower flexible sheets, the sheets having doughnut shaped configurations, inner perimeters defining a central core of the centrifuge bag, and outer perimeters, wherein the upper and lower sheets are superimposed and completely sealed together at their outer and inner perimeters;

a first compartment for receiving the fluid and separating the fluid into at least a first fraction and a second fraction;

a second compartment for separating the predetermined component from the first fraction;

a channel fluidly communicating the first compartment with the second compartment;

an inlet tube for introducing the fluid into the first compartment; and

an outlet tube for removing the predetermined component from the second compartment;

b) providing a centrifuge rotor comprising an annular base having a first annular channel and an annular cover having a second channel for holding the flexible centrifuge bag therebetween, whereby when the base and the cover are superimposed, the first and second annular channels define an annular interior chamber having an off-centered figure eight configuration, the cover further comprising one or more radially spaced apart concentric indicator lines for monitoring the separation of the predetermined component;

c) securing the flexible centrifuge bag in said centrifuge rotor, whereby the centrifuge bag is fixably rotatable with the centrifuge;

d) rotating the centrifuge rotor;

e) providing the fluid to the centrifuge bag via the inlet tube; and

f) removing the predetermined component from the second compartment via the outlet tube.

13. The method of claim 12 , further comprising a multiple lumen tubing having an inlet lumen and an outlet lumen, wherein the inlet tube is fluidly connected at its proximal end to the inlet lumen and the outlet tube is fluidly connected at its axis to the outlet lumen.

14. The method of claim 13 , wherein the tubing is prevented from completely rotating about its axis during rotation of the centrifuge rotor.

15. The method of claim 12 , wherein the fluid is whole blood.

16. The method of claim 12 , wherein the predetermined component is selected from the group consisting of platelets, platelet rich plasma, platelet poor plasma, white blood cells and red blood cells.

17. The method of claim 12 , wherein the inlet tube extends radially from the central core and is sealed between the upper and lower sheets at the inner perimeter.

18. The method of claim 12 , wherein the second chamber comprises diametrically opposed first and second wells for collecting and holding the predetermined component.

19. The method of claim 12 , wherein the base further comprises a first outer rim extending from the base top surface and a first grooved column axially centered and extending from the base top surface, wherein the height of the first rim is equal to the height of the first column, the first rim and the first column defining the first annular channel, and wherein the cover further comprises a second outer rim extending from the cover bottom surface and a second grooved column axially centered and extending from the cover bottom surface, wherein the height of the second rim is equal to the height of the second column, the second rim and the second column defining the second annular channel.
