Abstract

Protein formulations and methods for reducing the viscosity of a protein formulation are provided. The method for reducing the viscosity of a protein formulation comprises adding a viscosity reducing agent, such as calcium chloride or magnesium chloride to the protein formulation.

Claims (40)

1. A method for reducing the viscosity of an antibody formulation, the method comprising;

(a) providing the antibody formulation; and

(b) adding magnesium chloride to the antibody formulation to a concentration of between about 0.5 mM and about 20 mM, wherein the viscosity of the antibody formulation with the magnesium chloride is reduced compared to the viscosity of an antibody formulation without magnesium chloride.

2. The method of claim 1 , wherein the antibody is a humanized antibody.

3. The method of claim 1 , wherein the antibody is an antigen-binding fragment.

4. The method of claim 1 , wherein the antibody is an anti-myostatin antibody or an antigen-binding fragment thereof, an anti-IL-12 antibody or an antigen-binding fragment thereof, or an anti-IL-13 antibody or an antigen-binding fragment thereof.

5. The method of claim 4 , wherein the anti-myostatin antibody is MYO-029, wherein the anti-IL-12 antibody is J695, and wherein the anti-IL-13 antibody is IMA-638.

6. The method of claim 1 , wherein the viscosity of the antibody formulation is reduced by at least about 5% compared to the viscosity of the formulation in the absence of magnesium chloride.

7. A method for reducing the viscosity of an antibody formulation, the method comprising

(i) providing the antibody formulation; and

(ii) adding magnesium chloride to the antibody formulation, wherein the concentration of magnesium chloride in the formulation is between about 1 mM and about 25 mM.

8. The method of claim 1 , wherein the method further comprises determining the stability of the antibody formulation.

A method for reducing the viscosity of an antibody formulation, wherein the antibody of the formulation is selected from the group consisting of an anti-myostatin antibody or an antigen-binding fragment thereof, an anti-IL-12 antibody or an antigen-binding fragment thereof, and an anti-IL-13 antibody or an antigen-binding fragment thereof, the method comprising adding calcium chloride or magnesium chloride to the antibody formulation to a concentration of between about 0.5 mM and about 25 mM, wherein the viscosity of the antibody formulation with the calcium chloride or magnesium chloride is reduced compared to the viscosity of an antibody formulation without calcium chloride or magnesium chloride.

10. The method of claim 9 , wherein the antibody of the formulation is an anti-myostatin antibody or an antigen-binding fragment thereof.

11. The method of claim 10 , wherein the anti-myostatin antibody is MYO-022, MYO-028, or MYO-029.

12. The method of claim 9 , wherein the antibody of the formulation is an anti-IL-12 antibody or an antigen-binding fragment thereof.

13. The method of claim 12 , wherein the anti-IL-12 antibody is J695.

14. The method of claim 9 , wherein the antibody of the formulation is an anti-IL-13 antibody or an antigen-binding fragment thereof.

15. The method of claim 14 , wherein the anti-IL-13 antibody is IMA-638.

16. The method of claim 9 , wherein calcium chloride is added to the antibody formulation to a concentration of 5 mM.

17. The method of claim 9 , wherein calcium chloride is added to the antibody formulation to a concentration of 10 mM.

18. The method of claim 9 , wherein calcium chloride is added to the antibody formulation to a concentration of 20 mM.

19. The method of claim 9 , wherein the antibody formulation has a concentration of between about 50 mg/ml to about 200 mg/ml of the antibody.

20. The method of claim 9 , wherein the antibody formulation comprises about 4 mM to about 60 mM histidine, a cryoprotectant, and an antioxidant.

21. The method of claim 9 , wherein the antibody formulation further comprises 0.1% to 0.2% of a surfactant.

22. The method of claim 1 , wherein magnesium chloride is added to the antibody formulation to a concentration of 5 mM.

23. The method of claim 1 , wherein magnesium chloride is added to the antibody formulation to a concentration of 10 mM.

24. The method of claim 1 , wherein magnesium chloride is added to the antibody formulation to a concentration of 20 mM.

25. The method of claim 4 , wherein the antibody is an anti-myostatin antibody or an antigen-binding fragment thereof.

26. The method of claim 4 , wherein the antibody is an anti-IL-12 antibody or an antigen-binding fragment thereof.

27. The method of claim 4 , wherein the antibody is an anti-IL-13 antibody or an antigen-binding fragment thereof.

28. The method of claim 7 , wherein the antibody is an anti-myostatin antibody or an antigen-binding fragment thereof.

29. The method of claim 28 , wherein the anti-myostatin antibody is MYO-022, MYO-028, or MYO-029.

30. The method of claim 7 , wherein the antibody is an anti-IL-12 antibody or an antigen-binding fragment thereof.

31. The method of claim 30 , wherein the anti-IL-12 antibody is J695.

32. The method of claim 7 , wherein the antibody is an anti-IL-13 antibody or an antigen-binding fragment thereof.

33. The method of claim 32 , wherein the anti-IL-13 antibody is IMA-638.

34. The method of claim 7 , wherein magnesium chloride is added to the antibody formulation to a concentration of 2 mM.

35. The method of claim 7 , wherein magnesium chloride is added to the antibody formulation to a concentration of 15 mM.

36. The method of claim 7 , wherein magnesium chloride is added to the antibody formulation to a concentration of 25 mM.
