8-[3-amino-piperidin-1-yl]-xanthines, the preparation thereof and their use as pharmaceutical compositions
View Patent ↗The present invention relates to substituted xanthines of general formula wherein R 1 to R 3 are defined as in claims 1 to 16 , the tautomers, the stereoisomers, the mixtures, the prodrugs thereof and the salts thereof which have valuable pharmacological properties, particularly an inhibiting effect on the activity of the enzyme dipeptidylpeptidase-IV (DPP-IV).

(3) 1-(2-{2-[(aminocarbonyl)methoxy]-phenyl}-2-oxo-ethyl)-3-methyl-7-(3-methyl-2-buten-1-yl)-8-(3-amino-piperidin-1-yl)-xanthine,

3. A physiologically acceptable salt of a compound according to claim 1 or 2 with an inorganic or organic acid or base.
4. A pharmaceutical composition comprising a pharmaceutically acceptable amount of a compound according to claim 1 or 3 optionally together with one or more inert carriers and/or diluents.
5. A compound of formula (I)

R 1 is 4-methyl-2-quinazolinylmethyl;
R 2 is methyl; and
R 3 is 2-butyn-1-yl.
6. A pharmaceutical composition comprising one or more inert carriers or diluents and a compound of formula (I)
wherein
R 1 is 4-methyl-2-quinazolinylmethyl;
R 2 is methyl; and
R 3 is 2-butyn-1-yl.
7. A salt of a compound of formula (I)
wherein
R 1 is 4-methyl-2-quinazolinylmethyl;
R 2 is methyl; and
R 3 is 2-butyn-1-yl.
8. A physiologically acceptable salt of a compound of formula (I)
wherein
R 1 is 4-methyl-2-quinazolinylmethyl;
R 2 is methyl; and
R 3 is 2-butyn-1-yl.
9. A pharmaceutical composition comprising one or more inert carriers or diluents and a physiologically acceptable salt of a compound of formula (I)

R 1 is 4-methyl-2-quinazolinylmethyl;
R 2 is methyl; and
R 3 is 2-butyn-1-yl.
10. A salt of a compound of formula (I)
wherein
R 1 is 4-methyl-2-quinazolinylmethyl;
R 2 is methyl; and
R 3 is 2-butyn-1-yl,
with an inorganic or organic acid.
11. A physiologically acceptable salt of a compound of formula (I)
wherein
R 1 is 4-methyl-2-quinazolinylmethyl;
R 2 is methyl; and
R 3 is 2-butyn-1-yl,
with an inorganic or organic acid.
12. A pharmaceutical composition comprising one or more inert carriers or diluents and a physiologically acceptable salt of a compound of formula (I)

R 2 is methyl; and
R 3 is 2-butyn-1-yl,
with an inorganic or organic acid.
13. The compound 1-[(4-methyl-quinazolin-2-yl) methyl]-3-methyl-7-(2-butyn-1-yl)-8-(3-(R)-amino-piperidin-1-yl)-xanthine of the formula

with an inorganic or organic acid.
21. A pharmaceutical composition comprising a pharmaceutically acceptable amount of a physiologically acceptable salt according to claim 3 , optionally together with one or more inert carriers and/or diluents.
