Abstract

Analogs of bacteriocidal peptide microcin J25 (MccJ25) are provided that have an amino acid sequence that differs from that of MccJ25 by having at least one amino acid substitution; and that inhibit bacterial cell growth with a potency at least equal to that of MccJ25.

Claims (6)

1. An analog of bacteriocidal peptide microcin J25 (MccJ25) that (1) has an amino acid sequence that differs from that of MccJ25 by having at least one amino acid substitution; and (2) that inhibits bacterial cell growth with a potency at least equal to that of MccJ25, wherein the analog is selected from the group consisting of [Ser 3 ] MccJ25 (SEQ ID NO:2), [Ile 6 ] MccJ25 (SEQ ID NO:3), [Phe 6 ] MccJ25 (SEQ ID NO:4), [Ala 11 ] MccJ25 (SEQ ID NO:5), [Trp 11 ] MccJ25 (SEQ ID NO:6), [Thr 12 ] MccJ25 (SEQ ID NO:7), [Ser 12 ] MccJ25 (SEQ ID NO:8), [Met 12 ] MccJ25 (SEQ ID NO:9), [His 12 ] MccJ25 (SEQ ID NO:10) [Leu 13 ] MccJ25 (SEQ ID NO:11), [Ala 13 ] MccJ25 (SEQ ID NO:12), [Asn 13 ] MccJ25 (SEQ ID NO:13 [Arg 13 ] MccJ25 (SEQ ID NO:14), [Pro 13 ] MccJ25 (SEQ ID NO:15), [Thr 13 ] MccJ25 (SEQ ID NO:16), [His 13 ] MccJ25 (SEQ ID NO:17), [Val 13 ] MccJ25 (SEQ ID NO:18), [Met 13 ] MccJ25 (SEQ ID NO:19), [Phe 13 ] MccJ25 (SEQ ID NO:20), [Trp 13 ] MccJ25 (SEQ ID NO:21), [Ser 14 ] MccJ25 (SEQ ID NO:22), [Thr 14 ] MccJ25 (SEQ ID NO:23), [Phe 15 ] MccJ25 (SEQ ID NO:24), [Gly 15 ] MccJ25 (SEQ ID NO:25), [Leu 15 ] MccJ25 (SEQ ID NO:26), [His 15 ] MccJ25 (SEQ ID NO:27), [Asn 15 ] MccJ25 (SEQ ID NO:28), [Ala 15 ] MccJ25 (SEQ ID NO:29), [Ile 15 ] MccJ25 (SEQ ID NO:30), [Trp 15 ] MccJ25 (SEQ ID NO:31), [Met 15 ] MccJ25 (SEQ ID NO:32), [Gln 16 ] MccJ25 (SEQ ID NO:33), [Glu 17 ] MccJ25 (SEQ ID NO:34), [Val 17 ] MccJ25 (SEQ ID NO:35) and combinations thereof.

The analog of claim 1 , wherein the analog is selected from the group consisting of [Leu 13 ] MccJ25 (SEQ ID NO:11), [Ala 13 ] MccJ25 (SEQ ID NO:12), [Asn 13 ] MccJ25 (SEQ ID NO:13), [Arg 13 ] MccJ25 (SEQ ID NO:14), [Pro 13 ] MccJ25 (SEQ ID NO:15), [Thr 13 ] MccJ25 (SEQ ID NO:16), and [His 13 ] MccJ25 (SEQ ID NO:17).

3. The analog of claim 1 , wherein the analog is selected from the group consisting of [Phe 15 ] MccJ25 (SEQ ID NO:24), [Gly 15 ] MccJ25 (SEQ ID NO:25), [Leu 15 ] MccJ25 (SEQ ID NO:26), [His 15 ] MccJ25 (SEQ ID NO:27), [Asn 15 ] MccJ25 (SEQ ID NO:28), [Ala 15 ] MccJ25 (SEQ ID NO:29) and [Ile 15 ] MccJ25 (SEQ ID NO:30).

4. The analog of claim 1 , wherein the analog includes a detectable group.

5. The analog of claim 4 , wherein the detectable group is selected from the group consisting of chromophores, fluorophores, and cyanine dyes.

6. The analog of claim 4 , wherein the detectable group is located at a site of the amino acid substitution.
