Abstract

In accordance with the present invention, there are provided novel Siah-Mediated-Degradation-Proteins (SMDPs) and/or SCF-Complex Proteins (SCPs). Nucleic acid sequences encoding such proteins and assays employing same are also disclosed. The invention SMDPs and/or SCPs can be employed in a variety of ways, for example, for the production of anti-SMDP and/or SCP antibodies thereto, in therapeutic compositions, and methods employing such proteins and/or antibodies for drug screening, functional genomics and other applications. Also provided are transgenic non-human mammals that express the invention protein. Also provided are compositions and methods for targeting the destruction of selected polypeptides in eukaryotic cells based on the ubiquitin-independent mechanism by which ornithine decarboxylase is degraded by the 26S proteasome.

Claims (21)

1. An isolated polynucleotide that comprises a promoter sequence operably linked to a sequence that encodes a chimeric protein that comprises a target-protein binding domain operatively linked to a protein-degradation binding domain of ornithine decarboxylase and a flexible linker sequence [Gly-Gly-Gly-Gly-Ser] 3 (SEQ ID NO: 61) between the target-protein binding domain and the protein-degradation binding domain of ornithine decarboxylase, wherein binding of the target-protein binding domain of the chimeric protein to a target protein in a cell induces degradation of the target protein in the cell.

2. The isolated polynucleotide of claim 1 , wherein the protein-degradation binding domain of ornithine decarboxylase is an ornithine decarboxylase C-terminal region polypeptide.

3. The isolated polynucleotide of claim 1 , wherein the protein-degradation binding domain of ornithine decarboxylase is a human ornithine decarboxylase C-terminal region polypeptide.

4. The isolated polynucleotide of claim 1 , wherein the chimeric protein comprises, in order from N-terminus to C-terminus, (a) the target-protein binding domain, (b) the flexible linker sequence, and (c) the protein-degradation binding domain of ornithine decarboxylase.

5. The isolated polynucleotide of claim 1 , wherein the chimeric protein further comprises an epitope tag sequence.

6. The polynucleotide of claim 1 , further comprising a second promoter sequence operably linked to a polynucleotide sequence that encodes an antizyme protein or a fragment of an antizyme protein that has antizyme activity.

7. The polynucleotide of claim 6 wherein the polynucleotide encodes a fragment of antizyme protein that has antizyme activity.

8. A cell comprising an isolated polynucleotide of claim 1 .

9. An isolated polynucleotide that comprises

(a) a first promoter sequence operably linked to a sequence that encodes a chimeric protein that comprises a target-protein binding domain operatively linked to a protein-degradation binding domain of ornithine decarboxylase, and

(b) a second promoter sequence operably linked to a sequence that encodes a human antizyme protein or a fragment of a human antizyme protein that has antizyme activity,

wherein binding of the target-protein binding domain of the chimeric protein to a target protein in a cell induces degradation of the target protein in the cell.

10. The isolated polynucleotide of claim 9 , wherein the chimeric protein further comprises a linker sequence between the target-protein binding domain and the protein-degradation binding domain of ornithine decarboxylase.

11. The isolated polynucleotide of claim 10 , wherein the linker sequence is a flexible linker sequence.

12. The isolated polynucleotide of claim 11 , wherein the flexible linker sequence is [Gly-Gly-Gly-Gly-Ser] 3 (SEQ ID NO: 61).

13. The isolated polynucleotide of claim 11 , wherein the chimeric protein comprises, in order from N-terminus to C-terminus, (a) the target-protein binding domain, (b) the flexible linker sequence, and (c) the protein-degradation binding domain of ornithine decarboxylase.

14. The isolated polynucleotide of claim 9 , wherein the protein-degradation binding domain of ornithine decarboxylase is an ornithine decarboxylase C-terminal region polypeptide.

15. The isolated polynucleotide of claim 9 , wherein the protein-degradation binding domain of ornithine decarboxylase is a human ornithine decarboxylase C-terminal region polypeptide.

16. The polynucleotide of claim 9 , wherein the second promoter sequence is operably linked to a polynucleotide sequence that encodes a fragment of antizyme protein that has antizyme activity.

17. The isolated polynucleotide of claim 9 , wherein the chimeric protein further comprises an epitope tag sequence.

18. A kit comprising: (a) a composition comprising an isolated polynucleotide that comprises a promoter sequence operably linked to a sequence that encodes a chimeric protein that comprises a target-protein binding domain operatively linked to a protein-degradation binding domain of ornithine decarboxylase and a flexible linker sequence [Gly-Gly-Gly-Gly-Ser] 3 (SEQ ID NO: 61) between the target-protein binding domain and the protein-degradation binding domain of ornithine decarboxylase, wherein binding of the target-protein binding domain of the chimeric protein to a target protein in a cell induces degradation of the target protein in the cell; and (b) a composition comprising an isolated polynucleotide that comprises a promoter sequence operably linked to a sequence that encodes an antizyme protein or encodes a fragment of antizyme protein that has antizyme activity; and (c) instructions for use.
