Abstract

The invention provides the identification and characterization of disease and cancer-associated antigen, RAAG10. The invention also provides a family of monoclonal antibodies that bind to antigen RAAG10, methods of diagnosing and treating various human cancers and diseases that express RAAG10.

Claims (45)

1. An isolated cell line selected from the group consisting of ATCC No. PTA-4217, ATCC No. PTA-4218, ATCC No. PTA-4244, and ATCC No. PTA-4245.

2. An isolated antibody that binds to B7H3L comprising three complementarity determining regions from the heavy chain and three complementarity determining regions from the light chain of the monoclonal antibody expressed by the cell line having ATCC No. PTA-4217.

3. The isolated antibody of claim 2 , wherein the isolated antibody is a humanized antibody.

4. The isolated antibody of claim 2 , wherein the isolated antibody is a chimeric antibody comprising a heavy chain variable region and a light chain variable region from the heavy chain and light chain variable regions of the monoclonal antibody expressed by the cell line having ATCC No. PTA-4217.

5. The isolated antibody of claim 4 , wherein the chimeric antibody comprises a heavy chain constant region and a light chain constant region from a human antibody.

6. An antigen-binding fragment of the isolated antibody of claim 2 , wherein the antigen-binding fragment is selected from the group consisting of a Fab, a Fab′, a F(ab′) 2 and a Fv; and wherein the antigen-binding fragment retains the binding specificity of the monoclonal antibody expressed by the cell line having ATCC No. PTA-4217.

7. An isolated antibody that binds to B7H3L comprising a heavy chain variable region from the heavy chain variable region of the monoclonal antibody expressed by the cell line having ATCC No. PTA-4217.

8. An isolated antibody that binds to B7H3L comprising a light chain variable region from the light chain variable region of the monoclonal antibody expressed by the cell line having ATCC No. PTA-4217.

9. An isolated antibody that binds B7H3L comprising three complementarity determining regions from the heavy chain and three complementarity determining regions from the light chain of the monoclonal antibody expressed by the cell line having ATCC No. PTA-4218.

10. The isolated antibody of claim 9 , wherein the isolated antibody is a humanized antibody.

11. The isolated antibody of claim 9 , wherein the isolated antibody is a chimeric antibody comprising a heavy chain variable region and a light chain variable region from the heavy chain and light chain variable regions of the monoclonal antibody expressed by the cell line having ATCC No. PTA-4218.

12. The isolated antibody of claim 11 , wherein the chimeric antibody comprises a heavy chain constant region and a light chain constant region from a human antibody.

13. An antigen-binding fragment of the isolated antibody of claim 9 , wherein the antigen-binding fragment is selected from the group consisting of a Fab, a Fab′, a F(ab′) 2 and a Fv; and wherein the antigen-binding fragment retains the binding specificity of the monoclonal antibody expressed by the cell line having ATCC No. PTA-4218.

14. An isolated antibody that binds to B7H3L comprising a heavy chain variable region from the heavy chain variable region of the monoclonal antibody expressed by the cell line having ATCC No. PTA-4218.

15. An isolated antibody that binds to B7H3L comprising a light chain variable region from the light chain variable region of the monoclonal antibody expressed by the cell line having ATCC No. PTA-4218.

16. An isolated antibody that binds B7H3L comprising three complementarity determining regions from the heavy chain and three complementarity determining regions from the light chain of the monoclonal antibody expressed by the cell line having ATCC No. PTA-4244.

17. The isolated antibody of claim 16 , wherein the isolated antibody is a humanized antibody.

18. The isolated antibody of claim 16 , wherein the isolated antibody is a chimeric antibody comprising a heavy chain variable region and a light chain variable region from the heavy chain and the light chain variable regions of the monoclonal antibody expressed by the cell line having ATCC No. PTA-4244.

19. The isolated antibody of claim 18 , wherein the chimeric antibody comprises a heavy chain constant region and a light chain constant region from a human antibody.

20. An antigen-binding fragment of the isolated antibody of claim 16 , wherein the antigen-binding fragment is selected from the group consisting of a Fab, a Fab′, a F(ab′) 2 and a Fv; and wherein the antigen-binding fragment retains the binding specificity of the monoclonal antibody expressed by the cell line having ATCC No. PTA-4244.

21. An isolated antibody that binds to B7H3L comprising a heavy chain variable region from the heavy chain variable region of the monoclonal antibody expressed by the cell line having ATCC No. PTA-4244.

22. An isolated antibody that binds to B7H3L comprising a light chain variable region from the light chain variable region of the monoclonal antibody expressed by the cell line having ATCC No. PTA-4244.

23. An isolated antibody that binds B7H3L comprising three complementarity determining regions from the heavy chain and three complementarity determining regions from the light chain of the monoclonal antibody expressed by the cell line having ATCC No. PTA-4245.

24. The isolated antibody of claim 23 , wherein the isolated antibody is a humanized antibody.

25. The isolated antibody of claim 23 , wherein the isolated antibody is a chimeric antibody comprising a heavy chain variable region and a light chain variable region from the heavy chain and the light chain variable regions of the monoclonal antibody expressed by the cell line having ATCC No. PTA-4245.

26. The isolated antibody of claim 25 , wherein the chimeric antibody comprises a heavy chain constant region and a light chain constant region from a human antibody.

27. An antigen-binding fragment of the isolated antibody of claim 23 , wherein the antigen-binding fragment is selected from the group consisting of a Fab, a Fab′, a F(ab′) 2 and a Fv; and wherein the antigen-binding fragment retains the binding specificity of the monoclonal antibody expressed by the cell line having ATCC No. PTA-4245.

28. An isolated antibody that binds to B7H3L comprising a heavy chain variable region from the heavy chain variable region of the monoclonal antibody expressed by the cell line having ATCC No. PTA-4245.

29. An isolated antibody that binds to B7H3L comprising a light chain variable region from the light chain variable region of the monoclonal antibody expressed by the cell line having ATCC No. PTA-4245.

30. An isolated antibody expressed by a cell line having ATCC No. PTA-4217, or an antigen-binding fragment thereof.

31. An isolated antibody expressed by a cell line having ATCC No. PTA-4218, or an antigen-binding fragment thereof.

32. An isolated antibody expressed by a cell line having ATCC No. PTA-4244, or an antigen-binding fragment thereof.

33. An isolated antibody expressed by a cell line having ATCC No. PTA-4245, or an antigen-binding fragment thereof.

34. A pharmaceutical composition comprising a therapeutically effective dose of the isolated antibody of claim 2 or an antigen-binding fragment thereof, to together with a pharmaceutically acceptable carrier.

35. The pharmaceutical composition of claim 34 , wherein the composition comprises an additional therapeutic moiety.

36. The pharmaceutical composition of claim 34 , wherein the isolated antibody is linked to a therapeutic agent.

37. A pharmaceutical composition comprising a therapeutically effective dose of the isolated antibody of claim 9 or an antigen-binding fragment thereof, together with a pharmaceutically acceptable carrier.

38. The pharmaceutical composition of claim 37 , wherein the composition comprises an additional therapeutic moiety.

39. The pharmaceutical composition of claim 37 , wherein the isolated antibody is linked to a therapeutic agent.

40. A pharmaceutical composition comprising a therapeutically effective dose of the isolated antibody of claim 16 or an antigen-binding fragment thereof, together with a pharmaceutically acceptable carrier.

41. The pharmaceutical composition of claim 40 , wherein the composition comprises an additional therapeutic moiety.

42. The pharmaceutical composition of claim 40 , wherein the isolated antibody is linked to a therapeutic agent.

43. A pharmaceutical composition comprising a therapeutically effective dose of the isolated antibody of claim 23 or an antigen-binding fragment thereof, together with a pharmaceutically acceptable carrier.

44. The pharmaceutical composition of claim 43 , wherein the composition comprises an additional therapeutic moiety.

45. The pharmaceutical composition of claim 43 , wherein the isolated antibody is linked to a therapeutic agent.
