Abstract

Methods and compositions for modulating plant development are provided. Nucleotide sequences and amino acid sequences encoding Ovule Development Protein 2 (ODP2) proteins are provided. The sequences can be used in a variety of methods including modulating development, developmental pathways, altering oil content in a plant, increasing transformation efficiencies, modulating stress tolerance, and modulating the regenerative capacity of a plant. Transformed plants, plant cells, tissues, and seed are also provided.

Claims (38)

1. An isolated polynucleotide that encodes an amino acid sequence having at least 95% sequence identity to SEQ ID NO:2,wherein said polynucleotide encodes a polypeptide that renders a plant cell embryogenic, when said polynucleotide is expressed in the plant cell.

2. An expression cassette comprising the polynucleotide of claim 1 , wherein said polynucleotide is operably linked to a promoter that drives expression in a plant.

3. A plant comprising a heterologous polynucleotide operably linked to a promoter that drives expression in the plant, wherein said polynucleotide encodes an amino acid sequence having at least 95% sequence identity to SEQ ID NO: 2, wherein said polynucleotide encodes a polypeptide that renders a plant cell embryogenic, when said polynucleotide is expressed in the plant cell.

4. The plant of claim 3 , wherein said plant comprises a plant part selected from the group consisting of a cell, a seed, and a grain.

5. The plant of claim 3 , wherein said promoter is a constitutive promoter, a tissue-preferred promoter, an inducible promoter, or a developmentally regulated promoter.

6. The plant of claim 3 , wherein said plant is a monocot.

7. The plant of claim 6 , wherein said monocot is maize, wheat, rice, barley, sorghum, or rye.

8. The plant of claim 3 , wherein said plant is a dicot.

9. The plant of claim 3 , wherein said heterologous polynucleotide is stably incorporated into the genome of the plant.

10. The plant of claim 3 ,wherein said plant has an increased level of a polypeptide selected from the group consisting of

(a) the polypeptide comprising the amino acid sequence of SEQ ID NO: 2; and,

(b) the polypeptide having at least 95% sequence identity to SEQ ID NO: 2, wherein said polypeptide renders a plant cell embryogenic, when said polynucleotide is expressed in the plant cell.

11. The plant of claim 3 , wherein said plant has a phenotype selected from the group consisting of:

(a) an altered oil phenotype;

(b) a modified regenerative capacity;

(c) an ability to produce an asexually derived embryo;

(d) an increased transformation efficiency;

(e) an altered starch production; and,

(f) an increased or maintained yield under abiotic stress.

A method of increasing activity of a polypeptide in a plant comprising introducing into said plant a heterologous polynucleotide that encodes an amino acid sequence having at least 95% sequence identity to SEQ ID NO: 2, wherein said polynucleotide encodes a polypeptide that renders a plant cell of said plant embryogenic, when said polynucleotide is expressed in the plant cell.

13. The method of claim 12 , wherein said heterologous polynucleotide is transiently expressed.

14. The method of claim 12 , wherein said heterologous polynucleotide is stably integrated into the genome of the plant.

15. The method of claim 12 , wherein said heterologous polynucleotide is operably linked to a constitutive promoter, a tissue-preferred promoter, an inducible promoter, or a developmentally regulated promoter.

16. The method of claim 12 , wherein said plant comprises a plant part selected from the group consisting of a plant cell and a seed.

17. The method of claim 12 , wherein said plant is a dicot.

18. The method of claim 12 , wherein said plant is a monocot.

19. The method of claim 18 , wherein said monocot is maize, wheat, rice, barley, sorghum, or rye.

20. The plant of claim 3 , wherein said promoter is an inducible promoter.

21. The method of claim 12 , wherein said heterologous polynucleotide is operably linked to an inducible promoter.

22. The isolated polynucleotide of claim 1 , wherein said polynucleotide comprises the sequence set forth in SEQ ID NO: 1 or 3.

23. The isolated polynucleotide of claim 1 , wherein said polynucleotide encodes the amino acid sequence of SEQ ID NO: 2.

24. The isolated polynucleotide of claim 1 wherein said polynucleotide has at least 95%sequence identity to SEQ ID NO: 1 or 3, wherein said polynucleotide encodes a polypeptide that renders a plant cell embryogenic, when said polynucleotide is expressed in the plant cell.

25. The plant of claim 3 , wherein said polynucleotide comprises the sequence set forth in SEQ ID NO:1 or 3.

26. The plant of claim 3 , wherein said polynucleotide encodes the amino acid sequence of SEQ ID NO:2.

27. The plant of claim 3 , wherein said polynucleotide has at least 95% sequence identity to SEQ ID NO: 1 or 3, wherein said polynucleotide encodes a polypeptide that renders a plant cell embryogenic, when said polynucleotide is expressed in the plant cell.

28. The method of claim 12 , wherein said polynucleotide comprises the sequence set forth in SEQ ID NO:1 or 3.

29. The method of claim 12 , wherein said polynucleotide encodes the amino acid sequence of SEQ ID NO:2.

30. The method of claim 12 , wherein said polynucleotide has at least 95% sequence identity to SEQ ID NO: 1 or 3, wherein said polynucleotide encodes a polypeptide that renders a plant cell embryogenic, when said polynucleotide is expressed in the plant cell.
