Abstract

A bioengineered synthesis scheme for the production of quinic acid from a carbon source is provided. Methods of producing quinic acid from a carbon source based on the synthesis scheme as well as conversion of quinic acid to hydroquinone are also provided.

Claims (24)

1. A recombinant microbe comprising:

(A) a 3-deoxy-D-arabino-heptulosonic acid-7-phosphate (DAHP) synthase enzyme that is insensitive to feedback inhibition by aromatic amino acids encoded by an aroF FBR gene from E. coli,

(B) a 3-dehydroquinate (DHQ) synthase enzyme encoded by an aroB gene from E. coli , and

(C) a shikimate dehydrogenase enzyme encoded by an aroE gene from E. coli,

wherein

(I) the gene of at least one of said enzymes is recombinant,

(II) the microbe's DNA that encodes dehydroquinate dehydratase has been inactivated so that the recombinant microbe does not cause dehydration of 3-dehydroquinate to dehydroshikimate, and

(III) the recombinant microbe is capable of producing quinic acid from a carbon source by a synthesis scheme that uses all three of said enzymes.

2. The recombinant microbe according to claim 1 , wherein the gene encoding the DAHP synthase enzyme is recombinant.

3. The recombinant microbe according to claim 1 , wherein the gene encoding the DHQ synthase enzyme is recombinant.

4. The recombinant microbe according to claim 1 , wherein the gene encoding the shikimate dehydrogenase enzyme is recombinant.

5. The recombinant microbe according to claim 1 , wherein the microbe is selected from the group consisting of fungi, yeasts, and bacteria.

6. The recombinant microbe according to claim 1 , wherein the aroE gene is comprised by a plasmid.

7. The recombinant microbe according to claim 6 , wherein the plasmid is pKD12.112.

8. The recombinant microbe according to claim 1 , wherein the recombinant microbe is a recombinant E. coli.

9. The recombinant microbe according to claim 1 , wherein the microbe's inactivated DNA that encodes dehydroquinate dehydratase comprises an inactivated aroD gene from E. coli.

10. The recombinant microbe according to claim 9 , wherein the recombinant microbe is E. coli QP1.1.

11. A recombinant microbe comprising a biosynthetic pathway for converting a carbon source to quinic acid, said biosynthetic pathway comprising:

(A) means for catalyzing the synthesis of 3-deoxy-D-arabino-heptulosonic acid-7-phosphate (DAHP) from phosphoenolpyruvate and erythrose-4-phosphate comprising a 3-deoxy-D-arabino-heptulosonic acid-7-phosphate (DAHP) synthase enzyme that is insensitive to feedback inhibition by aromatic amino acids encoded by an aroF FBR gene from E. coli;

(B) means for catalyzing the synthesis of 3-dehydroquinate (DHQ) from DAHP comprising a 3-dehydroquinate (DHQ) synthase enzyme encoded by an aroB gene from E. coli ; and

(C) means for catalyzing the synthesis of shikimic acid from 3-dehydroshikimic acid and for catalyzing the synthesis of quinic acid from DHQ comprising a shikimate dehydrogenase enzyme encoded by an aroE gene from E. coli;

wherein at least one of the aroF FBR gene, the aroB gene, and the aroE gene is recombinant.

12. The recombinant microbe of claim 11 wherein the microbe's DNA that encodes dehydroquinate dehydratase has been inactivated so that the recombinant microbe does not cause dehydration of 3-dehydroquinate to dehydroshikimate.

13. A recombinant microbe comprising E. coli QP1.1 pKD12.112.
