Abstract

The present invention features polypeptides, such as antibodies, and their use in the treatment and diagnosis of neoplasms.

Claims (22)

1. A purified antibody or antigen binding fragment thereof, comprising a light chain (V L ) variable region sequence and a heavy chain (V H ) variable region sequence, wherein the light chain (V L ) variable region sequence comprises amino acids 26-34, 52-58 and 97-103 of SEQ ID NO:5, or wherein the heavy chain (V H ) variable region sequence comprises amino acids 11-18, 36-43, and 82-100 of SEQ ID NO:7, and wherein said antibody or said antigen binding fragment specifically binds to an epitope of a polypeptide having an approximate molecular weight of 115 kDa using sodium dodecyl sulfate polyacrylamide gel electrophoresis, wherein said polypeptide is expressed by ASPC-1(ATCC Accession No. CRL-1682) or BXPC-3 (ATCC Accession No. CRL-1687) cells and wherein PM-2 antibody produced by a cell line deposited as DSM ACC2600 specifically binds to said epitope of the polypeptide having an approximate molecular weight of 115 kD expressed by ASPC-1 (ATCC Accession No. CRL-1682) or BXPC-3 (ATCC Accession No. CRL-1687) cell.

A purified antibody or antigen binding fragment thereof comprising a light chain (V L ) variable region sequence and a heavy chain (V H ) variable region sequence, wherein the light chain (V L ) variable region sequence comprises amino acids 26-34, 52-58 and 97-103 of SEQ ID NO:5, and wherein the heavy chain (V H ) variable region sequence comprises amino acids 11-18, 36-43, and 82-100 of SEQ ID NO:7.

3. The purified antibody or antigen binding fragment thereof of claim 1 , wherein said light chain (V L ) variable region sequence comprises amino acids 26-34, 52-58 and 97-103 of SEQ ID NO:5.

4. The purified antibody or antigen binding fragment thereof of claim 1 , wherein said heavy chain (V H )variable region sequence comprises amino acids 11-18, 36-43 and 82-100 of SEQ ID NO:7.

5. The purified antibody or antigen binding fragment thereof of claim 1 or 2 , wherein said light chain (V L ) variable region sequence comprises SEQ ID NO:5.

6. The purified antibody or antigen binding fragment thereof of claim 1 or 2 , wherein said heavy chain (V H ) variable region sequence comprises SEQ ID NO:7.

7. The purified antibody or antigen binding fragment thereof of claim 1 or 2 , wherein said antibody or said antigen binding fragment is a monoclonal antibody.

8. The purified antibody or antigen binding fragment thereof of claim 1 or 2 , wherein said antibody or said antigen binding fragment is a human monoclonal antibody.

The purified antibody or antigen binding fragment thereof of claim 1 , wherein said antigen binding fragment comprises a light chain (V L ) variable region sequence comprising amino acids 26-34, 52-58 and 97-103 of SEQ ID NO:5, and a heavy chain (V H ) variable region sequence comprising amino acids 11-18, 36-43, and 82-100 of SEQ ID NO:7.

The purified antigen binding fragment of claim 1 , wherein said antigen binding fragment comprises a light chain (V L ) variable region sequence comprising amino acids 26-34, 52-58 and 97-103 of SEQ ID NO:5, or a heavy chain (V H ) variable region sequence comprising amino acids 11-18, 36-43, and 82-100 of SEQ ID NO:7.

11. The purified antibody or antigen binding fragment thereof of claim 1 or 2 , wherein said antibody or antigen binding fragment thereof comprises the amino acid sequence of SEQ ID NO:5 or SEQ ID NO:7.

12. The purified antibody or antigen binding fragment thereof of claim 1 or 2 , wherein said antigen binding fragment is selected from the group consisting of F v , Fab, Fab′, and F(ab′) 2 .

13. The purified antibody or antigen binding fragment thereof of claim 1 or 2 , wherein said antibody or said antigen binding fragment is linked or conjugated to a detectable agent.

The purified antibody or antigen binding fragment thereof of claim 1 or 2 , wherein said antibody or antigen binding fragment thereof specifically binds to a stomach adenocarcinoma, colorectal adenocarcinoma, squamous cell lung carcinoma, lung adenocarcinoma, squamous cell carcinoma of the esophagus, adenocarcinoma of the pancreas, urothel carcinoma of the urinary bladder, renal cell carcinoma of the kidney, adenocarcinoma of the prostate, ductal carcinoma of the breast, lobular carcinoma of the breast, adenocarcinoma of the ovary, adenocarcinoma of the endometrium, or adenocarcinoma of the uterus cell.

15. The purified antibody or antigen binding fragment thereof of claim 1 or 2 , wherein said antibody or antigen binding fragment thereof specifically binds to and inhibits proliferation of adenocarcinoma cells of the pancreas in vitro.

16. The purified antibody or antigen binding fragment thereof of claim 1 or 2 , wherein said antibody or antigen binding fragment thereof inhibits proliferation of BXPC-3 (ATCC Accession No. CRL-1687) cells.

17. The purified antibody or antigen binding fragment thereof of claim 1 or 2 , wherein said antibody or antigen binding fragment thereof induces apoptosis of BXPC-3 (ATCC Accession No. CRL-1687) cells.

The purified antibody or antigen binding fragment thereof of claim 1 or 2 , wherein said antibody or antigen binding fragment thereof specifically binds to CACO-2 cell line (ATCC Accession No. HBT-37), COLO-320 cell line (DSMZ Accession No. ACC 144), or COLO-206F cell line (DSMZ Accession No. ACC 21).

19. A purified antibody or antigen binding fragment thereof, comprising the amino acid sequence of SEQ ID NO:5 and SEQ ID NO:7.

20. An antibody produced by the PM-2 cell line having DSMZ Accession No. DSM ACC2600.

21. A purified or isolated cell that expresses the antibody or antigen binding fragment of claim 1 or 2 .

22. The purified or isolated cell of claim 21 , wherein said cell is a hybridoma.
