Abstract

Sequences of a serotype 8 adeno-associated virus and vectors and host cells containing these sequences are provided. Also described are methods of using such host cells and vectors in production of rAAV particles. rAAV8-mediated delivery of therapeutic and immunogenic genes, including Factor VIII, is also provided.

Claims (17)

1. An adeno-associated virus (AAV) 8 vector comprising a capsid comprising an AAV8 vp3 protein having the sequence of amino acid 203 to 737 of SEQ ID NO: 2 or a sequence at least 95% identical thereto, said vector further comprising a minigene comprising AAV inverted terminal repeats and a heterologous nucleic acid sequence operably linked to regulatory sequences which direct expression of a product from the heterologous nucleic acid sequence in a host cell.

2. The AAV8 vector according to claim 1 , wherein the capsid comprises an AAV8 vp3 protein having the sequence of amino acids 203 to 737 of SEQ ID NO: 2.

3. The AAV8 vector according to claim 1 , wherein the capsid comprises an AAV8 vp3 protein having the sequence at least 99% identical to amino acids 203 to 737 of SEQ ID NO: 2.

4. The AAV8 vector according to claim 1 , wherein the capsid further comprises an AAV8 vp2 protein having the sequence of amino acids 138 to 737 of SEQ ID NO: 2 or a sequence at least 99% identical thereto.

5. The AAV8 vector according to claim 1 , wherein the expression product of the heterologous nucleic acid sequence is an HIV protein.

6. The AAV8 vector according to claim 1 , wherein the expression product of the heterologous nucleic acid sequence is a Factor VIII protein.

7. The AAV8 vector according to claim 6 , wherein the Factor VIII protein is selected from the group consisting of the light chain of Factor VIII; the heavy chain of Factor VIII; and the B-domain mutant of Factor VIII.

8. The adeno-associated virus (AAV) vector according to claim 1 , wherein the AAV8 vp3 capsid protein is fused to a heterologous AAV capsid protein or fragment thereof selected from the group consisting of:

a fragment of an AAV8 encompassing hypervariable region (HVR)1 through 12 selected from the group consisting of: aa 146 to 152; aa 182 to 186; aa 262 to 264; aa 381 to 383; aa 450 to 474; aa 490 to 495; aa500 to 504; aa 514 to 522; aa 534 to 555; aa 581 to 594; aa 658 to 667; aa 705 to 719, wherein the amino acid numbers are relative to the sequence of the AAV2 capsid, SEQ ID NO:4;

a fragment of an AAV8 selected from the group consisting of: aa 24 to 42, aa 25 to 28; aa 81 to 85; aa 133 to 165; aa 134 to 165; aa 137 to 143; aa 154 to 156; aa 194 to 208; aa 261 to 274; aa 262 to 274; aa 171 to 173; aa 185 to 198; aa 413 to 417; aa 449 to 478; aa 494 to 525; aa 534 to 571; aa 581 to 601; aa 660 to 671; aa 709 to 723, wherein the amino acid numbers are relative to the sequence of the AAV2 capsid, SEQ ID NO:4; and

a fragment of an AAV8 selected from the group consisting of: aa 1 to 184, aa 199 to 259; aa 274 to 446; aa 603 to 659; aa 670 to 706; aa 724 to 738; aa 185 to 198; aa 260 to 273; aa 447 to 477; aa 495 to 602; aa 603 to 659; aa 660 to 669; and aa 707 to 723, wherein the amino acid numbers are relative to the sequence of the AAV8 capsid, SEQ ID NO:2.

9. The AAV8 vector according to claim 1 , wherein the expression product of the heterologous nucleic acid sequence is an IgG, IgM, IgA, IgD, IgE, chimeric immunoglobulin, humanized antibody, or a single chain antibody.

10. The AAV8 vector according to claim 9 , wherein the expression product of the heterologous nucleic acid sequence is a chimeric immunoglobulin.

11. The AAV8 vector according to claim 9 , wherein the expression product of the heterologous nucleic acid sequence is a single chain antibody.

12. A pharmaceutical composition comprising the AAV8 vector according to claim 1 and a physiological compatible carrier therefor.

13. A cell culture comprising the AAV8 vector according to claim 1 .

14. A method of delivering a transgene to a cell, said method comprising the step of contacting the cell with the AAV vector according to claim 1 , wherein said AAV vector comprises the transgene.
