Methods of identifying anti-cancer agents and uses thereof
View Patent ↗The present invention provides methods of identifying and/or detecting anti-cancer agents. The present invention provides methods of identifying and/or detecting compounds that can activate PARP and/or induce necrosis. The present invention also provides for methods of treating cancer in an individual. The present invention also provides kits for identifying and/or detecting anti-cancer agents.
1. A kit for identifying a PARP activator comprising:
1) a container comprising a plurality of immortalized cells that are apoptosis resistant, wherein said cells are deficient in Bax gene expression and Bak gene expression; and
2) an instruction manual; and
3) at least one reagent to detect PARP activation.
2. The kit of claim 1 , wherein said reagent to detect PARP activation is a reagent used in measuring PAR polymers, NAD depletion, or ATP depletion.
3. The kit of claim 1 , wherein the kit further comprises a positive control, wherein the positive control induces PARP activation or induces necrosis of a plurality of immortalized cells upon contact with said plurality of immortalized cells.
4. A kit for identifying a PARP activator comprising:
1) a container comprising a plurality of immortalized cells that are apoptosis resistant, wherein said cells are deficient in Bax gene expression and Bak gene expression; and
2) an instruction manual; and
3) a positive control, wherein the positive control induces PARP activation or induces necrosis of a plurality of immortalized cells upon contact with said plurality of immortalized cells.
5. The kit of claim 4 , wherein the positive control is N-methyl-N′-nitro N-nitrosoguandidine (MNNG).
6. The kit of claim 4 further comprising at least one reagent to detect PARD activation.
7. The kit of claim 6 , wherein said reagent to detect PARP activation is a reagent used in measuring PAR polymers, NAD depletion, or ATP depletion.
8. The kit of claim 4 further comprising at least one reagent to detect necrosis.
9. The kit of claim 8 , wherein said reagent to detect necrosis is a reagent used in measuring organelle swelling, intracellular vacuole formation, plasma membrane disintegration, or nuclear degradation without condensation.
