Delivery system for heparin-binding growth factors
A device comprising a scaffold coated with a protein having at least 83% identity to the amino acid sequence of the core protein of domain I of a mammalian perlecan, wherein at least one glycosaminoglycan is attached to the protein is described. The device may be used in vitro or in vivo to induce replication and/or differentiation of connective tissue cells and to repair tissue.
1. An implantable device comprising a scaffold, the scaffold coated with a proteoglycan that consists of an amino acid sequence having at least 84% homology to the amino acid sequence of the core protein of domain I of a mammalian perlecan, wherein the proteoglycan has at least one heparan sulfate glycosaminoglycan chain attached, and wherein the proteoglycan stimulates mesenchymal stem cell differentiation to cartilage.
2. The device of claim 1 , further comprising at least one growth factor attached to the glycosaminoglycan chain.
3. The device of claim 2 , wherein the growth factor is a heparin-binding growth factor.
4. The device of claim 1 , wherein the device is a medical device.
5. The device of claim 1 , wherein the scaffold comprises a polymer, biologically derived material, ceramic, metal, or combinations thereof.
6. The device of claim 1 , wherein the device is implanted in a mammal.
7. The device of claim 2 , wherein the growth factor induces differentiation of connective tissue cells.
8. The device of claim 7 , wherein the connective tissue cells are chondrocytes or fibroblasts.
9. The device of claim 1 , wherein the proteoglycan has two or three glycosaminoglycan chains attached.
10. A device for producing connective tissue cells for the repair of tissues or organs comprising a scaffold coated with a proteoglycan that consists of an amino acid sequence having at least 84% homology to the amino acid sequence of the core protein of domain I of a mammalian perlecan, wherein the proteoglycan has at least one heparan sulfate glycosaminoglycan chain attached, wherein the proteoglycan stimulates mesenchymal stem cell differentiation to cartilage, and wherein at least one growth factor capable of inducing connective tissue cells to replicate or to differentiate is attached to the glycosaminoglycan chain.
11. The device of claim 10 , wherein the growth factor is a heparin-binding growth factor.
12. The device of claim 10 , wherein the scaffold comprises a polymer, biologically derived material, ceramic, metal, or combinations thereof.
13. The device of claim 10 , wherein the cells are connective tissue cells.
14. The device of claim 13 , wherein the connective tissue cells are chondrocytes or fibroblasts.
15. The device of claim 10 , wherein the proteoglycan has two or three glycosaminoglycan chains attached to the protein.
16. The device of claim 1 , wherein the protein has at least 84% homology to the amino acid sequence of SEQ ID NO: 1.
17. The device of claim 1 , wherein the protein consists of the amino acid sequence of SEQ ID NO: 1.
18. The device of claim 1 , wherein the protein has at least 84% homology to the amino acid sequence of SEQ ID NO: 2.
19. The device of claim 1 , wherein the protein consists of the amino acid sequence of SEQ ID NO: 2.
