Abstract

The present invention is directed to methods for predicting or diagnosing a hazelnut-induced systemic reaction, and for methods for treating such a reaction.

Claims (16)

1. A method of diagnosing a hazelnut allergy characterized by a hazelnut-induced systemic reaction comprising: contacting an IgE-containing biological specimen of a human with an isolated and purified polypeptide that forms a complex with serum IgE from a human with said allergy, wherein said polypeptide is encoded by a first nucleic acid having the sequence of nucleotides 25-924 of SEQ ID NO:1 or a second nucleic acid that hybridizes over the full length of the complement of said first nucleic acid under high stringency conditions comprising 68° C. in aqueous buffered solution and wherein said second nucleic acid encodes a polypeptide that is recognized by antibodies specific for the polypeptide encoded by said first nucleic acid; determining the presence or absence of a complex between IgE in the specimen and the polypeptide; and diagnosing hazelnut allergy characterized by a hazelnut-induced systemic reaction if the complex is present.

A method of diagnosing a hazelnut allergy characterized by a hazelnut-induced systemic reaction comprising: contacting an IgE-containing biological specimen of a human with an isolated and purified polypeptide that comprises the amino acid sequence of SEQ ID NO:2; determining the presence or absence of a complex between IgE in the specimen and the polypeptide; and diagnosing hazelnut allergy characterized by a hazelnut-induced systemic reaction if the complex is present.

3. The method of claim 1 wherein the biological specimen is whole blood, sputum, serum, plasma, cerebrospinal fluid or urine.

4. The method of claim 1 wherein the presence of the complex is determined by an immunoassay.

5. The method of claim 4 wherein the polypeptide is immobilized on a solid support.

A method of diagnosing a hazelnut allergy characterized by a hazelnut-induced systemic reaction comprising: contacting an IgE-containing biological specimen of a human with an isolated and purified polypeptide that comprises amino acids 1-320 of SEQ ID NO:2; determining the presence or absence of a complex between IgE in the specimen and the polypeptide; and diagnosing hazelnut allergy characterized by a hazelnut-induced systemic reaction if the complex is present.

7. The method of claim 6 wherein the biological specimen is whole blood, sputum, serum, plasma, cerebrospinal fluid or urine.

8. The method of claim 6 wherein the presence of the complex is determined by an immunoassay.

9. The method of claim 6 wherein the polypeptide is immobilized on a solid support.

10. The method of claim 2 wherein the biological specimen is whole blood, sputum, serum, plasma, cerebrospinal fluid or urine.

11. The method of claim 2 wherein the presence of the complex is determined by an immunoassay.

12. The method of claim 2 wherein the polypeptide is immobilized on a solid support.

13. A method of diagnosing a hazelnut allergy characterized by a hazelnut-induced systemic reaction comprising: recombinantly producing a polypeptide that forms a complex with serum IgE from a human with said allergy, wherein said polypeptide is encoded by a first nucleic acid having the sequence of nucleotides 25-924 of SEQ ID NO:1 or a second nucleic acid that hybridizes with over the full length of the complement of said first nucleic acid under high stringency conditions comprising 68° C. in aqueous buffered solution and wherein said second nucleic acid encodes a polypeptide that is recognized by antibodies specific for the polypeptide encoded by said first nucleic acid; contacting an IgE-containing biological specimen of a human with said polypeptide; determining the presence or absence of a complex between IgE in the specimen and said polypeptide; and diagnosing hazelnut allergy characterized by a hazelnut-induced systemic reaction if the complex is present.

14. The method of claim 13 wherein the biological specimen is whole blood, sputum, serum, plasma, cerebrospinal fluid or urine.

15. The method of claim 13 wherein the presence of the complex is determined by an immunoassay.

16. The method of claim 13 wherein the polypeptide is immobilized on a solid support.
