Abstract

The present invention relates to nucleic acid sequences and amino acid sequences which influence bone deposition, the Wnt pathway, ocular development, tooth development, and may bind to LRP. The nucleic acid sequence and polypeptides include Wise and Sost as well as a family of molecules which express a cysteine knot polypeptide. Additionally, the present invention relates to various molecular tools derived from the nucleic acids and polypeptides including vectors, transfected host cells, monochronal antibodies, Fab fragments, and methods for impacting the pathways.

Claims (9)

1. A method for modulating Sost binding to an LRP comprising contacting a Sost polypeptide or a nucleic acid encoding a Sost polypeptide with an agent that binds to the Sost polypeptide or nucleic acid encoding the Sost polypeptide and prevents or decreases Sost polypeptide binding to an LRP selected from the group consisting of LRP 1-11 and Arrow.

2. The method according to claim 1 , wherein the agent is selected from the group consisting of a monoclonal antibody, a Fv fragment of an antibody, a Fab fragment of an antibody, a Fab 2 fragment of an antibody, and an antibody combining region.

3. The method according to claim 2 , wherein the agent is a monoclonal antibody, or a Fab fragment thereof.

4. The method according to claim 3 , wherein the monoclonal antibody, or a Fab fragment thereof specifically binds to a Sost polypeptide that is encoded by a nucleic acid sequence of SEQ ID NO: 10.

5. The method according to claim 3 , wherein the monoclonal antibody, or a Fab fragment thereof prevents or decreases Sost binding to an LRP selected from the group consisting of SEQ ID Nos: 81-82.

6. The method according to claim 1 , wherein the LRP is selected from the group consisting of LRP 1, 2, 5, and 6.

7. The method according to claim 1 , wherein the LRP is selected from the group consisting of LRP 5 and 6.

8. The method according to claim 1 , wherein the LRP is LRP5.

9. The method according to claim 1 , wherein the LRP is LRP6.
