Methods and compositions for treating cancer using proteasome inhibitors
The present invention relates to therapeutic compositions for treating cancer or preventing the growth of cancer cells, e.g., tumor growth, in a subject. The present invention also relates to methods for treating cancer, e.g., inhibiting tumor growth, in a subject who has become resistant to treatment, by administering to a subject an effective amount of a proteasome inhibitor and an effective amount of a therapeutic agent, e.g., a chemotherapeutic agent. The present invention further relates to methods for purging bone marrow, i.e., removing cancer cells from bone marrow, by exposing the bone marrow cells to a proteasome inhibitor and a therapeutic agent, e.g., a chemotherapeutic agent.
1. A method for treating multiple myeloma in a subject wherein the subject's cancer cells are resistant to treatment with the proteasome inhibitor PS-341, comprising administering to the subject an effective amount of PS-341 and an effective amount of doxorubicin such that multiple myeloma is treated.
2. The method of claim 1 , wherein the treatment of multiple myeloma is due to the inhibition of tumor growth.
3. The method of claim 1 , further comprising the administration of at least one additional proteasome inhibitor or chemotherapeutic agent.
4. The method of claim 1 , wherein PS-341 is administered prior to the administration of doxorubicin.
5. The method of claim 1 , wherein PS-341 is administered simultaneously with the administration of doxorubicin.
6. The method of claim 1 , wherein PS-341 is administered after the administration of doxorubicin.
7. The method of claim 1 , wherein PS-341 and doxorubicin are administered intravenously.
8. The method of claim 1 , wherein PS-341 and doxorubicin are administered intraperitoneally.
9. The method of claim 1 , wherein PS-341 and doxorubicin are administered orally.
10. A method for treating multiple myeloma in a subject, wherein the subject's cancer cells are resistant to treatment with the proteasome inhibitor PS-341, comprising administering to the subject an effective amount of PS-341 and an effective amount of doxorubicin such that multiple myeloma is treated, wherein PS-341 is administered at a dose of about 0.001 mg/m 2 body surface area/day to about 4.0 mg/m 2 body surface area/day.
11. The method of claim 10 , wherein the treatment of multiple myeloma is due to the inhibition of tumor growth.
12. The method of claim 10 , further comprising the administration of at least one additional proteasome inhibitor or chemotherapeutic agent.
13. The method of claim 10 , wherein PS-341 is administered prior to the administration of doxorubicin.
14. The method of claim 10 , wherein PS-341 is administered simultaneously with the administration of doxorubicin.
15. The method of claim 10 , wherein PS-341 is administered after the administration of doxorubicin.
16. The method of claim 10 , wherein PS-341 and doxorubicin are administered intravenously.
17. The method of claim 10 , wherein PS-341 and doxorubicin are administered intraperitoneally.
18. The method of claim 10 , wherein PS-341 and doxorubicin are administered orally.
