Abstract

Provided are cyclobutyl nucleosides and methods for their use in treatment of infections including Retroviridae (including HIV), Hepadnaviridae (including HBV), or Flaviviridae (including BVDV and HCV) infection, or conditions related to abnormal cellular proliferation, in a host, including animals, and especially humans.

Claims (41)

1. A nucleoside:

or a salt, acyl ester, salt of an acyl ester, enantiomer, diastereomer, or tautomer thereof.

2. A nucleoside selected from the group consisting of

wherein, Base is a purine or pyrimidine base or a pharmaceutically acceptable salt, acyl ester, salt of an acyl ester, enantiomer, diastereomer, or tautomer thereof.

3. The nucleoside of claim 2 , wherein the base is a pyrimidine.

4. The nucleoside of claim 3 , wherein the pyrimidine is a 5-fluorocytidine.

5. The nucleoside of claim 2 , wherein the base is a purine.

6. The nucleoside of claim 5 , wherein the purine is guanine or adenine.

7. A nucleoside selected from the group consisting of:

wherein, Base is a purine or pyrimidine base or a pharmaceutically acceptable salt, acyl ester, salt of an acyl ester, enantiomer, diastereomer, or tautomer thereof.

8. The nucleoside of claim 7 , wherein the base is a pyrimidine.

9. The nucleoside of claim 8 , wherein the pyrimidine is a 5-fluorocytidine.

10. The nucleoside of claim 7 , wherein the base is purine.

11. The nucleoside of claim 10 , wherein the purine is guanine or adenine.

12. A nucleoside selected from the group consisting of:

wherein, Base is a purine or pyrimidine base or a pharmaceutically acceptable salt, acyl ester, salt of an acyl ester, enantiomer, diastereomer, or tautomer thereof.

13. The nucleoside of claim 12 , wherein the pyrimidine.

14. The nucleoside of claim 13 , wherein the pyrimidine is a 5-fluorocytidine.

15. The nucleoside of claim 12 , wherein the base is a purine.

16. The nucleoside of claim 15 , wherein the purine is guanine or adenine.

17. A nucleoside selected from the group consisting of:

wherein, Base is a purine or pyrimidine base and Z is hydrogen or a pharmaceutically acceptable salt, acyl ester, salt of an acyl ester, enantiomer, diastereomer, or tautomer thereof.

18. The nucleoside of claim 17 , wherein the base is a pyrimidine.

19. The nucleoside of claim 18 , wherein the pyrimidine is a 5-fluorocytidine.

20. The nucleoside of claim 17 , wherein the base is a purine.

21. The nucleoside of claim 20 , wherein the purine is guanine or adenine.

22. A pharmaceutical composition comprising an effective amount of the nucleoside of claim 1 , or a pharmaceutically acceptable salt thereof together with a pharmaceutically acceptable carrier or diluent.

23. A pharmaceutical composition comprising an effective amount of the nucleoside of claim 1 , or a pharmaceutically acceptable salt thereof together with a pharmaceutically acceptable carrier or diluent and in combination with one or more other antivirally effective agents.

24. A pharmaceutical composition comprising an effective amount of the nucleoside of claim 2 , or a pharmaceutically acceptable salt thereof together with a pharmaceutically acceptable carrier or diluent.

25. A pharmaceutical composition comprising an effective amount of the nucleoside of claim 2 , or a pharmaceutically acceptable salt thereof together with a pharmaceutically acceptable carrier or diluent and in combination with one or more other antivirally effective agents.

26. A pharmaceutical composition comprising an effective amount of the nucleoside of claim 7 , or a pharmaceutically acceptable salt thereof together with a pharmaceutically acceptable carrier or diluent.

27. A pharmaceutical composition comprising an effective amount of the nucleoside of claim 7 , or a pharmaceutically acceptable salt thereof together with a pharmaceutically acceptable carrier or diluent and in combination with one or more other antivirally effective agents.

28. A pharmaceutical composition comprising an effective amount of the nucleoside of claim 12 , or a pharmaceutically acceptable salt thereof together with a pharmaceutically acceptable carrier or diluent.

29. A pharmaceutical composition comprising an effective amount of the nucleoside of claim 12 , or a pharmaceutically acceptable salt thereof together with a pharmaceutically acceptable carrier or diluent and in combination with one or more other antivirally effective agents.

30. A pharmaceutical composition comprising an effective amount of the nucleoside of claim 17 , or a pharmaceutically acceptable salt thereof together with a pharmaceutically acceptable carrier or diluent.

31. A pharmaceutical composition comprising an effective amount of the nucleoside of claim 17 , or a pharmaceutically acceptable salt thereof together with a pharmaceutically acceptable carrier or diluent and in combination with one or more other antivirally effective agents.

32. A nucleoside:

wherein, Base is a pyrimidine base and Z is hydrogen or a pharmaceutically acceptable salt, acyl ester, salt of an acyl ester, enantiomer, diastereomer, or tautomer thereof.

33. The nucleoside of claim 32 , Wherein the pyrimidine is a 5-fluorocytidine.

34. A pharmaceutical composition comprising an effective amount of the nucleoside of claim 32 , or a pharmaceutically acceptable salt thereof together with a pharmaceutically acceptable carrier or diluent.

35. A pharmaceutical composition comprising an effective amount of the nucleoside of claim 32 , or a pharmaceutically acceptable salt thereof together with a pharmaceutically acceptable carrier or diluent and in combination with one or more other antivirally effective agents.
