Abstract

The present invention relates to zcytor17lig polynucleotide, polypeptide and anti-zcytor17 antibody molecules. The zcytor17lig is a novel cytokine. The polypeptides may be used within methods for stimulating the immune system, and proliferation and/or development of hematopoietic cells in vitro and in vivo. The present invention also includes methods for producing the protein, uses therefor and antibodies thereto.

Claims (37)

1. An isolated polynucleotide, wherein the polynucleotide encodes a polypeptide comprising the sequence of amino acid residues having at least 96% identity to: (a) amino acid residues 31 to 163 as shown in SEQ ID NO:11; or (b) amino acid residues 1 to 163 as shown in SEQ ID NO: 11.

2. The isolated polynucleotide of claim 1 , wherein amino acid residues 74, 137, and 151 of the polypeptide are cysteine.

3. An isolated polynucleotide, wherein the polynucleotide encodes a polypeptide comprising the sequence of amino acid residues selected from the group consisting of: (a) amino acid residues 31 to 163 as shown in SEQ ID NO: 11; and (b) amino acid residues 1 to 163 as shown in SEQ ID NO: 11.

4. The isolated polynucleotide of claim 3 , wherein the sequence of amino acid residues of the polypeptide consists of amino acid residues 31 to 163 as shown in SEQ ID NO: 11.

5. The isolated polynucleotide of claim 3 , wherein the sequence of amino acid residues of the polypeptide consists of amino acid residues 1 to 163 as shown in SEQ ID NO; 11.

6. The isolated polynucleotide of claim 1 , wherein the sequence of amino acid residues of the polypeptide has at least 97% sequence identity to:

(a) amino acid residues 31 to 163 as shown in SEQ ID NO:11; or

(b) amino acid residues 1 to 163 as shown in SEQ ID NO:11.

7. The isolated polynucleotide of claim 1 , wherein the sequence of amino acid residues of the polypeptide has at least 98% sequence identity to:

(a) amino acid residues 31 to 163 as shown in SEQ ID NO:11; or

(b) amino acid residues 1 to 163 as shown in SEQ ID NO:11.

8. The isolated polynucleotide of claim 1 , wherein the sequence of amino acid residues of the polypeptide has at least 99% sequence identity to:

(a) amino acid residues 31 to 163 as shown in SEQ ID NO:11; or

(b) amino acid residues 1 to 163 as shown in SEQ ID NO:11.

9. The isolated polynucleotide of claim 1 , wherein the polypeptide comprises an affinity tag.

10. The isolated polynucleotide of claim 1 , wherein the polypeptide comprises PEGylation.

11. An isolated polynucleotide comprising the sequence of nucleotides from nucleotide 91 to nucleotide 489 of SEQ ID NO: 10.

12. An isolated expression vector comprising the following operably linked elements:

(a) a transcription promoter;

(b) a DNA segment comprising a polynucleotide selected from the group consisting of:

(i) the polynucleotide of claim 1 ;

(ii) the polynucleotide of claim 7 ; and

(iii) the polynucleotide of claim 11 , and

(c) a transcription terminator.

13. A cultured cell comprising the expression vector according claim 12 .

14. A method of producing a comprising:

culturing a cell according to claim 13 under conditions wherein the polypeptide encoded by the DNA segment is expressed; and

recovering the polypeptide.

15. An isolated polynucleotide, wherein the polynucleotide encodes a polypeptide comprising the amino acid sequence selected from the group consisting of:

(a) a polypeptide comprising the amino acid sequence of SEQ ID NO:11 from amino acid number 38-157; and

(b) a polypeptide comprising the amino acid sequence of SEQ ID NO:11 from amino acid number 24-163.

16. An isolated polynucleotide encoding an antigenic polypeptide comprising a sequence of at least 30 contiguous amino acid residues of SEQ ID NO: 11, wherein the antigenic polypeptide comprises the amino acid sequence selected from:

(a) amino acid residues 34 to 39 of SEQ ID NO: 11;

(b) amino acid residues 46 to 51 of SEQ ID NO: 11;

(c) amino acid residues 131 to 136 of SEQ ID NO:11;

(d) amino acid residues 158 to 163 of SEQ ID NO: 11; and

(e) amino acid residues 157 to 162 of SEQ ID NO:11.
