Methods of using molecular conjugates comprising monoclonal antibodies to dendritic cells
The present invention provides novel methods for targeting an antigen to a dendritic cell using an antibody linked to the antigen. The present invention further provides methods of inducing or enhancing an immune response and methods of immunization using an antibody linked to the antigen.
1. A method of targeting an antigen to a dendritic cell comprising contacting the cell with an antibody, or antigen-binding portion thereof, that binds to the human macrophage mannose receptor, wherein the antibody, or antigen-binding portion thereof, (a) is linked to an antigen and (b) comprises heavy and light chain variable regions as shown in SEQ ID NO:2 and SEQ ID NO:4, respectively.

A method of inducing or enhancing an immune response against an antigen in a subject comprising administering to the subject an antibody, or an antigen-binding portion thereof, that binds to the human macrophage mannose receptor, wherein the antibody, or an antigen-binding portion thereof, (a) is linked to an antigen and (b) comprises heavy and light chain variable regions as shown in SEQ ID NO:2 and SEQ ID NO:4, respectively.

A method of immunizing a subject comprising administering to the subject an antibody, or antigen-binding portion thereof, that binds to the human macrophage mannose receptor, wherein the antibody, or antigen-binding portion thereof, (a) is linked to an antigen and (b) comprises heavy and light chain variable regions as shown in SEQ ID NO:2 and SEQ ID NO:4, respectively.
4. The method of claim 2 , wherein the immune response comprises presentation of the antigen as a component of an MHC-I or MHC-II conjugate.
5. The method of claim 3 , wherein the antibody is administered in an amount sufficient to induce cytokine release by dendritic cells.
6. The method of any one of claims 1 - 3 , wherein the antigen is a tumor antigen, an autoantigen, or a component of a pathogen.
7. The method of any one of claims 1 - 3 , wherein the antigen is selected from the group of antigens consisting of Pmel-17, HER2/neu, CEA, gp100, MART1, TRP-2, melan-A, NY-ESO-1, MN (gp250), idiotype, MAGE-1, MAGE-3, Tyrosinase, Telomerase, and MUC-1.
8. The method of any one of claims 1 - 3 , wherein the antibody, or an antigen-binding portion thereof, is linked to the antigen to form a molecular conjugate which is produced as a recombinant fusion protein or a chemical conjugate or is encoded by the nucleotide sequence shown in SEQ ID NO:8.
