Antibodies to human IL-22
View Patent ↗The present application provides human antibodies and antigen binding fragments thereof that specifically bind to the human interleukin-22 (IL-22). The antibodies can act as antagonists of IL-22 activity, thereby modulating immune responses in general, and those mediated by IL-22 in particular. The disclosed compositions and methods may be used for example, in diagnosing, treating or preventing inflammatory disorders, autoimmune diseases, allergies, septic shock, infectious disorders, transplant rejection, cancer, and other immune system disorders.
1. An isolated antibody, or antigen-binding fragment thereof, that specifically binds to IL-22, wherein the antibody, or antigen-binding fragment thereof, comprises a V H domain comprising an amino acid sequence which is at least 95% identical to the amino acid sequence shown in SEQ ID NO:599 or a V L domain comprising an amino acid sequence which is at least 95% identical to the amino acid sequence shown in SEQ ID NO:600.
2. The antibody of claim 1 , wherein the V L domain comprises three complementarity determining regions (CDRs) comprising the amino acid sequences of SEQ ID NO:605, 606, and 607.
3. The antibody of claim 1 , wherein the V H domain comprises three complementarity determining regions (CDRs) comprising the amino acid sequences of SEQ ID NO: 602, 603, and 604.
4. The antibody of claim 1 , wherein the association constant of the antibody for human IL-22 is at least 10 10 M −1 .
5. The antibody of claim 1 , wherein the antibody blocks IL-22 mediated proliferation of BaF3 cells with an IC 50 of 150 pM or less and wherein the BaF3 cells comprise a human IL-22 receptor.
6. The antibody of claim 1 , wherein the antibody blocks IL-22 mediated GROa secretion from HT29 cells with an IC 50 of 150 pM or less.
7. The antibody of claim 1 , wherein the antibody inhibits the binding of IL-22 to IL-22R or a receptor complex comprising IL-22R and IL-10R2.
8. The antibody of claim 1 , wherein the antibody is human.
9. The antibody of claim 1 , wherein the antibody is IgG1 or IgG4.
10. A pharmaceutical composition comprising the antibody of claim 1 .
11. An isolated antibody, or antigen-binding fragment thereof, that specifically binds to IL-22, wherein the antibody, or antigen-binding fragment thereof, comprises 1) a V H domain comprising an amino acid sequence that is at least 85% identical to the amino acid sequence of SEQ ID NO:599 and 2) a V L domain comprising an amino acid sequence that is at least 85% identical to the amino acid sequence of SEQ ID NO:600.
12. The antibody of claim 11 , wherein the V H domain comprises an amino acid sequence that is at least 90% identical to the amino acid sequence of SEQ ID NO:599 and the V L domain comprises an amino acid sequence that is at least 90% identical to the amino acid sequence of SEQ ID NO:600.
13. The antibody of claim 11 , wherein the V H domain comprises an amino acid sequence that is at least 95% identical to the amino acid sequence of SEQ ID NO:599 and the V L domain comprises an amino acid sequence that is at least 95% identical to the amino acid sequence of SEQ ID NO:600.
14. The antibody of claim 11 , wherein the V H domain comprises an amino acid sequence that is at least 98% identical to the amino acid sequence of SEQ ID NO:599 and the V L domain comprises an amino acid sequence that is at least 98% identical to the amino acid sequence of SEQ ID NO:600.
15. The antibody of claim 11 , wherein the V H domain differs by no more than 5 amino acids from the amino acid sequence of SEQ ID NO:599 and wherein the V L domain differs by no more than 5 amino acids from the amino acid sequence of SEQ ID NO:600.
16. An isolated antibody, or antigen-binding fragment thereof, that specifically binds to IL-22, wherein the antibody, or antigen-binding fragment thereof, comprises 1) a V H domain comprising an amino acid sequence that is at least 85% identical to SEQ ID NO:599, wherein the V H domain comprises three complementarity determining regions (CDRs) comprising the amino acid sequences of SEQ ID NO:602, 603, and 604, and 2) a V L domain comprising an amino acid sequence that is at least 85% identical to SEQ ID NO:600, wherein the V L domain comprises three CDRs comprising the amino acid sequences of SEQ ID NO:605, 606, and 607.
17. The antibody of claim 16 , wherein the V H domain comprises an amino acid sequence that is at least 90% identical to SEQ ID NO:599 and the V L domain comprises an amino acid sequence that is at least 90% identical to SEQ ID NO:600.
18. The antibody of claim 16 , wherein the V H domain comprises an amino acid sequence that is at least 95% identical to SEQ ID NO:599 and the V L domain comprises an amino acid sequence that is at least 95% identical to SEQ ID NO:600.
19. The antibody of claim 16 , wherein the V H domain comprises an amino acid sequence that is at least 98% identical to SEQ ID NO:599 and the V L domain comprises an amino acid sequence that is at least 98% identical to SEQ ID NO:600.
20. The antibody of claim 16 , wherein the V H domain differs by no more than 5 amino acids from the amino acid sequence of SEQ ID NO:599 and wherein the V L domain differs by no more than 5 amino acids from the amino acid sequence of SEQ ID NO:600.
