Method for preparing largazole analogs and uses thereof
View Patent ↗Analogs of largazole are described herein. Methods of treating cancer and blood disorders using largazole and largazole analogs and pharmaceutical compositions comprising the same are additionally described herein. Methods for preparing largazole analogs are likewise described.
1. A compound of Formula (XIX)
wherein X═O or NR 6 , wherein R 6 ═H, lower alkyl, or lower arylalkyl;
G=S, O, or NR 9 , wherein R 9 ═H, lower alkyl, or lower arylalkyl;
Q, Y, W, Z are, independently, N or CH, wherein at least one of Q, Y, W, and Z is CH;
R 1 ═H, C(O)(CH 2 ) 6 CH 3 , C(O)R 7 (wherein R 7 =lower alkyl, lower aryl, or lower arylalkyl), R 2 is H, lower alkyl, or lower arylalkyl; R 3 and R 4 is lower alkyl; R 5 is H; or a pharmaceutically acceptable salt, prodrug, or stereoisomer thereof.
2. The compound of claim 1 , having the Formula (XX)
or a pharmaceutically acceptable salt, prodrug, or stereoisomer thereof.
3. A pharmaceutical composition comprising a therapeutically effective amount of the compound of claim 1 and at least one pharmaceutically acceptable excipient for treating a blood disorder in a subject.
4. The pharmaceutical composition of claim 3 , wherein the blood disorder is at least one of a hemoglobinopathy or a thalassemia.
5. The pharmaceutical composition of claim 3 or 4 , wherein the subject is human.
