Abstract

The invention provides isolated polypeptide and nucleic acid sequences derived from Streptococcus pneumoniae that are useful in diagnosis and therapy of pathological conditions; antibodies against the polypeptides; and methods for the production of the polypeptides. The invention also provides methods for the detection, prevention and treatment of pathological conditions resulting from bacterial infection.

Claims (22)

1. A method of making a S. pneumoniae polypeptide, comprising the step of culturing a host cell that includes a recombinant expression vector, wherein the recombinant expression vector comprises a nucleic acid sequence that encodes a polypeptide having the amino acid sequence as set forth in SEQ ID NO: 5326.

2. The method of claim 1 , further including the step of isolating the polypeptide from the host cell.

3. The method of claim 1 , wherein the host cell is a prokaryotic host cell.

4. The method of claim 3 , wherein the prokaryotic host cell is selected from the group consisting of E. coli, B. subtilis and S. aureus.

5. The method of claim 4 , wherein the prokaryotic host cell is E. coli.

6. The method of claim 1 , wherein the host cell is a eukaryotic host cell.

7. The method of claim 6 , wherein the eukaryotic host cell is selected from the group consisting of an insect cell, a yeast cell and a mammalian cell.

8. The method of claim 7 , wherein the eukaryotic host cell is an insect cell.

9. The method of claim 7 , wherein the mammalian cell is a Chinese hamster ovary cell.

10. The method of claim 1 , wherein the polypeptide elicits an immune response.

11. The method of claim 10 , wherein the immune response is an immune response against S. pneumoniae infection.

12. A method of making a S. pneumoniae polypeptide, comprising the step of culturing a host cell that includes a recombinant expression vector, wherein the recombinant expression vector comprises a nucleic acid sequence that encodes a polypeptide having at least 80% sequence identity to the amino acid sequence as set forth in SEQ ID NO: 5326.

13. The method of claim 12 , further including the step of isolating the polypeptide from the host cell.

14. The method of claim 12 , wherein the nucleic acid sequence encodes a polypeptide having at least 90% identity to SEQ ID NO: 5326.

15. The method of claim 12 , wherein the nucleic acid sequence encodes a polypeptide having at least 95% identity to SEQ ID NO: 5326.

16. The method of claim 12 , wherein the nucleic acid sequence encodes a polypeptide having at least 98% identity to SEQ ID NO: 5326.

17. The method of claim 12 , wherein the nucleic acid sequence encodes a polypeptide having at least 99% identity to SEQ ID NO: 5326.

18. The method of claim 12 , wherein the polypeptide elicits an immune response.

19. The method of claim 12 , wherein the immune response is an immune response against S. pneumoniae infection.

20. The method of claim 8 , wherein the recombinant expression vector is a baculovirus expression vector.

21. The method of claim 1 , wherein the polypeptide is a surface protein.

22. The method of claim 12 , wherein the polypeptide is a surface protein.
