Chemical compounds
The present invention relates to compounds that are a non-nucleoside reverse transcriptase inhibitors, and to processes for the preparation and use of the same. Specifically, the present invention includes methods of using such compounds in the treatment of human immunodeficiency virus infection.
1. A compound of formula (I):
wherein m is 1, 2, 3 or 4;
n is 2;
each R 1 independently is halogen, —CN, C 1 -C 5 alkyl, C 2 -C 5 alkenyl, C 2 -C 6 alkynyl, C 3 -C 7 cycloalkyl, hydroxyl, C 1 -C 8 alkoxy, —C(O)OR 5 , —C(O)N(R 5 ) 2 , —OR 5 , —R 3 CN, or —N(R 5 ) 2 ;
each R 2 independently is halogen;
A is C 5 -C 12 aryl or C 5 -C 12 heterocycle;
Q is hydrogen or C 1 -C 4 alkyl;
Y is —C(O)—, —S(O) 2 —, or —S(O)—;
Z is C 4 -C 12 aryl, C 3 -C 14 heterocycle, R 3 Het, or R 3 Ar, each optionally substituted with one or more of C 1 -C 8 alkyl, C 3 -C 7 cycloalkyl, C 1 -C 8 alkoxy, halogen, oxo, hydroxyl, —CN, —NO 2 , —N(R 5 )C(O)R 5 , —N(R 5 ) 2 , —OR 5 , —C 3 -C 12 Het, —C(O)N(R 5 ) 2 , C 4 -C 12 Ar, —SR 5 , —S(O) 2 N(R 5 ) 2 , —OR 3 HetC(O)R 5 , —OCF 3 , —S(O) 2 R 5 , —OR 3 R 5 , —N(R 5 )C(O)R 3 OR 5 ; —N(R 5 )C(O)R 3 N(R 5 ) 2 , —N(R 5 )C(O)R 3 R 5 , —OR 3 N(R 5 ) 2 , —R 3 Het, —R 3 N(R 5 ) 2 , —R 3 N(R 5 )C(O)R 5 , —OR 3 SR 5 , —C(O)R 5 , —C(R 5 ) 3 , —R 3 C(O)OR 5 , —R 3 C(O)N(R 5 ) 2 , or —N(R 5 )S(O) 2 R 5 ; or Z is linked to Q to form a C 4 -C 14 heterocycle together with the nitrogen atom and Y group to which they are attached and are optionally substituted with —C(O)OR 5 or C 1 -C 6 alkoxy;
each R 5 independently is hydrogen, C 1 -C 8 alkyl, C 3 -C 7 cycloalkyl, oxo, C 3 -C 12 heterocycle, C 4 -C 12 aryl or C 1 -C 8 alkoxy, each optionally substituted with hydroxyl, C 1 -C 4 alkoxy or C 3 -C 7 cycloalkyl;
R 3 is C 2 -C 6 alkenylene or C 1 -C 4 alkylene, each optionally substituted with hydroxyl or C 1 -C 8 alkoxy;
Het is C 3 -C 12 heterocycle and is optionally substituted with one or more of C 1 -C 6 alkyl, —C(O)N(R 5 ) 2 , —R 3 S(O) 2 R 5 , or halogen; and
Ar is C 4 -C 12 aryl and is optionally substituted with one or more of C 1 -C 6 alkyl or halogen; or a pharmaceutically acceptable salt thereof.
2. A compound of formula (I) according to claim 1 wherein m is 2.

A compound of formula (I) according to claim 1 wherein Z is C 3 -C 14 heterocycle optionally substituted with one or more of C 1 -C 8 alkyl, C 3 -C 7 cycloalkyl, C 1 -C 8 alkoxy, halogen, oxo, hydroxyl, —CN, —NO 2 , —N(R 5 )C(O)R 5 , —N(R 5 ) 2 , —OR 5 , —C 3 -C 12 Het, —C(O)N(R 5 ) 2 , C 4 -C 12 Ar, —SR 5 , —S(O) 2 N(R 5 ) 2 , —OR 3 HetC(O)R 5 , —OCF 3 , —S(O) 2 R 5 , —OR 3 R 5 , —N(R 5 )C(O)R 3 OR 5 ; —N(R 5 )C(O)R 3 N(R 5 ) 2 , —N(R 5 )C(O)R 3 R 5 , —OR 3 N(R 5 ) 2 , —R 3 Het, —R 3 N(R 5 ) 2 , —R 3 N(R 5 )C(O)R 5 , —OR 3 SR 5 , —C(O)R 5 , —C(R 5 ) 3 , —R 3 C(O)OR 5 , —R 3 C(O)N(R 5 ) 2 , or —N(R 5 )S(O) 2 R 5 .

A compound of formula (I) according to claim 1 wherein Y is —C(O)—, Q is hydrogen, and Z is C 3 -C 14 heterocycle optionally substituted with one or more of C 1 -C 8 alkyl, C 3 -C 7 cycloalkyl, C 1 -C 8 alkoxy, halogen, oxo, hydroxyl, —CN, —NO 2 , —N(R 5 )C(O)R 5 , —N(R 5 ) 2 , —OR 5 , —C 3 -C 12 Het, —C(O)N(R 5 ) 2 , C 4 -C 12 Ar, —SR 5 , —S(O) 2 N(R 5 ) 2 , —OR 3 HetC(O)R 5 , —OCF 3 , —S(O) 2 R 5 , —OR 3 R 5 , —N(R 5 )C(O)R 3 OR 5 ; —N(R 5 )C(O)R 3 N(R 5 ) 2 , —N(R 5 )C(O)R 3 R 5 , —OR 3 N(R 5 ) 2 , —R 3 Het, —R 3 N(R 5 ) 2 , —R 3 N(R 5 )C(O)R 5 , —OR 3 SR 5 , —C(O)R 5 , —C(R 5 ) 3 , —R 3 C(O)OR 5 , —R 3 C(O)N(R 5 ) 2 , or —N(R 5 )S(O) 2 R 5 .

A compound of formula (I) according to claim 1 wherein Y is —C(O)—, Q is C 1 -C 8 alkyl, and Z is C 3 -C 14 heterocycle optionally substituted with one or more of C 1 -C 8 alkyl, C 3 -C 7 cycloalkyl, C 1 -C 8 alkoxy, halogen, oxo, hydroxyl, —CN, —NO 2 , —N(R 5 )C(O)R 5 , —N(R 5 ) 2 , —OR 5 , —C 3 -C 12 Het, —C(O)N(R 5 ) 2 , C 4 -C 12 Ar, —SR 5 , —S(O) 2 N(R 5 ) 2 , —OR 3 HetC(O)R 5 , —OCF 3 , —S(O) 2 R 5 , —OR 3 R 5 , —N(R 5 )C(O)R 3 OR 5 ; —N(R 5 )C(O)R 3 N(R 5 ) 2 , —N(R 5 )C(O)R 3 R 5 , —OR 3 N(R 5 ) 2 , —R 3 Het, —R 3 N(R 5 ) 2 , —R 3 N(R 5 )C(O)R 5 , —OR 3 SR 5 , —C(O)R 5 , —C(R 5 ) 3 , —R 3 C(O)OR 5 , —R 3 C(O)N(R 5 ) 2 , or —N(R 5 )S(O) 2 R 5 .
6. A compound of formula (I) according to claim 1 wherein m is 2 and one R 1 is halogen and one R 1 is —CN.

A compound of formula (I) according to claim 1 wherein m is 2, n is 2, and Z is C 3 -C 14 heterocycle optionally substituted with one or more of C 1 -C 8 alkyl, C 3 -C 7 cycloalkyl, C 1 -C 8 alkoxy, halogen, oxo, hydroxyl, —CN, —NO 2 , —N(R 5 )C(O)R 5 , —N(R 5 ) 2 , —OR 5 , —C 3 -C 12 Het, —C(O)N(R 5 ) 2 , C 4 -C 12 Ar, —SR 5 , S(O) 2 N(R 5 ) 2 , —OR 3 HetC(O)R 5 , —OCF 3 , —S(O) 2 R 5 , —OR 3 R 5 , —N(R 5 )C(O)R 3 OR 5 ; —N(R 5 )C(O)R 3 N(R 5 ) 2 , —N(R 5 )C(O)R 3 R 5 , —OR 3 N(R 5 ) 2 , —R 3 Het, —R 3 N(R 5 ) 2 , —R 3 N(R 5 )C(O)R 5 , —OR 3 SR 5 , —C(O)R 5 , —C(R 5 ) 3 , —R 3 C(O)OR 5 , —R 3 C(O)N(R 5 ) 2 , or —N(R 5 )S(O) 2 R 5 .
8. A compound of formula (I) according to claim 1 wherein Z is selected from the group consisting of

each R 8 is independently hydrogen, halogen, N(R 15 ) 2 , OR 15 , SR 15 , C(O)N(R 15 ) 2 , C(O)OR 15 , CF 3 , C 1 -C 8 alkyl, C 1 -C 8 alkoxy, or —CN, wherein each C 1 -C 8 alkyl or C 1 -C 8 alkoxy is optionally substituted with hydroxyl, C 1 -C 4 alkoxy or C 3 -C 7 cycloalkyl;
R 9 is hydrogen or halogen;
R 10 is hydrogen, hydroxyl, C 1 -C 8 alkoxy, —N(R 15 )C(O)R 15 , —N(R 15 )C(O)R 13 N(R 15 ) 2 , —N(R 15 ) 2 , or —R 13 N(R 15 )C(O)R 15 ;
R 13 is C 1 -C 6 alkylene;
each R 15 is independently hydrogen, C 1 -C 8 alkyl, C 3 -C 7 cycloalkyl, C 3 -C 12 heterocycle, C 4 -C 12 aryl or C 1 -C 8 alkoxy, each optionally substituted with hydroxyl, C 1 -C 4 alkoxy or C 3 -C 7 cycloalkyl;
R 11 is hydrogen or C 1 -C 6 alkoxy; and
each R 12 is independently hydrogen, halogen, C 1 -C 8 alkyl, C 1 -C 8 alkoxy, C 3 -C 6 cycloalkyl, N(R 15 ) 2 , OR 15 , SR 15 , C(O)N(R 15 ) 2 , C(O)OR 15 , or —N(O) 2 , wherein each C 1 -C 8 alkyl or C 1 -C 8 alkoxy is optionally substituted with hydroxyl, C 1 -C 4 alkoxy or C 3 -C 7 cycloalkyl.
9. A compound of formula (I) according to claim 1 wherein Y is —C(O)— and Z is selected from the group consisting of

11. A pharmaceutically acceptable salt of a compound according to claim 10 .
12. A pharmaceutical composition comprising a compound according to claim 1 , and a pharmaceutically acceptable carrier.
13. A pharmaceutical composition according to claim 12 in the form of a tablet, capsule, liquid or suspension.
14. A method for inhibiting the replication of a human immunodeficiency virus in a cell infected with said virus comprising contacting the cell infected with the virus with an antiviral effective amount of a compound according to claim 1 .
15. A method for inhibiting the replication of a human immunodeficiency virus in a cell infected with said virus, comprising contacting the cell infected with the virus with an antiviral effective amount of a first therapeutic agent that is a compound according to claim 1 and at least one additional therapeutic agent selected from the group consisting of zidovudine, didanosine, lamivudine, zalcitabine, abacavir, stavidine, adefovir, adefovir dipivoxil, nevirapine, delavirdine, efavirenz, TMC-278, TMC-125, saquinavir, ritonavir, indinavir, nelfinavir, amprenavir, fosamprenavir, brecanavir, palinavir, lasinavir, atazanavir, tipranavir, enfuvirtide, T-1249, L-870,810, raltegravir, vicriviroc, and maraviroc.
16. A method for inhibiting a human immunodeficiency virus in a cell infected with said virus, comprising contacting the cell infected with the virus with an antiviral effective amount of a compound according to claim 10 .

A method for inhibiting a human immunodeficiency virus in a cell infected with said virus, comprising contacting the cell infected with the virus with an antiviral effective amount of a first therapeutic agent that is a compound according to claim 10 and at least one additional therapeutic agent selected from the group consisting of zidovudine, didanosine, lamivudine, zalcitabine, abacavir, stavidine, adefovir, adefovir dipivoxil, nevirapine, delavirdine, efavirenz, TMC-278, TMC-125, saquinavir, ritonavir, indinavir, nelfinavir, amprenavir, fosamprenavir, brecanavir, palinavir, lasinavir, atazanavir, tipranavir, enfuvirtide, T-1249, L-870,810, raltegravir, vicriviroc, and maraviroc.
