Abstract

The invention provides novel TRIM polypeptides, proteins, and nucleic acid molecules. In addition to isolated, full-length TRIM proteins, the invention further provides isolated TRIM fusion proteins, antigenic peptides and anti-TRIM antibodies. The invention also provides TRIM nucleic acid molecules, recombinant expression vectors containing a nucleic acid molecule of the invention, host cells into which the expression vectors have been introduced and non-human transgenic animals in which an TRIM gene has been introduced or disrupted. The present invention also provides methods and compositions for the diagnosis and treatment of viral infection and/or replication, e.g., HIV infection. The invention further provides methods for identifying a compound capable of treating or preventing viral infection and/or replication, e.g., HIV infection and AIDS. In addition, the invention provides a method for treating a subject having a viral infection and/or replication, e.g., HIV infection using the modulators of the invention.

Claims (19)

1. A method of inhibiting HIV infection in one or more cells or conferring HIV infection resistance to one or more cells, comprising administering to one or more cells an agent selected from the group consisting of:

(a) an isolated nucleic acid molecule comprising a nucleic acid sequence having at least 95% identity to the nucleotide sequence of SEQ ID NO:1, wherein the nucleic acid molecule encodes a Trim5α polypeptide and the Trim5α polypeptide is stably expressed in the cell; and

(b) an isolated Trim5α polypeptide comprising an amino acid sequence having at least 95% identity to the amino acid sequence of SEQ ID NO:2.

2. The method of claim 1 , wherein the isolated nucleic acid molecule comprises the nucleic acid sequence of SEQ ID NO:1.

3. The method of claim 1 , wherein the isolated nucleic acid molecule consists of the nucleic acid sequence of SEQ ID NO:1.

4. The method of claim 1 , wherein the isolated nucleic acid molecule encodes a polypeptide comprising the amino acid sequence of SEQ ID NO:2.

5. The method of claim 1 , wherein the isolated nucleic acid molecule encodes a polypeptide consisting of the amino acid sequence of SEQ ID NO:2.

6. The method of claim 1 , wherein the isolated Trim5α polypeptide comprises the amino acid sequence of SEQ ID NO:2.

7. The method of claim 1 , wherein the isolated Trim5α polypeptide consists of the amino acid sequence of SEQ ID NO:2.

8. The method of any one of claims 1 - 7 , wherein the agent is administered in a pharmaceutically acceptable formulation.

9. The method of any one of claims 1 - 7 , further comprising administering at least one additional anti-viral agent.

10. The method of any one of claims 1 - 7 , wherein the agent is encapsulated in a liposome.

11. The method of any one of claims 1 - 5 , wherein the isolated nucleic acid molecule is administered using a vector.

12. The method of claim 11 , wherein said vector is a viral vector.

13. The method of any one of claims 1 - 7 , wherein the cells are in vitro, in vivo or ex vivo.

14. The method of any one of claims 1 - 7 , wherein the cells are human cells.

15. The method of any one of claims 1 - 7 , wherein the cells are in a subject at risk for HIV infection.

16. The method of claim 15 , wherein the subject is a human.

17. The method of 15 , wherein the cells are present in a mucosal membrane of the subject.
