Abstract

Antibodies which specifically bind heregulin-coupled HER3, at a site distinct from the heregulin binding site, are described. These antibodies are particularly useful in treating cancer.

Claims (23)

1. Monoclonal antibodies including fragments and recombinant forms thereof which bind to HER3 not complexed to heregulin, and which bind to HER3:heregulin complex with greater affinity as compared to binding to uncomplexed HER3.

2. The antibodies of claim 1 which bind to HER3 not complexed to heregulin with an affinity of at least 5 nM and bind to a HER3:heregulin complex with an affinity at least 5 times that of binding to HER3.

3. An antibody of claim 1 which is 1G4.

4. The antibodies of claim 1 which are human or humanized.

5. The antibodies of claim 1 which are prepared recombinantly.

6. The antibodies of claim 5 which are single chain antibodies.

7. Conjugates of the antibodies of claim 1 coupled to a cytotoxic agent or to a chemotherapeutic agent.

8. A pharmaceutical composition that contains the antibodies of claim 1 .

9. A pharmaceutical composition that contains the conjugates of claim 7 .

10. The antibodies of claim 1 wherein the heavy chain comprises CDR1: GYTFTDYVS (SEQ ID NO:4), CDR2: IYPSGRY (SEQ ID NO:5) and CDR3: TRSLQRLRYFDV (SEQ ID NO:6).

11. The antibodies of claim 1 wherein the light chain comprises CDR1: RASQSISDYLH (SEQ ID NO:10), CDR2: YGSQSIS (SEQ ID NO:11) and CDR3: QQSNSWPLT (SEQ ID NO:12).

12. The antibodies of claim 10 wherein the light chain comprises CDR1: RASQSISDYLH (SEQ ID NO:10), CDR2: YGSQSIS (SEQ ID NO:11) and CDR3: QQSNSWPLT (SEQ ID NO:12).

13. The antibodies of claim 2 which are human or humanized.

14. The antibodies of claim 2 which are prepared recombinantly.

15. The antibodies of claim 14 which are single chain antibodies.

16. Conjugates of the antibodies of claim 2 coupled to a cytotoxic agent or to a chemotherapeutic agent.

17. A pharmaceutical composition that contains the antibodies of claim 2 .

18. A pharmaceutical composition that contains the conjugates of claim 16 .

19. The antibodies of claim 12 which are prepared recombinantly.

20. The antibodies of claim 12 which are single chain antibodies.

21. Conjugates of the antibodies of claim 12 coupled to a cytotoxic agent or to a chemotherapeutic agent.

22. A pharmaceutical composition that contains the antibodies of claim 12 .

23. A pharmaceutical composition that contains the conjugates of claim 21 .
