Abstract

A novel gene 202P5A5 and its encoded protein, and variants thereof, are described wherein 202P5A5 exhibits tissue specific expression in normal adult tissue, and is aberrantly expressed in the cancers listed in Table I. Consequently, 202P5A5 provides a diagnostic, prognostic, prophylactic and/or therapeutic target for cancer. The 202P5A5 gene or fragment thereof, or its encoded protein, or variants thereof, or a fragment thereof, can be used to elicit a humoral or cellular immune response; antibodies or T cells reactive with 202P5A5 can be used in active or passive immunization.

Claims (16)

1. An isolated polynucleotide encoding a protein having the amino acid sequence set forth in SEQ ID NO: 12, wherein:

the polynucleotide comprises the nucleic acid sequence set forth in SEQ ID NO: 2, wherein the nucleotide at position 138 is G, the nucleotide at position 1269 is G, the nucleotide at position 1288 is A, and the nucleotide at position 1662 is A;

the polynucleotide consists of residues 29-1858 of SEQ ID NO: 2, with or without the stop codon, wherein the nucleotide at position 138 is G, the nucleotide at position 1269 is G, the nucleotide at position 1288 is A, and the nucleotide at position 1662 is A; or

the polynucleotide comprises residues 29-1858 of SEQ ID NO: 2, with or without the stop codon, wherein the nucleotide at position 138 is G, the nucleotide at position 1269 is G, the nucleotide at position 1288 is A, the nucleotide at position 1552 is G, and the nucleotide at position 1662 is A.

The isolated polynucleotide of claim 1 , wherein the polynucleotide comprises residues 29-1858 of SEQ ID NO: 2, with or without the stop codon, wherein the nucleotide at position 138 is G, the nucleotide at position 1269 is G, the nucleotide at position 1288 is A, the nucleotide at position 1552 is G, and the nucleotide at position 1662 is A.

3. The isolated polynucleotide of claim 1 , wherein the polynucleotide comprises the nucleic acid sequence set forth in SEQ ID NO: 2, wherein the nucleotide at position 138 is G, the nucleotide at position 1269 is G, the nucleotide at position 1288 is A, and the nucleotide at position 1662 is A.

The isolated polynucleotide of claim 1 , wherein the polynucleotide consists of the nucleic acid sequences set forth in SEQ ID NO: 2, wherein the nucleotide at position 138 is G, the nucleotide at position 1269 is G, the nucleotide at position 1288 is A, and the nucleotide at position 1662 is A.

5. The isolated polynucleotide of claim 4 , wherein the nucleotide at position 1552 is A.

The isolated polynucleotide of claim 3 , wherein the nucleotide at position 1552 is A, the nucleotide at position 1899 is G, the nucleotide at position 2057 is T, the nucleotide at position 2143 is C, the nucleotide at position 2144 is G, the nucleotide at position 2249 is C, the nucleotide at position 2576 is A, the nucleotide at position 2812 is T, the nucleotide at position 2836 is G, the nucleotide at position 3059 is T, the nucleotide at position 3101 is G, the nucleotide at position 3309 is A, the nucleotide at position 3332 is T, the nucleotide at position 4208 is C, the nucleotide at position 4264 is C, the nucleotide at position 4409 is C, and the nucleotide at position 4645 is A.

7. A recombinant vector, comprising the isolated polynucleotide of claim 1 .

8. The recombinant vector of claim 7 , wherein the vector is a viral vector.

9. The recombinant vector of claim 8 , wherein the viral vector is a vaccinia, fowlpox, canarypox, adenovirus, influenza, poliovirus, adeno-associated virus, lentivirus, or Sindbis virus vector.

10. An isolated host cell, comprising the vector of claim 8 .

11. An isolated host cell, comprising the vector of claim 7 .

12. A process for producing a protein having the amino acid sequence set forth in SEQ ID NO: 12, comprising culturing the host cell of claim 11 under conditions sufficient for the production of a protein, and recovering the protein from the culture.

13. The process of claim 12 , wherein the protein is recovered using chromatography.
