Abstract

Activation sites on the alpha subunit of sodium potassium ATPase have been discovered. It has also been discovered that certain antibodies that bind to the alpha subunit of sodium potassium ATPase dramatically increase enzyme activity. There has never before been a report of precise activation sites or drug interaction sites for sodium potassium ATPase. Certain methods have also been discovered for treating or preventing diseases associated with low sodium potassium ATPase activity by administering antibodies, antibody fragments and small molecules that bind to the activation sites on the alpha subunit of sodium potassium ATPase.

Claims (5)

1. A pharmaceutical composition that increases (Na + +K + )-ATPase activity in an animal cell or bodily fluid, comprising an antibody or an antibody fragment that binds to an activation site on the alpha subunit of any isoform of (Na + +K + )-ATPase, which activation site consists of HLLGIRETWDDRWIN (SEQ ID NO: 4).

The pharmaceutical composition of claim 1 , wherein the animal cell is a member selected from the group comprising hepatocytes, kidney cells, red blood cells, endothelial cells, lung cells, nerve cells, lens cells, brain cells, muscle cells, cardiac myocytes, and any cells that express (Na + +K + )-ATPase.

The pharmaceutical composition of claim 1 , wherein the isolated antibody or antibody fragment that recognizes and binds to an epitope consisting of the amino acid sequence HLLGIRETWDDRWIN (SEQ ID NO: 4) in the α-subunit of any isoform of (Na + +K + )-ATPase, wherein binding of the antibody to the amino acid sequence, HLLGIRETWDDRWIN (SEQ ID NO: 4) in a therapeutically effective amount increases myocyte intracellular diastolic or systolic calcium in an animal.

4. The pharmaceutical composition of claim 1 , wherein binding of the antibody or antibody fragment to the alpha unit of any isoform of (Na + +K + )-ATPase exerts a positive inotropic effect in cardiac myocytes or in a heart for treatment of heart diseases and muscle contractile disorders.

5. The pharmaceutical composition of claim 1 , wherein the antibody or antibody fragment is a polyclonal or monoclonal antibody; the antibody is a human or humanized antibody.
