Abstract

Methods for treating cancer using compounds that inhibit human DNA ligases. Methods for using compounds that inhibit human DNA ligases to provide insights into the reaction mechanisms of human DNA ligases, for example to identify the human DNA ligase involved in different DNA repair pathways. Screening methods for compounds that inhibit human DNA ligases.

Claims (20)

1. A method of treating chronic leukemia, breast cancer or colon cancer exhibiting an altered DNA damage response in a patient in need comprising administering to said patient an effective amount of at least one anti-cancer compound according to the chemical structures as set forth below:

in combination with a second cancer treatment comprising a DNA damaging agent.

2. The method according to claim 1 wherein said anti-cancer compound is

3. The method according to claim 1 wherein said DNA damaging agent is ionizing radiation, a PARP inhibitor or a DNA alkylating agent.

4. The method according to claim 3 wherein said DNA damaging agent is a PARP inhibitor.

5. The method according to claim 4 wherein said PARP inhibitor is 3-aminobenzamide.

6. The method according to claim 3 wherein said DNA damaging agent is ionizing radiation.

7. The method according to claim 3 wherein said DNA damaging agent is a DNA alkylating agent.

8. The method according to claim 7 wherein said DNA alkylating agent is methyl methanesulfonate (MMS).

9. The method according to claim 1 wherein said cancer is breast cancer.

10. The method according to claim 1 wherein said cancer is chronic leukemia.

11. The method according to claim 1 wherein said compound is

and said cancer is chronic leukemia.

12. The method according to 1 wherein said compound is

and said cancer is breast cancer.

13. The method according to claim 1 wherein said compound is

and said cancer is colon cancer.

14. The method of claim 1 , wherein said DNA damaging agent is administered at the same time with administering said at least one compound.

15. The method of claim 1 , wherein said DNA damaging agent is administered prior to administering said at least one compound.

16. The method of claim 1 , wherein said DNA damaging agent is administered after administering said at least one compound.
