Abstract

Methods are provided for analyzing characteristics of fluids in the context of an acoustic ejection system. Such a system has a controller, an acoustic radiation generator, and a coupling medium coupling the radiation to a reservoir holding fluid. The methods can use acoustic radiation to both perturb a surface of the fluid in the reservoir and analyze the effect of the perturbation. The methods may use information about prior fluids. The methods of the invention can determine physical characteristics such as speed of sound and viscosity. The methods also include ways to determine a level of acoustic energy suitable to eject a droplet. Preferably the methods are executed automatically under control of programming of a controller of an acoustic ejection system.

Claims (31)

1. A method for determining a speed of sound in a fluid, the method comprising:

immersing a solid object at a first location in a quantity of the fluid in a reservoir;

sending first acoustic energy to the solid object;

detecting second acoustic energy reflected from the solid object at the first location;

moving the solid object from the first location to a second location in the quantity of the fluid;

detecting third acoustic energy reflected from the solid object at the second location; and

calculating a speed of sound in the fluid by using as an input a distance between the first location of the solid object and the second location of the solid object.

2. The method of claim 1 , and further comprising generating the first acoustic energy by an acoustic ejection system capable of ejecting droplets of fluid from the reservoir.

3. The method of claim 1 , wherein the solid object is a pin tool.

4. The method of claim 1 , wherein the process for sending first acoustic energy to the solid object includes directing the first acoustic energy in a vertical direction towards the solid object.

5. The method of claim 1 , wherein the first acoustic energy is focused and associated with an F-number.

6. The method of claim 5 , wherein the F-number is approximately 2 or greater.

7. The method of claim 5 , wherein the process for calculating a speed of sound in the fluid is performed by using as an input the F-number.

8. The method of claim 1 , wherein the method is carried out for each of a plurality of fluids in a plurality of reservoirs respectively.

9. The method of claim 8 , wherein the plurality of reservoirs for the plurality of fluids for which the method is carried out form part of a well plate.

10. A method for determining a speed of sound in a fluid, the method comprising:

immersing a solid object in a quantity of the fluid in a reservoir;

supplying first acoustic energy by an acoustic ejection system capable of ejecting droplets of fluid from the reservoir; and

detecting second acoustic energy reflected from the solid object;

wherein the process for immersing a solid object is performed by using a positioning system also used by the acoustic ejection system for other purposes.

11. The method of claim 10 , and further comprising:

moving the solid object; and

detecting third acoustic energy reflected from the solid object in its position after being moved.

12. The method of claim 10 , and further comprising calculating a speed of sound in the fluid by using as an input a distance between the solid object and a portion of the fluid in the reservoir closest to the acoustic ejection system.

13. The method of claim 10 , wherein the solid object is a pin tool.

14. The method of claim 10 , wherein the process for supplying first acoustic energy includes directing the first acoustic energy in a vertical direction towards the solid object.

15. The method of claim 10 , wherein the method is carried out for each of a plurality of fluids in a plurality of reservoirs respectively.

16. The method of claim 15 , wherein the plurality of reservoirs for the plurality of fluids for which the method is carried out form part of a well plate.

17. The method of claim 10 , wherein the first acoustic energy is focused and associated with an F-number.

18. The method of claim 17 , wherein the F-number is approximately 2 or greater.

19. The method of claim 17 , and further comprising calculating a speed of sound in the fluid by using as an input the F-number.
