Abstract

The present invention is directed to nicotinamide derivatives, and their use in treating lipoprotein abnormalities, alone or in combination with a statin.

Claims (19)

1. A method for increasing HDL-cholesterol levels in a human subject in need thereof, comprising administering orally to said subject a therapeutically effective amount of 1-methylnicotinamide or a pharmaceutically acceptable salt thereof and a therapeutically effective amount of a statin.

2. The method of claim 1 wherein said subject has low HDL-cholesterol levels before said administration.

3. The method of claim 1 , wherein said 1-methylnicotinamide salt is selected from the group consisting of 1-methylnicotinamide chloride, 1-methylnicotinamide benzoate, 1-methylnicotinamide salicylate, 1-methylnicotinamide acetate, 1-methylnicotinamide citrate, and 1-methylnicotinamide lactate.

4. The method of claim 1 , wherein said 1-methylnicotinamide or a pharmaceutically acceptable salt thereof and said statin are administered concurrently.

5. The method of claim 1 , wherein said 1-methylnicotinamide or a pharmaceutically acceptable salt thereof and said statin are administered sequentially.

6. The method of claim 1 , wherein said statin is selected from the group consisting of mevastatin, lovastatin, simvastatin, pravastatin, fluvastatin, pitavastatin, atorvastatin, cerivastatin, rosuvastatin, pentostatin, nystatin, a pharmaceutically acceptable salt thereof, and combinations thereof.

7. The method of claim 1 , wherein said 1-methylnicotinamide or a pharmaceutically acceptable salt thereof and said statin are administered formulated in pharmaceutical compositions.

8. A method for decreasing triglyceride levels in a human subject in need thereof, comprising administering orally to said subject a therapeutically effective amount of 1-methylnicotinamide or a pharmaceutically acceptable salt thereof and a therapeutically effective amount of a statin.

9. The method of claim 8 , wherein said 1-methylnicotinamide salt is selected from the group consisting of 1-methylnicotinamide chloride, 1-methylnicotinamide benzoate, 1-methylnicotinamide salicylate, 1-methylnicotinamide acetate, 1-methylnicotinamide citrate, and 1-methylnicotinamide lactate.

10. The method of claim 8 , wherein said 1-methylnicotinamide or a pharmaceutically acceptable salt thereof and said statin are administered concurrently.

11. The method of claim 8 , wherein said 1-methylnicotinamide or a pharmaceutically acceptable salt thereof and said statin are administered sequentially.

12. The method of claim 8 , wherein said statin is selected from the group consisting of mevastatin, lovastatin, simvastatin, pravastatin, fluvastatin, pitavastatin, atorvastatin, cerivastatin, rosuvastatin, pentostatin, nystatin, a pharmaceutically acceptable salt thereof, and combinations thereof.

13. The method of claim 8 , wherein said 1-methylnicotinamide or a pharmaceutically acceptable salt thereof and said statin are administered formulated in pharmaceutical compositions.

14. A method of treating atherosclerosis in a human subject in need thereof, comprising administering orally to said subject a therapeutically effective amount of a 1-methylnicotinamide or a pharmaceutically acceptable salt thereof and a therapeutically effective amount of a statin.

15. The method of claim 14 , wherein said 1-methylnicotinamide salt is selected from the group consisting of 1-methylnicotinamide chloride, 1-methylnicotinamide benzoate, 1-methylnicotinamide salicylate, 1-methylnicotinamide acetate, 1-methylnicotinamide citrate, and 1-methylnicotinamide lactate.

16. The method of claim 14 , wherein said 1-methylnicotinamide or a pharmaceutically acceptable salt thereof and said statin are administered concurrently.

17. The method of claim 14 , wherein said 1-methylnicotinamide or a pharmaceutically acceptable salt thereof and said statin are administered sequentially.

18. The method of claim 14 , wherein said statin is selected from the group consisting of mevastatin, lovastatin, simvastatin, pravastatin, fluvastatin, pitavastatin, atorvastatin, cerivastatin, rosuvastatin, pentostatin, nystatin, a pharmaceutically acceptable salt thereof, and combinations thereof.

19. The method of claim 14 , wherein said 1-methylnicotinamide or a pharmaceutically acceptable salt thereof and said statin are administered formulated in pharmaceutical compositions.
