Fusion protein with HIV antigen
The present invention includes compositions and methods for the expression, secretion and use of novel compositions for use as, e.g., vaccines and antigen delivery vectors, to delivery antigens to antigen presenting cells. In one embodiment, the vector is an anti-CD40 antibody, or fragments thereof, and one or more antigenic peptides linked to the anti-CD40 antibody or fragments thereof, including humanized antibodies.
1. A fusion protein comprising the formula:
Ab-(PL-Ag) x ;
wherein Ab is an antibody or a fragment thereof;
PL is selected from SEQ ID NO.: 11, 12, 13 or 14;
Ag is at least one viral antigen; and
x is an integer from 1 to 20.
2. The fusion protein of claim 1 , wherein the fusion protein has more stability in solution than the fusion protein comprising a PL without the glycosylation site.
3. The fusion protein of claim 1 , wherein the Ag comprises a peptide from a retrovirus.
4. The fusion protein of claim 1 , wherein the Ag comprises at least one of HIV antigen.
5. The fusion protein of claim 1 , wherein the Ag is selected from:

6. The fusion protein of claim 1 , wherein the Ag is 19 to 32 residues.
7. The fusion protein of claim 1 , wherein the Ag is a cytotoxic T lymphocyte (CTL) epitope identified in the Gag protein presented in the context of MHC-class I molecules.
8. The fusion protein of claim 1 , wherein the Ag is HIV Gag.
9. The fusion protein of claim 1 , wherein x is 3.
10. The fusion protein of claim 1 , wherein the Ag comprises virus peptides from different antigens separated by different PLs selected from SEQ ID NO.: 11, 12, 13 or 14.
11. The fusion protein of claim 1 , wherein the Ab comprises SEQ ID NOS.: 37 and 38.
12. The fusion protein of claim 1 , wherein the Ab comprises at least the variable region of the antibody anti-CD40 — 12E12.3F3 (ATCC Accession No. PTA-9854).
13. The fusion protein of claim 1 , wherein the Ab comprises at least one variable domain having SEQ ID NO.: 148.
14. The fusion protein of claim 1 , wherein the Ag is 17 to 60 residues.
15. The fusion protein of claim 1 , wherein the Ag comprises at least one lipopeptide.
16. The fusion protein of claim 1 , wherein the Ag is at the carboxy-terminus and further comprises a carboxy-terminus (Palm)-NH 2 group.
17. A viral antigen delivery vector comprising: a fusion protein comprising an anti-CD40 antibody, or a fragment thereof, a peptide linker selected from SEQ ID NO.: 11, 12, 13, or 14, and one or more viral peptides at the carboxy-terminus of the anti-CD40 antibody, wherein, when two or more viral peptides are present, the viral peptides are separated by one or more peptide linkers selected from SEQ ID NO:11, 12, 13 or 14.
18. A fusion protein comprising an anti-CD40 antibody, or a fragment thereof, with one or more viral peptides at the carboxy-terminus of the antibody separated by one or more peptide linkers selected from SEQ ID NO.: 11, 12, 13, or 14.
19. The fusion protein of claim 18 , wherein the one or more viral peptides are HIV peptides.
